Based on the zymography analysis, Bacilfus sp. DJ-4 (screened from Doen-Jang, a Korean traditional fermented food) secretes seven extracellalar fibrinolytic enzymes (EFEs; 68, 64, 55, 4S, 33, 27, and 13 kDa) in culture broth. These seven EFEs were analyzed by newly applied SDSfibrin zymography combined with gradient polyacrylamide (SDS-FZGP). This improved gel system was used with a 5-20% acrylamide gradient in a fibrin zymogram gel for the separation of proteins with molecular masses from below 10 kDa to over 100 kDa on one gel plate. Using this system, high molecular weight bands (HMWBs) were clearly and sharply resolved. We also examined the relationship between an acrylamide concentration and the enzymatic activity of EFE using densitometric analysis.
Comparison of Three Substrates ( Casein , Fibrin , and Gelatin ) in Zymographic Gel

Three zymographic techniques using casein, fibrin, and gelatin as substrates in SDS-PAGE were compared based on three aspects: (1) The proteolytic pattern of extracellular enzymes from the three bacterial strains, Bacillus sp. DJ-l, DJ-2, and DJ-3. (2) The enzymatic sensitivity of their activity on zymogram gels. (3) The stability of stained zymogram gels with Coomassie brilliant blue in the destaining solution. There was no significant difference on the pattern of extracellular enzymes from the three strains. The bands in the fibrin gel were clearer and more distinct from the extensive destaining process. It was also shown that the gelatin gel revealed the highest enzymatic sensitivity among the three gels, based on the densitometric analysis. In the casein gel, a trace that could be mistaken as a proteolytic band appeared around 40-50 kDa.
생물막 유동층 반응기에서 고농도 합성 폐수의 처리효율과 미생물 성상

이 실험의 목적은 생물막 유동층 반응기내에서 미생물 성상의 변화를 관찰하여 유기물 처리효율과 최대 미생물 농도를 안정적으로 유지할 수 있는 조건을 고찰하는데 있다. 모래를 여재로 사용하여 수리학적 체류시간을 5시간, 상향유속을 17m/hr, 운전온도는 22±1℃, pH는 7±0.1로 고정한 상태에서 유기물 부하율을 10㎏ COD/㎥·day에서 80㎏ COD/㎥·day로 증가시키면서 실험을 수행하였다. 또한 실험에 사용한 폐수는 글루코오즈를 기질로 한 합성폐수를 조제하여 사용하였다. 유기물 부하율이 10㎏ COD/㎥·day에서 COD는 96%의 제거율을 나타내었고, 반응기내 미생물 농도는 6,530㎎/ℓ이었다. 또한, 유기물 부하율이 80㎏ COD/㎥·day로 증가되어도 COD는 73%의 높은 제거율과 반응기내 미생물 농도는 37,000㎎/ℓ를 나타내었다. 유기물 제거율의 관점에서 보면 F/M비가 1.7 이하, 미생물막 두께가 420㎛ 이하에서 운전하는 것이 가장 효율적인 것으로 나타났다. The purposes of this experiment were to investigate biomass characteristics and substrate removal efficiency and to find the optimum conditions on holding up higher biomass concentration in Fluidized-Bed Biofilm Reactor(FBBR). A number of experiments were conducted under the following conditions by increasing organic loading rate from 10㎏ COD/㎥·day to 80㎏ COD/㎥·day. Superficial upflow velocity was 17m/hr, operating temperature was 22±1℃, pH and 7±0.1. The synthetic wastewater based on glucose was used as a substrate in this experiment. The removal of chemical oxygen demand(COD) with volumetric loading rates of 10㎏ COD/㎥. day·80㎏ COD/㎥·day was investigated to show the efficiency of aerobic FBBR. The corresponding COD removal efficiencies were 96 and 73% with volumetric loading rate of 10 to 80 ㎏ COD/㎥ day, respectively, and the concentrations of biomass remaining inside the reactor were shown 6,500㎎/ℓand 37,000㎎/ℓ, respectively. These experimental conditions of the F/M ratio of 1.7 and the biofilm thickness of 420㎛ was proved to be a most appropriate condition in operating FBBR in the view of substrate removal.

A number of experiments were conducted in order to investigate the organic removal efficiency and biomass characteristics according to the organic shock loading rate in a fluidized bed biofilm reactor. At the operation conditions of HRT, 8.44 hour, superficial upflow velocity, 0.9 ㎝/sec and temperature, 22±1 ℃, the removal efficiency of SCOD was founded to be 96.5, 92 and 90 % with the organic shock loading rate of 3.5, 10.8 and 33 kgCOD/㎥·day, respectively. Within the F/M ratio ranged 0.4 to 2.0 ㎏COD/㎏VSS·day, the SCOD removal efficiency was shown as 90% at F/M ratio of 2.0 ㎏COD/㎏VSS·day, but the TCOD removal efficiency was 72 % at F/M ratio of 1.8 kgCOD/kgVSS·day. The average biomass concentrations were 7800, 14950 and 27532 ㎎/l on the organic shock loading rate of 3.5, 10.8 and 33 ㎏COD/㎥·day, respectively. This result was agreed with the fact that more biomass could be produced at high concentration of substrate, but some biomass was detached at the onset of shock and easily acclimated at the shock condition.

Objectives: This study was aimed to find out the physiological mechanism of craniosacral therapy, especially in CV4 technique and cranial rhythmic impulses. Methods: Recent studies were reviewed for cranial rhythmic impulses, and the ancient and present text were reviewed for the therapy of sub-occiput and cranial-suture part. Results and Conclusions: 1. Suboccipital muscles releasing may have made CV4 technique to effect. 2. The mechanism of the meridians of acupuncture and craniosacral system can be cooperative together in many parts of the clinical treatments. 3. Arterial vasomotor waves have a frequency similar to reports of cranial rhythmic impulses. The methods which have relation to heart rate variability, will be useful for the study of craniosacral therapy.
