        Spatial protein expression of Panax ginseng by in-depth proteomic analysis for ginsenoside biosynthesis and transportation

        Background: Panax ginseng, as one of the most widely used herbal medicines worldwide, has been studied comprehensively in terms of the chemical components and pharmacology. The proteins from ginseng are also of great importance for both nutrition value and the mechanism of secondary metabolites. However, the proteomic studies are less reported in the absence of the genome information. With the completion of ginseng genome sequencing, the proteome profiling has become available for the functional study of ginseng protein components. Methods: We optimized the protein extraction process systematically by using SDS-PAGE and one-dimensional liquid chromatography mass spectrometry. The extracted proteins were then analyzed by two-dimensional chromatography separation and cutting-edge mass spectrometry technique. Results: A total of 2,732 and 3,608 proteins were identified from ginseng root and cauline leaf, respectively, which was the largest data set reported so far. Only around 50% protein overlapped between the cauline leaf and root tissue parts because of the function assignment for plant growing. Further gene ontology and KEGG pathway revealed the distinguish difference between ginseng root and leaf, which accounts for the photosynthesis and metabolic process. With in-deep analysis of functional proteins related to ginsenoside synthesis, we interestingly found the cytochrome P450 and UDP-glycosyltransferase expression extensively in cauline leaf but not in the root, indicating that the post glucoside synthesis of ginsenosides might be carried out when growing and then transported to the root at withering. Conclusion: The systematically proteome analysis of Panax ginseng will provide us comprehensive understanding of ginsenoside synthesis and guidance for artificial cultivation.

        Overexpressing osa-miR171c decreases salt stress tolerance in rice

        The miRNA171 family is one of the well-conservedmiRNA families, and its role under stresses is not knownexcept its expression on genome-wide expression analyses. osa-miR171c was induced by high concentration of salt (150mM NaCl). A rice dh mutant with osa-miR171c overexpressiontriggered by a T-DNA insertion, significantly decreased salttolerance at the stages of germination and seedling. Thisphenotype was confirmed by osa-miR171c overexpressiontransgenic rice. Compared with wild-type (WT), dh mutantreduced amounts of free proline and increased the water lossrate after salt treatment. Stomatal density in the leafepidermis of dh mutant also increased. Moreover, dh mutantincreased sensitivity to ABA treatment. Several stressresponsivegenes were down-regulated in dh mutant than inWT under salty stress. These results indicate that osamiR171cis involved in modulating physiological changes,stomatal development, ABA-dependent pathways and expressionof stress-related genes; thereby, it possibly contributes tosalty tolerance.

      • The Construction of the Trust Relationship between the Platform and the Consumer in Mutual Economic Environment

        The mutual aid economy mainly comes from the sharing economy. The “mutual aid economy” model of taking out personal idle funds to share with others to obtain the most significant economic benefits is an area that the international community is currently striving to explore. After the establishment of the "Mutual Insurance Commune", parallel to the development of the digital economy and the significant improvement of the material living standards, the mutual aid economy with unique advantages has been experiencing rapid growth in China. Because China has a large population and a large base of the bottom poor groups, the traditional insurance business can no longer meet the needs of these groups. In this context, the network mutual assistance platform bred by the combination of the Internet and the traditional insurance industry has ushered in a development boom. In recent years, however, the Internet mutual aid platform is under a contentious and intensified debate,which has a great relationship with the standard use of funds. Some network mutual aid platforms put forward that they have the right to manage the mutual aid funds stored on the platform, but there is no clear limit on the scope of use. In addition, the information disclosure of some platforms is not transparent enough, which makes the use of mutual aid funds standardized to become a major problem, which severely limits the development of network mutual aid platforms. A lot of news of “xx mutual assistance platform shutting down” has shaken the rationality and longevity of the network mutual aid platform. In addition, due to China's unique domestic political environment and policies, for example, in September this year, The State Council issued the notice of "normalization of collective procurement of medicines”, which will reduce the potential population of mutual insurance and have a certain impact on its development. These show that its stable development has hidden risks that it cannot ignore. Since the closure of Fun in Funding and Water Droplets Mutual Aid in March of this year, two leading mutual-aid platforms, the credibility of the mutual aid economy, the sustainability of online mutual-aid platforms, and the future of mutual aid are all doubtful and faint. Factors such as establishing the trust relationship between the platform and the consumer, the financing of the risk of entering and exiting the risk, the breadth, depth and timeliness of mutual assistance behaviour challenge the survival and development of network mutual assistance platforms. It also means that the transformation and upgrading of the traditional insurance industry in the Internet age is not smooth. However, on the other hand, online mutual aid platforms have the following advantages: First, mutual aid platforms have outstanding social benefits, which can maximize the benefits of social resources. Second, the fund scale of the network platform is relatively small, and the funding object is scattered. Third, the mutual aid platform's motivational orientation to pursue benefits makes its risks more controllable than other platforms. Also,the introduction of the "Blockchain" technology enhances the transparency and reliability of the mutual assistance mechanism. In the process of mutual-treasure operation, the integration of blockchain technology realizes the establishment of mutual-treasure advantages. This study believes that if we can correctly guide network mutual aid, it can become a new model in financial and insurance innovation. The study aims to provide constructive suggestions to promote the transformation and development of the network mutual aid platform and achieve high-quality development of the mutual aid economy in China. According to the development status of low threshold, high security, low intermediate cost and high self-propagation of mutual aid economy and network mutual aid platform at home and abroad, taking “Xianghubao”, a domestic “late start, fast development, “strong strength” network mutual assistance platform, as the research object. This study focuses on building a trust relationship between the network mutual aid platform and consumers in the mutual aid economy from the normative perspective of using technology and controlling mutual aid funds. Employing semi-structured interviews, questionnaires and data modelling and analysis, the study has explored the prerequisites for the rise and prosperity of the mutual aid economy include increasingly abundant idle resources, the establishment of collaborative network platforms, mutual aid value consensus, industry environment,then offers a deep analysis towards the mutual trust mechanism between the enterprise and the user within “Xianghubao”, to further probe into the logical and critical elements which impact on building the relationship of trust between the network mutual aid platform and consumers in mutual aid economy. Also, the "Blockchain" technology is introduced to enhance the transparency and reliability of the mutual assistance mechanism. Based on the study, the company can make corresponding countermeasures from the four dimensions of internal management, trusted products, risk control, and mutual trust between customers to deal with the reformation and the closure of “Xianghubao”.         The adsorption and difusion of oxygen atom on the O-terminated ZnO ð 000?1 Þ surface havebeen systematically investigated based on ¯rst-principles density functional theory. The resultsshow that the surface relaxation of the ZnO ð 000?1 Þ surface is signi¯cant. In the view of themaximization of the adsorption energy, the preferred site for the adsorption of oxygen atom is thetop-O site above the oxygen atom of the ¯rst Zn – O bilayer. There is chemical bond formedbetween the adsorbed oxygen atom and the oxygen atom on the surface, which will result in theredistribution of the charges. The charges transfer from the ZnO surface to the adsorbed oxygenatom, which will heighten the surface potential of ZnO surface and increase the surface workfunction. Moreover, the di®usion of the oxygen atom on the ZnO surface has also been investi-gated, and the potential barriers of the difusion have been identi¯ed to reveal the adsorptionstability

        Transcriptome-Wide Analyses Provide Insights into Development of the Hedychium coronarium Flower, Revealing Potential Roles of PTL

        The flower of Hedychium coronarium possesses highly specialized floral organs: a synsepalous calyx, petaloid staminodes and a labellum. The formation of these organs is controlled by two gene categories: floral organ identity genes and organ boundary genes, which may function individually or jointly during flower development. Although the floral organogenesis of H. coronarium has been studied at the morphological level, the underlying molecular mechanisms involved in particular organ morphologies that emerge in flower development still remain poorly understood. Here, we used comparative transcriptomics combined with Real-time quantitative PCR to investigate gene expression patterns of ABC-class genes in H. coronarium flowers, as well as the homolog of PETAL LOSS (HcPTL). Our studies found that stamen/petal identity or stamen fertility in H. coronarium was not necessarily correlated with the differential expression of HcAP3 and HcAG. We also found a novel spatio-temporal expression pattern for HcPTL mRNA, suggesting it may have evolved a lineage-specific role in the morphogenesis of the Hedychium flower. Our study provides a new transcriptome reference and a functional hypothesis regarding the role of a boundary gene in organ fusion that should be further addressed through phylogenetic analyzes of this gene, as well as functional studies.
