Background: This study investigated the survival benefit of radiotherapy (RT) of the supra- and infraclavicular lymphatic drainage area in Chinese women with T1-2N1M0 breast cancer receiving mastectomy. Methods: A total of 593 cases were retrospectively reviewed from 1998 to 2007. The relationship between supra- or infraclavicular fossa relapse (SCFR) and post-operative RT at the supra-/infraclavicular lymphatic drainage area was evaluated. Results: The majority of patients (532/593; 89. 8%) received no RT while 61 patients received RT. The median follow-up was 85 months. Among patients without RT, 54 (10. 2%) developed recurrence in the chest wall or ipsilateral SCFR. However, none of the 61 patients who underwent RT demonstrated SCFR. One patient who received RT (1. 6%) experienced recurrence in the chest wall. Univariate analysis revealed that age and molecular subtype (both P < 0. 05) were two prognostic factors related to supraclavicular and infraclavicular fossa relapse-free survival (SFRFS). Multivariate analysis revealed that only Her-2 positive status (P = 0. 011) was an independent predictor of SFRFS. RT had no influence on distant metastasis (P = 0. 328) or overall survival (P = 0. 541). SCFR significantly affected probability of distant metastasis (P < 0. 001) and overall survival (P < 0. 001). Conclusion: Although RT was not significantly associated with SFRFS, postoperative RT was significantly associated with a lower locoregional (i. e., supraclavicular/infraclavicular and chest wall) recurrence rate. SCFR significantly influenced distant metastasis-free survival, which significantly influenced the overall survival of T1-2N1M0 breast cancer patients after mastectomy. Thus, prophylactic RT is recommended in T1-2N1M0 breast cancer patients, especially those who have Her-2 positive lesions.

Transient receptor potential melastain 7 (TRPM7) is a bifunctional protein with dual structure of both ion channel and protein kinase, participating in a wide variety of diseases including cancer. Recent researches have reported the mechanism of TRPM7 in human cancers. However, the correlation between TRPM7 and prostate cancer (PCa) has not been well studied. The objective of this study was to investigate the potential the role of TRPM7 in the apoptosis of PC-3 cells, which is the key cell of advanced metastatic PCa. In this study, we demonstrated the influence and potential function of TRPM7 on the PC-3 cells apoptosis induced by TNF-related apoptosis inducing-ligand (TRAIL). The study also found a novel up-regulated expression of TRPM7 in PC-3 cells after treating with TRAIL. Suppression of TRPM7 by TRPM7 non-specific inhibitors ($Gd^{3+}$ or 2-aminoethoxy diphenylborate (2-APB) ) not only markedly eliminated TRPM7 expression level, but also increased the apoptosis of TRAIL-treated PC-3 cells, which may be regulated by the phosphatidylinositol 3-kinase/protein kinase B (PI3K/AKT) signaling pathway accompany with up-regulated expression of cleaved Caspase-3, (TRAIL-receptor 1, death receptors 4) DR4, and (TRAIL-receptor 2, death receptors 5) DR5. Taken together, our findings strongly suggested that TRPM7 was involved in the apoptosis of PC-3 cells induced by TRAIL, indicating that TRPM7 may be applied as a therapeutic target for PCa.

An endophytic Streptomyces sp. (AC-2) was isolated from the root of Cistanches deserticola Y.C.Ma.. Chemical investigations of the culture broth of AC-2 afforded fifteen compounds including K1115 A (1), tyrosol (2), phenylethylamine derivatives (3, 4), cyclic dipeptides (5-8), nucleosides and their aglycones (9-13), N-acetyltryptamine (14), and pyrrole-2-carboxylic acid (15). Only tyrosol can promote an increase of intracellular cAMP special on GPR12 transfected cells, such as CHO and HEK293, which means it may be a possible ligand for GPR12.

<P>A novel displacement reaction has been observed to occur between conducting polymers (CP) and metal salts which can be used to fabricate nanostructured CP–metal composites in a one-pot manner. Vanadium pentoxide (V <SUB>2</SUB>O<SUB>5</SUB>) nanofiber is used during the synthesis as the reactive seeds to induce the nanofibril CP–metal network formation. The CP–metal nanocomposites exhibit excellent sensory properties for hydrogen peroxide (H<SUB>2</SUB>O<SUB>2</SUB>) detection, where both high sensitivity and a low detection limit can be obtained. The sensory performance of the CP–metal composite can be further enhanced by a facile microwave treatment. It is believed that the CP–metal nanofibril network can be converted to a carbon–metal network by a microwave-induced carbonization process and result in the sensory enhancement. </P>

Background/Aims: The application of glycated hemoglobin (HbA1c) for the diagnosis of diabetes is currently under extensive discussion. In this study, we explored the validity of using HbA1c as a screening and diagnostic test in Chinese subjects recruited in Nanjing, China. Methods: In total, 497 subjects (361 men and 136 women) with fasting plasma glucose (PG) ≥ 5.6 mmol/L were recruited to undergo the oral glucose tolerance test (OGTT) and HbA1c test. Plasma lipid, uric acid, and blood pressure were also measured. Results: Using a receiver operating characteristic curve, the optimal cutoff point of HbA1c related to diabetes diagnosed by the OGTT was 6.3%, with a sensitivity and specificity of 79.6% and 82.2%, respectively, and the area under the curve was 0.87 (95% confidence interval, 0.83 to 0.92). A HbA1c level of 6.5% had a sensitivity and specificity of 62.7% and 93.5%, respectively. When comparing the HbA1c ≥ 6.5% or OGTT methods for diagnosing diabetes, the former group had significantly higher HbA1c levels and lower levels of fasting and 2-hour PG than the latter group. No significant difference was observed in the other metabolism indexes between the two groups. Conclusions: Our results suggest that HbA1c ≥ 6.5% has reasonably good specificity for diagnosing diabetes in Chinese subjects, which is in concordance with the American Diabetes Association recommendations.

Rhein lysinate (RHL) is the salt of lysine andrhein and the objective of this study was to investigate theprotection of RHL to liver in diabetic mice. The model oftype 2 diabetes was established by high-fat diet andstreptozotocin treatment. Malondialdehyde, superoxidedismutase (SOD) and glutathione peroxidase (GSH-Px)were measured using a spectrophotometer. Inflammatoryfactors (TNF-a and IL-6) and related proteins (ERK1/2 andSREBP-1c) were analyzed by Western blot. Tissue profilewas determined by hematoxylin and eosin staining andaccumulation of fat was examined by Nile red staining. The results indicated that plasma glucose levels of type 2diabetic mice were over 13.9 mM. Compared with modelgroup, plasma glucose levels were decreased, howeverinsulin levels were increased in RHL (25 and 50 mg/kg)-treated group. Elevated plasma triglyceride and cholesterolwere also markedly attenuated after RHL treatment. Theactivities of SOD and GSH-Px of livers were increasedafter RHL treatment. Livers of RHL-treated mice had morenormal structure and less steatosis than that of diabeticmice. Moreover, RHL decreased the expression of TNF-aand IL-6 and the phosphorylation of SREBP-1c and ERK1/2. In conclusion, RHL has a noticeable hepatic protectionin diabetic mice.

Objective: To explore the application of multiplex nested methylated specific polymerase chain reaction (PCR) in the early diagnosis of epithelial ovarian carcinoma (EOC). Materials and Methods: Serum and fresh tissue samples were collected from 114 EOC patients. RUNX3, TFPI2 and OPCML served as target genes. Methylation levels of tissues were assessed by multiplex nested methylated specific PCR, the results being compared with those for carcinoma antigen 125 (CA125). Results: The serum free deoxyribose nucleic acid (DNA) methylation spectrum of EOC patients was completely contained in the DNA spectrum of cancer tissues, providing an accurate reflection of tumor DNA methylation conditions. Serum levels of CA125 and free DNA methylation in the EOC group were evidently higher than those in benign lesion and control groups (p<0.05). Patients with early EOC had markedly lower serum CA125 than those with advanced EOC (p<0.05), but there was no significant difference in free DNA methylation (p>0.05). The sensitivity, specificity and positive predicative value (PPV) of multiplex nested methylated specific PCR were significantly higher for detection of all patients and those with early EOC than those for CA125 (p<0.05). In the detection of patients with advanced EOC, the PPV of CA125 detection was obviously lower than that of multiplex nested methylated specific PCR (p>0.05), but there was no significant difference in sensitivity (p>0.05). Conclusions: Serum free DNA methylation can be used as a biological marker for EOC and multiplex nested methylated specific PCR should be considered for early diagnosis since it can accurately determine tumor methylation conditions.

Regeneration and functional recovery of facial nerves after injury are still far from satisfaction. Studies on whether Schwann cells of injured facial nerves could secrete neurotrophic factors to promote nerve regeneration are rare, and that is the pbjective of this study. Schwann cells from Wallerian degenerating segments of rats facial nerves were isolated and cultured in conditioned culture medium, collected, concentrated and fractionated by ultra-filtration. The neurotrophic bioactivity of the soluble secretions from cultured Schwann cells were examined on an established neuron culture model. Dil fluorescence labeling facial motoneurons from neonatal rats, MTT assay, and image analysis for survival of facial motoneurons were used. The results show that facial motoneuron survival activity in Schwann cell culture medium containing a molecular weight larger than 30 kDa component, was significantly higher than in medium with and without serum (p<0.05 and p<0.01 respectively). The effect could be extinguished by either boiling or trypsinizing. At the 2nd, 4th and 7th days after conditioned culture, mean perikaryon area and neurite length of facial motoneurons in a medium containing molecular weight larger than 30kDa component, were significantly larger than in serum containing medium. The results show that Schwann cells from Wallerian degenerating facial nerves in serum-free and neurite re-growth of facial motoneurons in vitro. The results also suggest that the neurotrophic effect might be derived from protein(s) or peptide(s) with molecular weight larger than 30 kDa component of the soluble secretions. It may be potent for functional repair of facial verve and other neural disorders.
