Species Specific Antiviral Activity of Porcine Interferon-α8 (IFNα8)

<P>Interferons (IFNs) have been known as antiviral genes and they are classified by type 1, type 2, and type 3 IFN. The type 1 IFN consists of IFNα, IFNβ, IFNτ, and IFNω whereas the type 2 IFN consists of only IFNγ, which is a key cytokine driving T helper cell type 1 immunity. IFNλ belongs to the type 3 IFN, which is also known as IL-28 and IL-29 possessing antiviral activities. Type 1 IFN is produced by viral infection whereas type 2 IFN is induced by mitogenic or antigenic T-cell stimuli. The IFNτ of bovine was first discovered in an ungulate ruminant recognition hormone. IFNτ belongs to the type 1 IFN with the common feature of type 1 IFN such as antiviral activity. IFNs have been mostly studied for basic research and clinical usages therefore there was no effort to investigate IFNs in industrial animals. Here we cloned porcine IFNα8 from peripheral blood mononuclear cells of Korean domestic pig (<I>Sus scrofa domestica</I>). The newly cloned IFNα8 amino acid sequence from Korean domestic pig shares 98.4% identity with the known porcine IFNα8 in databank. The recombinant porcine IFNα8 showed potent antiviral activity and protected bovine Madin-Darby bovine kidney epithelial (MDBK) cells from the cytopathic effect of vesicular stomatitis virus, but it failed to protect human Wistar Institute Susan Hayflick (WISH) cells and canine Madin-Darby canine kidney epithelial-like (MDCK) cells. The present study demonstrates species specific antiviral activity of porcine IFNα8.</P>

<P>Allogenic adipose-derived mesenchymal stem cells (Ad-MSCs) are an alternative source for cytotherapy owing to their antioxidant and anti-inflammatory effects. Frozen-thawed allogenic Ad-MSCs can be used instantly for this purpose. However, the viability and function of frozen-thawed Ad-MSCs have not been clearly evaluated. The purpose of this study was to compare the viability and function of Ad-MSCs and heme oxygenase-1 (HO-1)-overexpressed Ad-MSCs <I>in vitro</I> after freeze-thawing. The viability, proliferation, antioxidant capacity and mRNA gene expression of growth factors were evaluated. Frozen-thawed cells showed significantly lower viability than fresh cells (77% for Ad-MSCs and 71% for HO-1 Ad-MSCs, <I>P</I><0.01). However, the proliferation rate of frozen-thawed Ad-MSCs increased and did not differ from that of fresh Ad-MSCs after 3 days of culture. In contrast, the proliferation rate of HO-1-overexpressed Ad-MSCs was lower than that of Ad-MSCs. The mRNA expression levels of <I>TGF-β</I>, <I>HGF</I> and <I>VEGF</I> did not differ between fresh and frozen-thawed Ad-MSCs, but <I>COX-2</I> and <I>IL-6</I> had significantly higher mRNA expression in frozen cells than fresh cells (<I>P</I><0.05). Fresh Ad-MSCs exhibited higher <I>HO-1</I> mRNA expression than frozen-thawed Ad-MSCs, and fresh HO-1-overexpressed Ad-MSCs exhibited higher than fresh Ad-MSCs (<I>P</I><0.05). However, there was no significant difference between fresh and frozen HO-1-overexpressed Ad-MSCs. The antioxidant capacity of HO-1-overexpressed Ad-MSCs was significantly higher than that of Ad-MSCs. Cryopreservation of Ad-MSCs negatively affects viability and antioxidant capacity, and HO-1-overexpressed Ad-MSCs might be useful to maximize the effect of Ad-MSCs for cytotherapy.</P>

<P>The aim of this study was to investigate the in vitro osteogenic capacity of bone morphogenetic protein 7 (BMP-7) overexpressing adipose-derived (Ad-) mesenchymal stem cells (MSCs) sheets (BMP-7-CS). In addition, BMP-7-CS were transplanted into critical-sized bone defects and osteogenesis was assessed. BMP-7 gene expressing lentivirus particles were transduced into Ad-MSCs. BMP-7, at the mRNA and protein level, was up-regulated in BMP-7-MSCs compared to expression in Ad-MSCs. Osteogenic and vascular-related gene expressions were up-regulated in BMP-7-CS compared to Ad-MSCs and Ad-MSC sheets. In a segmental bone-defect model, newly formed bone and neovascularization were enhanced with BMP-7-CS, or with a combination of BMP-7-CS and demineralized bone matrix (DBM), compared to those in control groups. These results demonstrate that lentiviral-mediated gene transfer of BMP-7 into Ad-MSCs allows for stable BMP-7 production. BMP-7-CS displayed higher osteogenic capacity than Ad-MSCs and Ad-MSC sheets. In addition, BMP-7-CS combined with demineralized bone matrix (DBM) stimulated new bone and blood vessel formation in a canine critical-sized bone defect. The BMP-7-CS not only provides BMP-7 producing MSCs but also produce osteogenic and vascular trophic factors. Thus, BMP-7-CS and DBM have therapeutic potential for the treatment of critical-sized bone defects and could be used to further enhance clinical outcomes during bone-defect treatment.</P>
Mapping potential landscapes of semiconducting carbon nanotubes with scanning gate microscopy

<P>Inhomogeneities in the energy bands of semiconducting carbon nanotubes (CNTs) are investigated with scanning gate microscopy (SGM). Non-uniform responses of a semiconducting CNT to a local electric field are found in SGM images which are obtained at the different depletion states of the CNT. The results indicate that local potential perturbations affect the transport characteristics of semiconducting CNTs significantly, especially near the off-state of the device.</P>
Power Controlled Concurrent Transmissions in mmWave WPANs

<P>This letter considers power-controlled transmission from directional antennas in mmWave wireless personal area network (WPAN) systems. The attributes of these systems are studied; these include the number of concurrent transmissions and the power consumption with different system parameters, such as the antenna's beamwidth and radiating efficiency. Numerical results are presented to show that the power controlled transmission enables more concurrent transmissions than the non-power controlled transmission. The results also show that the number of concurrent transmissions increases as the beamwidth and the path loss component become smaller and the antenna's radiating efficiency increases. In addition, the power controlled system generally uses less power than the non-power controlled transmission set up; the overall analysis is verified by simulation.</P>

The shock absorber is installed on the suspension device of the vehicle and enters the outside, and foreign substances such as dust and soil are introduced from the road surface while driving to form scratches on the piston rod, which may damage or breakage the function of the shock absorber, so a bellows-shaped dust cover is installed. In this treatise, a product development in accordance with an optimal design that prevents buckling of a dust cover for a shock absorber is described.
Delay and Throughput Performance of Uncompressed Video Streaming Service in mmWave WPAN

Existing WPAN solutions (IEEE 802.11 a/b/g, UWB) only have limited bandwidth and cannot support uncompressed video transmission. Instead, 60㎓ millimeter-wave(mmWave) band is an attracting method because it can provide huge bandwidth and achieve very high data rate(over 3Gbps). In this paper, we present the comparison of different superframe sizes in terms of delay and throughput for uncompressed video streaming in mmWave WPANs. We use the Constant Bit Rate (CBR) model to emulate the 1080p (30f 20b) video streaming. For simplification, we assume that an error-free PHY wireless channel is adopted. Simulation results show that the superframe size of 1/60s can result in better performance than that of 2㎳ for the uncompressed video service in mmWave WPAN. It is necessary to increase superframe size and use aggregation method, in order to achieve high throughput.
