      • Weight Loss Correlates with Macrophage Inhibitory Cytokine-1 Expression and Might Influence Outcome in Patients with Advanced Esophageal Squamous Cell Carcinoma

        Background: Weight loss during chemotherapy has not been exclusively investigated. Macrophage inhibitory cytokine-1 (MIC-1) might play a role in its etiology. Here, we investigated the prognostic value of weight loss before chemotherapy and its relationship with MIC-1 concentration and its occurrence during chemotherapy in patients with advanced esophageal squamous cell carcinoma (ESCC). Materials and Methods: We analyzed 157 inoperable locally advanced or metastatic ESCC patients receiving first-line chemotherapy. Serum MIC-1 concentrations were assessed before chemotherapy. Patients were assigned into two groups according to their weight loss before or during chemotherapy:>5% weight loss group and ${\leq}5%$ weight loss group. Results: Patients with weight loss>5% before chemotherapy had shorter progression-free survival period (5.8 months vs. 8.7 months; p=0.027) and overall survival (10.8 months vs. 20.0 months; p=0.010). Patients with weight loss >5% during chemotherapy tended to have shorter progression-free survival (6.0 months vs. 8.1 months; p=0.062) and overall survival (8.6 months vs. 18.0 months; p=0.022), and if weight loss was reversed during chemotherapy, survival rates improved. Furthermore, serum MIC-1 concentration was closely related to weight loss before chemotherapy (p=0.001) Conclusions: Weight loss both before and during chemotherapy predicted poor outcome in advanced ESCC patients, and MIC-1 might be involved in the development of weight loss in such patients.

        Cathepsin D deficiency delays central nervous system myelination by inhibiting proteolipid protein trafficking from late endosome/lysosome to plasma membrane

        This study aimed to investigate the role of cathepsin D (CathD) in central nervous system (CNS) myelination and its possible mechanism. By using CathD knockout mice in conjunction with immunohistochemistry, immunocytochemistry and western blot assays, the myelination of the CNS and the development of oligodendrocyte lineage cells in vivo and in vitro were observed. Endocytosis assays, real-time-lapse experiments and total internal reflection fluorescence microscopy were used to demonstrate the location and movement of proteolipid protein in oligodendrocyte lineage cells. In addition, the relevant molecular mechanism was explored by immunoprecipitation. The increase in Fluoromyelin Green staining and proteolipid protein expression was not significant in the corpus callosum of CathD−/− mice at the age of P11, P14 and P24. Proteolipid protein expression was weak at each time point and was mostly accumulated around the nucleus. The number of oligodendrocyte lineage cells (olig2+) and mature oligodendrocytes (CC1+) significantly decreased between P14 and P24. In the oligodendrocyte precursor cell culture of CathD−/− mice, the morphology of myelin basic protein-positive mature oligodendrocytes was simple while oligodendrocyte precursor cells showed delayed differentiation into mature oligodendrocytes. Moreover, more proteolipid protein gathered in late endosomes/lysosomes (LEs/Ls) and fewer reached the plasma membrane. Immunohistochemistry and immunoelectron microscopy analysis showed that CathD, proteolipid protein and VAMP7 could bind with each other, whereas VAMP7 and proteolipid protein colocalized with CathD in late endosome/lysosome. The findings of this paper suggest that CathD may have an important role in the myelination of CNS, presumably by altering the trafficking of proteolipid protein.

        Difficulties of facelift in patients with scar after treatment of facial liposuction or thread lift sutures

        Objective: To explore the methods and safety of general abnormal anatomical layers and structure in rhytidectomy, which is also of clinical significance. In this group, 39 patients with static and dynamic deformities formed at different levels after liposuction and thread technique more than one time were treated with stable and active separation skills in high SMAS facelift operation. The results showed that the patients of this group could get the same effect of younger postoperative operation as “initial face” and solve dynamic adhesion deformity. Methods: Magnetic resonance imaging and ultrasound were performed before operation, focusing on the abnormal changes of facial soft tissue. The results showed that the signal of the foreign body, the position and texture of scar, the relationship between scar and masseter fascia, buccal fat pad, and whether there was tissue defect or hyperplasia. In the operation, the normal separation was made in the second and fourth layers of the previously safe and easy to separate plane, instead of in the dense one or two layers of the scar. It is an alternative microsurgical separation technique. The key point is to separate the incomplete SMAS flap to achieve the expected good effect of rhytidectomy, release the scar adhesion and facial spasm. At the same time, the severed injury of facial nerve branches should be strictly prevented. Results: 74 cases of facelift were taken in recent eight months and 67 patients were treated with micro plastic surgery. Among them, 39 patients received more than one facial liposuction and more than one thread lift. In the scar formed by liposuction and thread technology, liposuction is especially serious; liposuction can still absorb the structural layer, and scar directly involves the SMAS and buccal fat pad envelope containing facial nerve branches, one of which directly causes the injury of the mandibular marginal branch. Most of the chronic granuloma scar formed after the degradation of the thread technology is in the facial space, which causes the scar atresia of the space. The flap and SMAS flap of 39 cases were separated, most of which were facial nerve branches in scar. All of them were difficult to separate and form the SMAS flap of superficial temporal fascia-Orbicularis oculi-platysma, and 89.6% of the excellent facelift effect was obtained. Facial nerve injury was found in 2 cases, 1 case was right mandible marginal branch traction injury, and the other side was left corner nerve injury. In the process of recovery. Conclusion: For patients with facial liposuction and thread technique, doctors must be prepared for the difficult separation of scar adhesion and have a high risk of facial nerve branch disconnection. If we don’t pursue the rejuvenation effect, it will lead to the existence of surgical effect discount, bilateral asymmetry, and dynamic adhesion deformity.

      • Historical Long-term Exposure to Pentachlorophenol Causing Risk of Cancer - A Community Study

        Background: Pervious studies suggested occupational workers exposure to pentachlorophenol (PCP) might contribute to increased risk of cancer. However, few studies have focused on associations between PCP and cancer risk at the community level. Objective: The present study was to explore the cancer risk for the community population living long-term in a PCP contaminated area. Methods: All the cancer cases diagnosed in 2009-2011 in Tongling City were collected. The cancer patients' residencies were geo-referenced in each district. The historical PCP usage for each district of Tongling was calculated as the PCP pollution index, which was further used to divide into PCP exposure categories. Standardized rate ratios (SRRs) of cancer incidence were applied to detect the cancer risk as exposure grade elevated. Correlation analysis was performed to analyze the relationship between PCP pollution and cancer incidence. Results: A total of 5,288 cancer cases (3,451 male and 1,837 female) were identified. PCP usage was correlated with the incidence of leukemia (r=0.88, P=0.002) for males, and with cancer of the esophagus for males (r=0.83, P=0.008) and females (r=0.71, P=0.020). Compared with the low exposure category, significant SRRs for total cancer sites was obtained for high PCP exposure category (SRR=1.61, 95%CI=1.59-1.62). Most SRR values of the cancer sites were significantly increased as exposure grade elevated and exposure time extended. Conclusion: The present study found that community residents living in the PCP contaminated area had increased risk of cancers. Leukemias, lymphomas and nasopharyngeal and esophageal cancers are most possibly associated with PCP exposure.

        Association between Fecal Bile Acids and Colorectal Cancer: A Meta-analysis of Observational Studies

        Purpose: To provide a systematic review with meta-analysis for addressing the relationship between fecal bile acids (FBAs) and colorectal cancer. Materials and Methods: Electronic databases were searched for all observational studies that examined the relationship between FBAs and colorectal cancer or adenoma, and calculated weighted mean difference (WMD) and 95% confidence interval (CI). Publication bias was assessed with funnel plot. Results: Twenty case-control or cohort studies were identified. All studies were pooled to assess the relationship between total FBAs and cancer/adenoma of the large bowel, however, no association was seen (WMD 0.61mg/g freeze-dried feces; 95% CI: -0.35-1.57). Significantly increased concentration of chenodeoxycholic acid (CDCA) was seen while pooling to assess the relationship between CDCA and cancer/adenoma of the large bowel (WMD 0.13 mg/g freeze-dried feces; 95% CI: 0.01-0.25), especially for colorectal cancer (WMD 0.28mg/g freeze-dried feces; 95% CI: 0.10-0.46). However, no significant differences in deoxycholic acid (DCA), lithocholic acid (LCA), and primary and secondary bile acids, were seen between patients with cancer and patients with matched controls regardless of fixed and random effects models. Conclusion: CDCA might play a role in the etiology of colorectal cancer.

        Vibration analysis of viscoelastic single-walled carbon nanotubes resting on a viscoelastic foundation

        Vibration responses were investigated for a viscoelastic Single-walled carbon nanotube (visco-SWCNT) resting on a viscoelastic foundation. Based on the nonlocal Euler-Bernoulli beam model, velocity-dependent external damping and Kelvin viscoelastic foundation model, the governing equations were derived. The Transfer function method (TFM) was then used to compute the natural frequencies for general boundary conditions and foundations. In particular, the exact analytical expressions of both complex natural frequencies and critical viscoelastic parameters were obtained for the Kelvin-Voigt visco-SWCNTs with full foundations and certain boundary conditions, and several physically intuitive special cases were discussed. Substantial nonlocal effects, the influence of geometric and physical parameters of the SWCNT and the viscoelastic foundation were observed for the natural frequencies of the supported SWCNTs. The study demonstrates the efficiency and robustness of the developed model for the vibration of the visco-SWCNT-viscoelastic foundation coupling system.
