The objective of this study was to analyze the target genes and regulatory function of miR-34a in Megalobrama amblycephala using second-generation highthroughput sequencing and bioinformatic tools. Functional enrichment analysis was performed by gene ontology. MiR- 34a and target gene expression levels were measured in M. amblycephala fed normal and high-carbohydrate diets. The results revealed that miR-34a was highly conserved in several species, and miR-34a of M. amblycephala has a close evolutionary relationship to that of zebrafish and common carp. miRanda, TargetScan, RNAhybrid predicted 5,185, 6,282 and 2,168 target genes, respectively, and 645 target genes were in common. According to annotation information, the target genes were enriched in phosphate metabolism, glycerophospholipid metabolism, Golgi vesicle transport, cell division, and other biological processes (P < 0.05). Pathway enrichment analysis revealed that these target genes were mainly enriched in alpha-linolenic acid and linoleic acid metabolism, ether lipid metabolism, VEGF signaling pathway, Fc epsilon RI signaling pathway, GnRH signaling pathway, and MAPK signaling pathway (P < 0.05). The regulatory role of miR-34a was more significant in the liver than in the brain of M. amblycephala. MiR-34a regulates glucose lipid homeostasis induced by high glucose diets by upregulating hepatic PI3K/Akt, FOXO, and TOR signaling pathways.
BMB Reports : Over-expression of OsHsfA7 enhanced salt and drought tolerance in transgenic rice

Heat shock proteins play an important role in plant stress tolerance and are mainly regulated by heat shock transcription factors (Hsfs). In this study, we generated transgenic rice over-expressing OsHsfA7 and carried out morphological observation and stress tolerance assays. Transgenic plants exhibited less, shorter lateral roots and root hair. Under salt treatment, over-expressing OsHsfA7 rice showed alleviative appearance of damage symptoms and higher survival rate, leaf electrical conductivity and malondialdehyde content of transgenic plants were lower than those of wild type plants. Meanwhile, transgenic rice seedlings restored normal growth but wild type plants could not be rescued after drought and re-watering treatment. These findings indicate that over-expression of OsHsfA7 gene can increase tolerance to salt and drought stresses in rice seedlings. [BMB Reports 2013; 46(1): 31-36]

Two genes coding for eukaryotic translation initiation factors, eIF4E.a and eIF4E.c, were isolated from twelve accessions of Chinese cabbage (Brassica rapa L. ssp. pekinensis). Polymorphism analysis revealed that 94and 142 polymorphic sites were characterized from allele of BraeIF4E.a and BraeIF4E.c which produced complex haplotype structures. Six novel haplotypes were characterized from the two alleles respectively. Among the six novel haplotypes of BraeIF4E.a, three loss-of-function mutations were identified in which a conserved single nucleotide deletion mutation cause the early termination of BraeIF4E.a coding product; while for six new BraeIF4E.c haplotypes, their coding product show amino acid substitution mutations on non-conservative amino acid residues which might affect TuMV infection in Chinese cabbage.

Myostatin represses muscle growth by negatively regulating the number and size of muscle fibers. Myostatin loss-of-function can result in the double-muscling phenotype and increased muscle mass. Thus, knockout of myostatin gene could improve the quality of meat from mammals. In the present study, zinc finger nucleases, a useful tool for generating gene knockout animals, were designed to target exon 1 of the myostatin gene. The designed ZFNs were introduced into porcine primary fibroblasts and early implantation embryos via electroporation and microinjection, respectively. Mutations around the ZFNs target site were detected in both primary fibroblasts and blastocysts. The proportion of mutant fibroblast cells and blastocyst was 4.81% and 5.31%, respectively. Thus, ZFNs can be used to knockout myostatin in porcine primary fibroblasts and early implantation embryos.
Expression Profiles of Class A Rice Heat Shock Transcription Factor Genes Under Abiotic Stresses

Expression profiles of 12 class A rice heat shock transcription factor genes (OsHsfAs) were analyzed by semi-quantitative reverse transcriptase polymerase chain reaction. The OsHsfA genes exhibited tissue-specific expressions under normal condition. OsHsfA1a, A2d, and A9 were predominantly expressed in young spike. Expression responses of the 12 OsHsfAs under abiotic stresses were analyzed in the shoots of rice seedling. Most OsHsfA genes responded quickly to heat stress except for OsHsfA1a, A3,and A9 which were almost unaffected. In particular,OsHsfA2a expression in response to heat stress was highest among the heat shock factors examined. However, the majority of the increased OsHsfAs expression responses to salt, polyethylene glycol (PEG), and cold treatments primarily occurred during the later stages (3 to 24 h) of stress exposure. Furthermore, most of OsHsfA gene expressions were little affected and only a few (OsHsfA3, A4d, A7,and A9) genes had slow responses to cold treatment. The results indicate that the transcript levels of OsHsfAs during heat stress exposure were distinct from those of plants subjected to salt, PEG, and cold stresses, suggesting that there might be different regulatory networks between heat and non-heat stress.

BackgroundHuman immunodeficiency virus (HIV)-negative plasmablastic lymphoma (PBL) is a rare entity of diffuse large B-cell lymphoma (DLBCL). The clinicopathological features of and optimal treatment for HIV-negative PBL remain largely unknown.MethodsTo gain insight into this distinct lymphoma, we summarized the clinicopathologic charac-teristics of 8 unpublished HIV-negative PBLs and performed a comprehensive review of 394 published cases.ResultsOf the 8 unpublished PBLs, the median patient age was 53.0 years. Four patients pre-sented with stage IV disease. All 8 patients showed a plasma cell-like immunophenotype. Of the six patients who received anthracycline-based chemotherapy, including two who received bortezomib, three patients achieved a continuous complete response, two pa-tients died due to disease progression, and one patient was lost to follow-up. The other two patients achieved continuous complete response after receiving chemotherapy com-bined with radiotherapy and surgery. Of the 402 patients, the majority were male, with a mean age of 58.0 years. EBV infection was detected in 55.7% of the patients. The median survival times of the patients who received CHOP or CHOP-like regimens and intensive regimens were not reached and 23.0 months, respectively, and the intensive regimen did not improve the survival outcome (P=0.981). Multivariate analysis showed that EBER remained the only independent factor affecting overall survival (OS).ConclusionHIV-negative PBL is a distinct entity with a predilection for elderly and immunosup-pressed individuals. Intensive chemotherapy had no apparent survival benefits over the CHOP regimen in terms of OS; the prognosis of this disease is poor with current chemo-therapy methods, and treatment remains a challenge.
