Comparison between residents with a 6-year medical program and a 7-year medical program in terms of objective structured clinical examination performance in postgraduate year training in Taiwan: a 2-group pre- and post-test non-synchronized study

        Ya-Ting Chang,Ying-Ying Yang,Chung-Pin Li,Chen-Huan Chen 한국보건의료인국가시험원 2022 보건의료교육평가 Vol.19 No.-

Purpose In 2013, medical schools in Taiwan implemented a 6-year medical program that replaced the previous 7-year medical education program. The postgraduate year (PGY) program was also extended from 1 year to 2 years. The new program is characterized by diversified teaching, integration of medical skills, a system-oriented curriculum, and the implementation of primary care and clinical thinking training. The purpose of this study was to examine whether postgraduate residents who learned under the new program have better patient care skills than those who learned under the previous program. Methods Of 101 residents in the PGY program at Taipei Veterans General Hospital, 78 were trained in the 6-year program, while 23 were trained in the 7-year program. During the PGY training, 2 objective structured clinical examinations (OSCEs) were used to evaluate clinical reasoning, communication skills, and procedural skills at the beginning of the training and after 11 months of training, respectively. The scores of each OSCE and the rate of improvement of the pre- and post-tests were analyzed. Results Residents trained in the new program scored higher on clinical reasoning (P<0.001) and the total scores of the 3 tested skills (P=0.019) on the pre-test. In terms of improvement, residents educated in the previous system improved more in clinical reasoning than those educated in the new education system. Conclusion The new medical education program, which emphasizes clinical thinking, improved residents’ clinical skills. The PGY program was effective in improving the clinical performance of residents who were educated in the previous system.

Fungichromin Production by Streptomyces padanus PMS-702 for Controlling Cucumber Downy Mildew

        Ya-Ting Fan,Kuang-Ren Chung,Jenn-Wen Huang 한국식물병리학회 2019 Plant Pathology Journal Vol.35 No.4

Streptomyces padanus PMS-702 strain produces a polyene macrolide antibiotic fungichromin and displays antagonistic activities against many phytopathogenic fungi. In the present study, experimental formulations were assessed to improve the production of fungichromin, the efficacy of PMS-702 on the suppression of sporangial germination, and the reduction of cucumber downy mildew caused by Pseudoperonospora cubensis. PMS-702 strain cultured in a soybean meal-glucose (SMG) medium led to low levels of fungichromin accumulation and sporangial germination suppression. Increasing medium compositions and adding plant oils (noticeably coconut oil) in SMG significantly increased fungichromin production from 68 to 1,999.6 μg/ml. Microscopic examination reveals that the resultant suspensions significantly reduced sporangial germination and caused cytoplasmic aggregation. Greenhouse trials reveal that the application of PMS-702 cultural suspensions reduced downy mildew severity considerably. The addition of Tween 80 into the synthetic medium while culturing PMS-702 further increased the suppressive efficacy of downy mildew severity, particularly when applied at 24 h before inoculation or co-applied with inoculum. Fungichromin at 50 μg/ml induced phytotoxicity showing minor necrosis surrounded with light yellowish halos on cucumber leaves. The concentration that leads to 90% inhibition (IC90) of sporangial germination was estimated to be around 10 μg/ml. The results provide a strong possibility of using the S. padanus PMS-702 strain as a biocontrol agent to control other plant pathogens.

Sport Gender Ideology, Past Contact Experiences and Attitudes toward Sexual Minority Athletes in Taiwan

        Ya-Ting Shang,Chu-Min Liao,Diane L. Gill 숙명여자대학교 아시아여성연구원 2012 Asian Women Vol.28 No.3

The purpose of this study was to examine sport gender ideology (belief that men in sport have to show their masculinity and women their femininity) and past contact experiences as predictors of coaches’ and athletes’ attitudes toward sexual minority athletes. Surveys were completed by 315 athletes and 94 coaches from college varsity teams in Taiwan. Hierarchical regression results showed that, for male athletes and female coaches, lower levels of sport gender ideology and positive previous experiences with homosexuals predicted more positive attitudes toward sexual minority athletes. For female athletes and male coaches, positive experiences with sexual minorities were associated with a more positive attitude toward gay and lesbian athletes. The different results may reflect position expectations/role constraints that lead male athletes and female coaches to abide by masculinity-dominated formal and informal rules. For female athletes, attitudes toward sexual minority athletes may be a self-expressive function of other values, such as justice.

An Investigation Into the Relationship Between Metabolic Responses and Energy Regulation in Antibody-Producing Cell

        Ya Ting Sun,Liang Zhao,Zhao Yang Ye,Li Fan,Xu Ping Liu,Wen Song Tan 한국미생물생명공학회(구 한국산업미생물학회) 2013 Journal of microbiology and biotechnology Vol.23 No.11

Energy-efficient metabolic responses were often noted in high-productive cultures. To better understand these metabolic responses, an investigation into the relationship between metabolic responses and energy regulation was conducted via a comparative analysis among cultures with different energy source supplies. Both glycolysis and glutaminolysis were studied through the kinetic analyses of major extracellular metabolites concerning the fast and slow cell growth stages, respectively, as well as the time-course profiles of intracellular metabolites. In three cultures showing distinct antibody productivities, the amino acid metabolism and energy state were further examined. Both the transition of lactate from production to consumption and steady intracellular pools of pyruvate and lactate were observed to be correlated with efficient energy regulation. In addition, an efficient utilization of amino acids as the replenishment for the TCA cycle was also found in the cultures with upregulated energy metabolism. It was further revealed that the inefficient energy regulation would cause low cell productivity based on the comparative analysis of cell growth and productivity in cultures having distinct energy regulation.

Metabolic engineering of erythritol production from glycerol by Yarrowia lipolytica

        Ya-Ting Wang,Ling-Xuan Zhao,Liu-Jing Wei,Jun Chen,Zhijie Liu,Feng Liu,Qiang Hua 한국생물공학회 2024 Biotechnology and Bioprocess Engineering Vol.29 No.1

Erythritol as a four-carbon polyol has been widely used in food, pharmaceutical and daily chemical industries with characteristics of low caloric value and high chemical stability. Here, a system metabolic engineering strategy was used to increase the yield of erythritol from glycerol in Yarrowia lipolytica by enhancing the substrate transformation and restricting the by-product synthesis. Specifi cally, we determined that over-expression of a newly identifi ed erythrose reductase YPR1 was able to improve the erythritol production as same as the well-known erythrose reductase ER27. Instead of its up-regulation, knockout of erythrose reductase ER10 was eff ective to improve erythritol synthesis. Moreover, both over-expression of YPR1 and deletion of ER10 signifi cantly accelerated the glycerol utilization in response to high osmotic stress. To further decrease the by-product accumulation, a restriction and recycling strategy was implemented by knockout of mannitol dehydrogenase MDH2 and enhancement of arabitol dehydrogenase ADH1 and fructokinase HXK1. The engineered strain YL13 produced a titer of 25 g/L erythritol and less than 0.5 g/L mannitol and arabitol. By over-expression of transketolase TKL1, the fi nal strain YL14 produced 28.5 g/L erythritol and none of mannitol and arabitol. This study provides a new idea for reducing the production of by-products and improving the glycerol conversion to erythritol.

Involvement of Alternative Oxidase in the Regulation of Sensitivity of Sclerotinia sclerotiorum to the Fungicides Azoxystrobin and Procymidone

        Ting Xu,Ya-Ting Wang,Wu-Sheng Liang,Fei Yao,Yong-Hong Li,Dian-Rong Li,Hao Wang,Zheng-Yi Wang 한국미생물학회 2013 The journal of microbiology Vol.51 No.3

Sclerotinia sclerotiorum is a filamentous fungal pathogen that can infect many economically important crops and vegetables. Alternative oxidase is the terminal oxidase of the alternative respiratory pathway in fungal mitochondria. The function of alternative oxidase was investigated in the regulation of sensitivity of S. sclerotiorum to two commercial fungicides, azoxystrobin and procymidone which have different fungitoxic mechanisms. Two isolates of S. sclerotiorum were sensitive to both fungicides. Application of salicylhydroxamic acid, a specific inhibitor of alternative oxidase, significantly increased the values of effective concentration causing 50% mycelial growth inhibition (EC50) of azoxystrobin to both S. sclerotiorum isolates, whereas notably decreased the EC50 values of procymidone. In mycelial respiration assay azoxystrobin displayed immediate inhibitory effect on cytochrome pathway capacity, but had no immediate effect on alternative pathway capacity. In contrast, procymidone showed no immediate impact on capacities of both cytochrome and alternative pathways in the mycelia. However, alternative oxidase encoding gene (aox) transcript and protein levels, alternative respiration pathway capacity of the mycelia were obviously increased by pre-treatment for 24 h with both azoxystrobin and procymidone. These results indicate that alternative oxidase was involved in the regulation of sensitivity of S. sclerotiorum to the fungicides azoxystrobin and procymidone, and that both fungicides could affect aox gene expression and the alternative respiration pathway capacity development in mycelia of this fungal pathogen.

Chinese Translation and Validation of the Child and Adolescent Sleep Checklist for Parents

        Yang Ya-Ting,Chang Han-Yun,Hsu Chung-Yao,Lin Cheng-Yu,Zeitzer Jamie M. 대한수면학회 2020 sleep medicine research Vol.11 No.2

Background and ObjectiveaaThe Child and Adolescent Sleep Checklist (CASC) for parents was originally developed and published in Japanese and has served as a valid and effective tool for the assessment of a parent’s perspective of their adolescent child’s sleep behaviors. To allow for comparisons of such perspectives among parents of different cultures, and with different linguistic backgrounds, reliable and valid measurement tools are necessary. The purpose of this study was to translate and validate a Chinese language version of this questionnaire. MethodsaaInternal consistency, factor structure, and criterion validity data were tested using 497 parent/adolescent dyads in Taiwan. Exploratory factor analysis (EFA) and internal consistency reliability were examined in a subset of the cohort (n = 241), while criterion validity and conduct confirmatory factor analysis (CFA) was examined in a second subset of the cohort (n = 256). Preliminary criterion validity was established through a comparison with a measure of chronotype, Morningness- Eveningness Scale for Children. ResultsaaEFA resulted in four factors, consistent with the original version, with the four factors accounting for 60% of the total variance. The results of CFA presented good fit to the data. The questionnaire demonstrated also acceptable to good internal consistency (α = 0.84–0.89). Preliminary criterion validity was evident as the total score and each of the subscale scores on the questionnaire were significantly higher (worse sleep-related behavior) in evening-types. ConclusionsaaThe Chinese language version of the CASC for parents appears to be a psychometrically sound measure to evaluate sleep behaviors in Chinese-speaking adolescents.
