        FSCB phosphorylation in mouse spermatozoa capacitation

        It is generally accepted that spermatozoa capacitation is associated with protein kinase A-mediated tyrosine phosphorylation. In our previous study, we identified the fibrous sheath CABYR binding protein (FSCB), which was phosphorylated by PKA. However, the phosphorylation status of FSCB protein during spermatozoa capacitation should be further investigated. To this aim, in this study, we found that phosphorylation of this 270-kDa protein occurred as early as 1 min after mouse spermatozoa capacitation, which increased over time and remained stable after 60 min. Immunoprecipitation assays demonstrated that the tyrosine and Ser/Thr phosphorylation of FSCB occurred during spermatozoa capacitation. The extent of phosphorylation and was closely associated with the PKA activity and spermatozoa motility characteristics. FSCB phosphorylation could be induced by PKA agonist DB-cAMP, but was blocked by PKA antagonist H-89.Therefore, FSCB contributes to spermatozoa capacitation in a tyrosine-phosphorylated format, which may help in further elucidating the molecular mechanism of spermatozoa capacitation.       • Research of Manufacturing Resource Sharing System Based on P2P

        To realize the equal sharing and full use of manufacturing resource, aiming at the convergence characteristics and dynamic changing behaviors of the distributed and stochastic manufacturing resources, according to the thought of peer-to-peer (P2P) and the Chord protocol, a distributed manufacturing resource sharing system model is proposed, making logic sub rings by the type of resources and the logic main rings by the super nodes. The relay of the resource rings and the convergence of different types of resources for application are completed by super nodes. The description template of the manufacturing resources and the Meta data model relating to the search keyword sets from the template are designed. The indexing process of the resources based on Meta data model and the hierarchical trust mechanism of the sharing system are elaborated; finally, an instance of the sharing system prototype is given.

        Comparison of <sup>18</sup>F-Labeled Fluoroalkylphosphonium Cations with <sup>13</sup>N-NH<sub>3</sub> for PET Myocardial Perfusion Imaging

        <P>Despite substantial advances in the diagnosis of cardiovascular disease, there is a need for <SUP>18</SUP>F-labeled myocardial perfusion agents for the diagnosis of ischemic heart disease because current PET tracers for myocardial perfusion imaging have a short half-life that limits their widespread clinical use in PET. Thus, <SUP>18</SUP>F-labeled fluoroalkylphosphonium derivatives (<SUP>18</SUP>F-FATPs), including (5-<SUP>18</SUP>F-fluoropentyl)triphenylphosphonium cation (<SUP>18</SUP>F-FPTP), (6-<SUP>18</SUP>F-fluorohexyl)triphenylphosphonium cation (<SUP>18</SUP>F-FHTP), and (2-(2-<SUP>18</SUP>F-fluoroethoxy)ethyl)triphenylphosphonium cation (<SUP>18</SUP>F-FETP), were synthesized. The myocardial extraction and image quality of the <SUP>18</SUP>F-FATPs were compared with those of <SUP>13</SUP>N-NH<SUB>3</SUB> in rat models. <B>Methods:</B> The first-pass extraction fraction (EF) values of the <SUP>18</SUP>F-FATPs (<SUP>18</SUP>F-FPTP, <SUP>18</SUP>F-FHTP, <SUP>18</SUP>F-FETP) and <SUP>13</SUP>N-NH<SUB>3</SUB> were measured in isolated rat hearts perfused with the Langendorff method (flow velocities, 0.5, 4.0, 8.0, and 16.0 mL/min). Normal and myocardial infarction rats were imaged with small-animal PET after intravenous injection of 37 MBq of <SUP>18</SUP>F-FATPs and <SUP>13</SUP>N-NH<SUB>3.</SUB> To determine pharmacokinetics, a region of interest was drawn around the heart, and time–activity curves of the <SUP>18</SUP>F-FATPs and <SUP>13</SUP>N-NH<SUB>3</SUB> were generated to obtain the counts per pixel per second. Defect size was analyzed on the basis of polar map images of <SUP>18</SUP>F-FATPs and <SUP>13</SUP>N-NH<SUB>3.</SUB> <B>Results:</B> The EF values of <SUP>18</SUP>F-FATPs and <SUP>13</SUP>N-NH<SUB>3</SUB> were comparable at low flow velocity (0.5 mL/min), whereas at higher flows EF values of <SUP>18</SUP>F-FATPs were significantly higher than those of <SUP>13</SUP>N-NH<SUB>3</SUB> (4.0, 8.0, and 16.0 mL/min, <I>P</I> < 0.05). Myocardium-to-liver ratios of <SUP>18</SUP>F-FPTP, <SUP>18</SUP>F-FHTP, <SUP>18</SUP>F-FETP, and <SUP>13</SUP>N-NH<SUB>3</SUB> were 2.10 ± 0.30, 4.36 ± 0.20, 3.88 ± 1.03, and 0.70 ± 0.09, respectively, 10 min after injection, whereas myocardium-to-lung ratios were 5.00 ± 0.25, 4.33 ± 0.20, 7.98 ± 1.23, and 2.26 ± 0.14, respectively. Although <SUP>18</SUP>F-FATPs and <SUP>13</SUP>N-NH<SUB>3</SUB> sharply delineated myocardial perfusion defects, defect size on the <SUP>13</SUP>N-NH<SUB>3</SUB> images was significantly smaller than on the <SUP>18</SUP>F-FATP images soon after tracer injection (0–10 min, <I>P</I> = 0.027). <B>Conclusion:</B> <SUP>18</SUP>F-FATPs exhibit higher EF values and more rapid clearance from the liver and lung than <SUP>13</SUP>N-NH<SUB>3</SUB> in normal rats, which led to excellent image quality in a rat model of coronary occlusion. Therefore, <SUP>18</SUP>F-FATPs are promising new PET radiopharmaceuticals for myocardial perfusion imaging.</P>

      • Bithionol residue analysis in animal-derived food products by an effective and rugged extraction method coupled with liquid chromatography–tandem mass spectrometry

        <P><B>Abstract</B></P> <P>Herein, we developed a simple analytical procedure for the quantitation of bithionol residues in animal-derived food products such as porcine muscle, eggs, milk, eel, flatfish, and shrimp using a modified quick, easy, cheap, effective, rugged, and safe (QuEChERS) extraction method coupled with liquid chromatography–electrospray ionization tandem mass spectrometry (LC-ESI<SUP>+</SUP>/MS-MS). Samples were extracted with 0.1% solution of formic acid in acetonitrile and the extract was purified using a C18 sorbent. Separation was performed on a Waters XBridge™ C18 reversed-phase analytical column using 0.1% solution of formic acid/acetonitrile as the mobile phase. Six-point matrix-matched calibration indicated good linearity, with the calculated coefficients of determination (<I>R</I> <SUP>2</SUP>) being≥0.9813. Intra- and inter-day recoveries (determined at spiking levels equivalent to 1×and 2×the limit of quantitation (0.25μg/kg)) ranged between 80.0 and 94.0%, with the corresponding relative standard deviations (RSDs) being≤8.2%. The developed experimental protocol was applied to different samples purchased from local markets in Seoul, which were tested negative for bithionol residues. In conclusion, the proposed method proved to be versatile and precise, being ideally suited for the routine detection of bithionol residues in animal-derived food products with various protein and fat contents.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Bithionol residues were quantified in porcine muscle, whole milk, eggs, eel, flatfish, and shrimp. </LI> <LI> Samples were extracted using a modified EN-QuEChERS method followed by LC–MS/MS analysis. </LI> <LI> Satisfactory extraction efficiency was obtained using 0.1% formic acid in acetonitrile as an extractant. </LI> <LI> None of the market samples tested positive for the target analyte. </LI> <LI> The developed method can help establish the MRL of bithionol, which is yet to be set by regulatory authorities. </LI> </UL> </P>

      • Momordica cochinchinensis Seed Extracts Suppress Migration and Invasion of Human Breast Cancer ZR-75-30 Cells Via Down-regulating MMP-2 and MMP-9

        Objective: Metastases and invasion are the main reasons for oncotherapy failure. Momordica cochinchinensis (Mu Bie Zi in Chinese) had been used for a variety of purposes, and shown anti-cancer action. In this article, we focused on effects on regulation of breast cancer cell ZR-75-30 metastases and invasion by extracts of Momordica cochinchinensis seeds (ESMCs). Methods: Effect of ESMCs on ZR-75-30 human breast cancer cells proliferation were evaluated by MTT assay and on invasion and migration by wound-healing and matrigel invasion chamber assays. Expression and protease activity of two matrix metalloproteinases (MMPs), MMP-2 and MMP-9, were analyzed by Western blotting and gelatin zymography, respectively. Results: ESMC revealed strong growth inhibitory effects on ZR-75-30 cells, and effectively inhibited ZR-75-30 cell invasion in a dose-dependent manner. Western blot and gelatin zymography analysis showed that ESMC significantly inhibited the expression and secretion of MMP-2 and MMP-9 in ZR-75-30 cells. Conclusions: ESMC has the potential to suppress the migration and invasion of ZR-75-30 cancer cells, and it might prove to of interest in the development of novel inhibitors for breast cancer.

        Critical Short Circuit Ratio Analysis on DFIG Wind Farm with Vector Power Control and Synchronized Control

        The introduction of renewable energy sources into the AC grid can change and weaken the strength of the grid, which will in turn affect the stability and robustness of the doubly-fed induction generator (DFIG) wind farm. When integrated with weak grids, the DFIG wind turbine with vector power control often suffers from poor performance and robustness, while the DFIG wind turbine with synchronized control provides better stability. This paper investigates the critical short circuit ratios of DFIG wind turbine with vector power control and synchronized control, to analyze the stability boundary of the DFIG wind turbine. Frequency domain methods based on sensitivity and complementary sensitivity of transfer matrix are used to investigate the stability boundary conditions. The critical capacity of DFIG wind farm with conventional vector power control at a certain point of common coupling (PCC) is obtained and is further increased by employing synchronized control properly. The stability boundary is validated by electromagnetic transient simulation of an offshore wind farm connected to a real regional grid.

        Filamin A (FLNA) is known to act as platform for the signaling and intracellular trafficking of various GPCRs including dopamine D<SUB>2</SUB> and D<SUB>3</SUB> receptors (D<SUB>2</SUB>R, D<SUB>3</SUB>R). To understand molecular mechanisms involved in the FLNA-mediated regulation of D<SUB>2</SUB>R and D<SUB>3</SUB>R, comparative studies were conducted on the signaling and intracellular trafficking of the D<SUB>2</SUB>R and D<SUB>3</SUB>R in FLNA-knockdown cells, with a specific focus on the roles of the proteins that interact with FLNA and the D<SUB>2</SUB>R and D<SUB>3</SUB>R. Lowering the level of cellular FLNA caused an elevation in RalA activity and resulted in selective interference with the normal intracellular trafficking and signaling of the D<SUB>2</SUB>R and D<SUB>3</SUB>R, through GRK2 and β-arrestins, respectively. Knockdown of FLNA or coexpression of active RalA interfered with the recycling of the internalized D<SUB>2</SUB>R and resulted in the development of receptor tolerance. Active RalA was found to interact with GRK2 to sequester it from D<SUB>2</SUB>R. Knockdown of FLNA or coexpression of active RalA prevented D<SUB>3</SUB>R from coupling with G protein. The selective involvement of GRK2- and β-arrestins in the RalA-mediated cellular processes of the D<SUB>2</SUB>R and D<SUB>3</SUB>R was achieved via their different modes of interactions with the receptor and their distinct functional roles in receptor regulation. Our results show that FLNA is a multi-functional protein that acts as a platform on which D<SUB>2</SUB>R and D<SUB>3</SUB>R can interact with various proteins, through which selective regulation of these receptors occurs in combination with GRK2 and β-arrestins.

        Short-Term Outcomes and Safety of Computed Tomography-Guided Percutaneous Microwave Ablation of Solitary Adrenal Metastasis from Lung Cancer: A Multi-Center Retrospective Study

        Objective: To retrospectively evaluate the short-term outcomes and safety of computed tomography (CT)-guided percutaneous microwave ablation (MWA) of solitary adrenal metastasis from lung cancer. Materials and Methods: From May 2010 to April 2014, 31 patients with unilateral adrenal metastasis from lung cancer who were treated with CT-guided percutaneous MWA were enrolled. This study was conducted with approval from local Institutional Review Board. Clinical outcomes and complications of MWA were assessed. Results: Their tumors ranged from 1.5 to 5.4 cm in diameter. After a median follow-up period of 11.1 months, primary efficacy rate was 90.3% (28/31). Local tumor progression was detected in 7 (22.6%) of 31 cases. Their median overall survival time was 12 months. The 1-year overall survival rate was 44.3%. Median local tumor progression-free survival time was 9 months. Local tumor progression-free survival rate was 77.4%. Of 36 MWA sessions, two (5.6%) had major complications (hypertensive crisis). Conclusion: CT-guided percutaneous MWA may be fairly safe and effective for treating solitary adrenal metastasis from lung cancer.
