lncRNA Epigenetic Landscape Analysis Identifies <i>EPIC1</i> as an Oncogenic lncRNA that Interacts with MYC and Promotes Cell-Cycle Progression in Cancer

        Wang, Zehua,Yang, Bo,Zhang, Min,Guo, Weiwei,Wu, Zhiyuan,Wang, Yue,Jia, Lin,Li, Song,Caesar-Johnson, Samantha J.,Demchok, John A.,Felau, Ina,Kasapi, Melpomeni,Ferguson, Martin L.,Hutter, Carolyn M.,Sof Cell Press 2018 Cancer Cell Vol. No.

<P><B>Summary</B></P> <P>We characterized the epigenetic landscape of genes encoding long noncoding RNAs (lncRNAs) across 6,475 tumors and 455 cancer cell lines. In stark contrast to the CpG island hypermethylation phenotype in cancer, we observed a recurrent hypomethylation of 1,006 lncRNA genes in cancer, including <I>EPIC1</I> (epigenetically-induced lncRNA1). Overexpression of <I>EPIC1</I> is associated with poor prognosis in luminal B breast cancer patients and enhances tumor growth <I>in vitro</I> and <I>in vivo.</I> Mechanistically, <I>EPIC1</I> promotes cell-cycle progression by interacting with MYC through <I>EPIC1</I>'s 129–283 nt region. <I>EPIC1</I> knockdown reduces the occupancy of MYC to its target genes (e.g., <I>CDKN1A</I>, <I>CCNA2</I>, <I>CDC20</I>, and <I>CDC45</I>). MYC depletion abolishes <I>EPIC1</I>'s regulation of MYC target and luminal breast cancer tumorigenesis <I>in vitro</I> and <I>in vivo</I>.</P> <P><B>Highlights</B></P> <P> <UL> <LI> LncRNAs show a hypomethylation phenotype, in contrast to a CIMP phenotype in cancer </LI> <LI> <I>EPIC1</I> promotes breast tumorigenesis through regulating cancer cell-cycle progression </LI> <LI> <I>EPIC1</I> directly interacts with MYC protein through <I>EPIC1</I>'s 129–283 nt region </LI> <LI> <I>EPIC1</I> regulates MYC targets by enhancing MYC occupancy on its target promoters </LI> </UL> </P> <P><B>Graphical Abstract</B></P> <P>[DISPLAY OMISSION]</P>

Tidal Effect on Grouting in Karst Fracture with Flowing Water: Experimental Investigation and Its Application

        Qing Jin,Zehua Bu,Dong-dong Pan,Xiaojie Gao,Pengshuai Yang,Haiyan Li,Zhaofeng Li,Zhenhao Xu 대한토목학회 2023 KSCE Journal of Civil Engineering Vol.27 No.2

To address the problem on grouting control of water inflow disaster in coastal area, a caisson-type grouting experimental system of a single fracture in flowing water under tidal action was developed independently. Through the tide simulation system, the tidal action of different intensity and frequency was effectively described. Tide simulation system, fracture system, grouting system, water supply system and information monitoring system were combined to realize the visualization of the whole grouting process under tidal action. Furthermore, the grouting experiments of a plate fracture under different tidal action were developed and the blocking mechanism under tidal action was revealed. The conclusions were as follow: 1) Under constant water head, the slurry had three typical diffusion patterns, which were circular distribution, double U-shaped distribution and elliptical distribution respectively. 2) Under tidal action, the pressure showed a periodic fluctuation growth, and the growth rate slowed down with the increase of fracture aperture. 3) Strong tidal action would promote the diffusion of and increased the outflow amount of slurry, which was not conducive to grouting. Based on the water inflow treatment project of limestone mine in Guangxi, Through the field grouting tests, the 2# water inflow point was blocked. The research results have a certain reference value for the technology optimization of grouting engineering in coastal areas.

Structural and component characterization of the B4C neutron conversion layer deposited by magnetron sputtering

        Zhu Jingtao,Liu Yang,Zhou Jianrong,Yang Zehua,Zhu Hangyu,Zhou Xiaojuan,Tan Jinhao,Cui Mingqi,Sun Zhijia 한국원자력학회 2023 Nuclear Engineering and Technology Vol.55 No.9

Neutron conversion detectors that use 10B-enriched boron carbide are feasible alternatives to 3 He-based detectors. We prepared boron carbide films at micron-scale thickness using direct-current magnetron sputtering. The structural characteristics of natural B4C films, including density, roughness, crystallization, and purity, were analyzed using grazing incidence X-ray reflectivity, X-ray diffraction, X-ray photoelectron spectroscopy, time-of-flight secondary ion mass spectrometry, and scanning electron microscopy. A beam profile test was conducted to verify the practicality of the 10B-enriched B4C neutron conversion layer. A clear profile indicated the high quality of the neutron conversion of the boron carbide layer.

Simultaneous Single-Position Oblique Lateral Interbody Fusion Combined With Unilateral Percutaneous Pedicle Screw Fixation for Single-Level Lumbar Tuberculosis: A 3-Year Retrospective Comparative Study

        Jinyue He,Jiezhong Deng,Yusheng Yang,Tingting Zheng,Fei Luo,Jianzhong Xu,Zehua Zhang 대한척추신경외과학회 2023 Neurospine Vol.20 No.4

Objective: To illustrate a simultaneous single-position oblique lateral interbody fusion (SPOLIF) combined with unilateral percutaneous pedicle screw fixation in treating single-level lumbar tuberculosis, compared with posterior-only approach in clinical and radiographic evaluations. Methods: Consecutive patients who had undergone surgeries for single-level lumbar tuberculosis from January 2018 to December 2020 were retrospectively reviewed. The patients included were divided into SP-OLIF and posterior-only groups according to surgical methods applied, with follow-up for at least 36 months. Outcomes included estimated blood loss, operative time, and complications for safety evaluation; visual analogue scale (VAS), Oswestry Disability Index (ODI) for efficacy evaluation; erythrocyte sedimentation rate (ESR) and C-reactive protein (CRP) for evaluating tuberculosis activity; x-ray and computed tomography scan were used for radiographic evaluation. Results: A total of 136 patients had been enrolled in the study (60 for SP-OLIF and 76 for Posterior-only). The median operative time, blood loss, and hospital stay in SP-OLIF group were significantly less, with a lower complication rate. Meanwhile, the SP-OLIF group showed substantially lower VAS in 1 and 7 days and decreased ODI in the first month postoperatively, without significant difference afterward. Similarly, the median CRP and ESR in SP-OLIF group were significantly lower in 3 and 7 days postoperatively. All indicators had reduced to normal after 3 months. No recurrence had been reported throughout the whole follow-up. Conclusion: SP-OLIF was an efficient minimally invasive protocol for single-level lumbar tuberculosis, facilitating earlier clinical improvement, with decreased blood loss, operative time and hospital stay compared with posterior-only approach.
