      • TWO’S COMPANY; THREE’S A CROWD: HOW SOCIAL MEDIA INFLUENCES GARMENT CONSUMPTION IN YOUTH FASHION

        The global apparel market is estimated to increase from US$1,105 billion in 2012 to US$2,110 billion in 2025 (Statista, 2014), with women’s wear accounting for approximately 55% of the total within the United Kingdom (UK) (Verdict, 2015). In 2013 and 2014 market research performed by Mintel identified the ‘shopping experience’ as one of the key factors in women’s shopping habits, noting that consumers aged 16-24, in particular, tended to browse online but to buy in-store, still seeing shopping for clothes as a day out with friends (Mintel, 2013). Nevertheless, the growing influence of social media was noted, and by 2015 this age group had overtaken the 25-34 year olds to become the main online clothes shoppers. Despite this, young people aged 16-24, all of whom potentially qualify as digital natives, still show a preference for shopping in-store (Mintel, 2015), although preliminary research indicates that more of them are now shopping alone than with friends. This suggests that there has been a shift in consumer shopping habits within the youth sector, with less importance being placed on the shopping experience. Given that, this study aims to explore the social media behaviours of the youngers aged between 18-24 with a particular focus on fashion consumption. Crowd sourcing has been identified as an emerging practice found in several sectors including fashion (Yeomans, 2013). Preliminary research suggests that the shift is just a perceived one and that technology savvy young fashion consumers are using smart-technology to upload ‘chelfies’ (‘selfies direct from the changing rooms’) to crowd source opinion about proposed purchases (Soar and Torn, 2015), using social media to replace ‘physical friends’ with ‘digital friends’. To explore this phenomenon further, this study employs qualitative research method and uses semi-structured interviews. The result of this research provides evidence of crowd sourcing behaviours via social media, as well as indicates the complex communication mechanism that embeds within fashion consumption among the generation of the youth.

      • Transcriptomes of major renal collecting duct cell types in mouse identified by single-cell RNA-seq

        <P><B>Significance</B></P><P>A long-term goal in mammalian biology is to identify the genes expressed in every cell type of the body. In the kidney, the expressed genes (i.e., transcriptome) of all epithelial cell types have already been identified with the exception of the cells that make up the renal collecting duct, which is responsible for regulation of blood pressure and body fluid composition. Here, single-cell RNA-sequencing was used in mouse to identify transcriptomes for the major collecting duct cell types: type A intercalated cells, type B intercalated cells, and principal cells. The information was used to create a publicly accessible online resource. The data allowed identification of genes that are selectively expressed in each cell type, which is informative for cell-level understanding of physiology and pathophysiology.</P><P>Prior RNA sequencing (RNA-seq) studies have identified complete transcriptomes for most renal epithelial cell types. The exceptions are the cell types that make up the renal collecting duct, namely intercalated cells (ICs) and principal cells (PCs), which account for only a small fraction of the kidney mass, but play critical physiological roles in the regulation of blood pressure, extracellular fluid volume, and extracellular fluid composition. To enrich these cell types, we used FACS that employed well-established lectin cell surface markers for PCs and type B ICs, as well as a newly identified cell surface marker for type A ICs, c-Kit. Single-cell RNA-seq using the IC- and PC-enriched populations as input enabled identification of complete transcriptomes of A-ICs, B-ICs, and PCs. The data were used to create a freely accessible online gene-expression database for collecting duct cells. This database allowed identification of genes that are selectively expressed in each cell type, including cell-surface receptors, transcription factors, transporters, and secreted proteins. The analysis also identified a small fraction of hybrid cells expressing aquaporin-2 and anion exchanger 1 or pendrin transcripts. In many cases, mRNAs for receptors and their ligands were identified in different cells (e.g., <I>Notch2</I> chiefly in PCs vs. <I>Jag1</I> chiefly in ICs), suggesting signaling cross-talk among the three cell types. The identified patterns of gene expression among the three types of collecting duct cells provide a foundation for understanding physiological regulation and pathophysiology in the renal collecting duct.</P>

        Sunitinib Versus Sorafenib in Advanced Hepatocellular Cancer: Results of a Randomized Phase III Trial

        <P><B>Purpose</B></P><P>Open-label, phase III trial evaluating whether sunitinib was superior or equivalent to sorafenib in hepatocellular cancer.</P><P><B>Patients and Methods</B></P><P>Patients were stratified and randomly assigned to receive sunitinib 37.5 mg once per day or sorafenib 400 mg twice per day. Primary end point was overall survival (OS).</P><P><B>Results</B></P><P>Early trial termination occurred for futility and safety reasons. A total of 1,074 patients were randomly assigned to the study (sunitinib arm, n = 530; sorafenib arm, n = 544). For sunitinib and sorafenib, respectively, median OS was 7.9 versus 10.2 months (hazard ratio [HR], 1.30; one-sided <I>P</I> = .9990; two-sided <I>P</I> = .0014); median progression-free survival (PFS; 3.6 <I>v</I> 3.0 months; HR, 1.13; one-sided <I>P</I> = .8785; two-sided <I>P</I> = .2286) and time to progression (TTP; 4.1 <I>v</I> 3.8 months; HR, 1.13; one-sided <I>P</I> = .8312; two-sided <I>P</I> = .3082) were comparable. Median OS was similar among Asian (7.7 <I>v</I> 8.8 months; HR, 1.21; one-sided <I>P</I> = .9829) and hepatitis B–infected patients (7.6 <I>v</I> 8.0 months; HR, 1.10; one-sided <I>P</I> = .8286), but was shorter with sunitinib in hepatitis C–infected patients (9.2 <I>v</I> 17.6 months; HR, 1.52; one-sided <I>P</I> = .9835). Sunitinib was associated with more frequent and severe adverse events (AEs) than sorafenib. Common grade 3/4 AEs were thrombocytopenia (29.7%) and neutropenia (25.7%) for sunitinib; hand-foot syndrome (21.2%) for sorafenib. Discontinuations owing to AEs were similar (sunitinib, 13.3%; sorafenib, 12.7%).</P><P><B>Conclusion</B></P><P>OS with sunitinib was not superior or equivalent but was significantly inferior to sorafenib. OS was comparable in Asian and hepatitis B–infected patients. OS was superior in hepatitis C–infected patients who received sorafenib. Sunitinib-treated patients reported more frequent and severe toxicity.</P>

      • <i>ITGB6</i> loss-of-function mutations cause autosomal recessive amelogenesis imperfecta

        <P>Integrins are cell-surface adhesion receptors that bind to extracellular matrices (ECM) and mediate cell–ECM interactions. Some integrins are known to play critical roles in dental enamel formation. We recruited two Hispanic families with generalized hypoplastic amelogenesis imperfecta (AI). Analysis of whole-exome sequences identified three <I>integrin beta 6</I> (<I>ITGB6</I>) mutations responsible for their enamel malformations. The female proband of Family 1 was a compound heterozygote with an <I>ITGB6</I> transition mutation in Exon 4 (g.4545G > A c.427G > A p.Ala143Thr) and an <I>ITGB6</I> transversion mutation in Exon 6 (g.27415T > A c.825T > A p.His275Gln). The male proband of Family 2 was homozygous for an <I>ITGB6</I> transition mutation in Exon 11 (g.73664C > T c.1846C > T p.Arg616*) and hemizygous for a transition mutation in Exon 6 of <I>Nance–Horan Syndrome</I> (<I>NHS</I> Xp22.13; g.355444T > C c.1697T > C p.Met566Thr). These are the first disease-causing <I>ITGB6</I> mutations to be reported. Immunohistochemistry of mouse mandibular incisors localized ITGB6 to the distal membrane of differentiating ameloblasts and pre-ameloblasts, and then ITGB6 appeared to be internalized by secretory stage ameloblasts. ITGB6 expression was strongest in the maturation stage and its localization was associated with ameloblast modulation. Our findings demonstrate that early and late amelogenesis depend upon cell–matrix interactions. Our approach (from knockout mouse phenotype to human disease) demonstrates the power of mouse reverse genetics in mutational analysis of human genetic disorders and attests to the need for a careful dental phenotyping in large-scale knockout mouse projects.</P>

        종자발아율, 수량, 그리고 화분발아율 비교를 통한 내서성 고추 자원 선발

        본 연구는 농촌진흥청 국립원예특작과학원 채소과에서 육성 중인 고추 34계통과 세계채소센터 육성 계통과 자원 12점 등 총 46점의 고추를 공시하여, 종자의 발아율, 수량 및 상품과율, 그리고 화분발아율을 조사하여 내서성이 강한 고추 자원을 선발하기 위하여 수행되었다. 1. 종자발아율 비교에서 여름종자는 가을종자 대비 상대적인 피해율은 4.3~100% 범위였고, H42(1.6%), H16(5.0%), H14(6.3%), H02(12.4%), H44(13.7%), H12(15.9%), H03 (17.6%), H17(23.4%) 순으로 대조 계통인 H37(26.2%)에 비해 피해율이 낮았다. 2. 고추 과실의 수량은 34.4~446.5g, 총 과실수는 2.0~134.3 개를 나타냈다. 상품과 비율에 있어서 75% 이상은 H22(95.0%), H16(89.1%), H37(88.4%), H40(86.4%), H6 (86.0), H27(83.7%), H04(83.6%), H03(82.6%), H08(81.1%), H44(81.4%), H02(80.0%), H45(79.1%), H17(78.7%), H19 (77.7%), H34(77.0%), H15(76.9%), H42(76.3%) 순으로 나타 났다. 3. 고온 처리에 의한 화분발아율의 피해율은 H06(59.5%), H14(74.4%), H44(85.3%), H03(90.2%)가 대조구인 H37 (94.7%)과 나머지 다른 고추에 비해 상대적으로 낮았다. 이상의 결과, 가을종자 대비 여름종자 종자발아율의 피해율, 수량 및 상품과율, 그리고 고온 처리에 의한 화분발아율 등을 고려하여, H03, H04, H06, H14, H17, H22, H44, H45 계 통을 내서성이 높은 자원으로 선발하였다. This study was conducted to select heat tolerant pepper accessions in tropical regions. Fruit yield and pollen germination rate between heat stress and normal condition as well as germination rate between seeds harvested in summer and fall were compared using 46 pepper accessions including 34 from Rural Development Administration and 12 from World Vegetable Center. For seed germination rate, the relative injury rate of summer seed ranged between 4.3 to 100%. Among accessions, H42 (1.6%), H16 (5.0%), H14 (6.3%), H02 (12.4%), H44 (13.7%), H12 (15.9%), H03 (17.6%) and H17 (23.4%) showed lower injury rate in seed germination than H37 (26.2%) which is known for its heat tolerance. Fruit yield and number per plant were in the range of between 34.4 to 446.5g and 2.0 to 134.3 fruits, respectively. The marketable fruit ratio ranged between 41.0 to 95.0% and accessions producing marketable fruits more than 75% were H22 (95.0%), H16 (89.1%), H37 (88.4%), H40 (86.4%), H6 (86.0), H27 (83.7%), H04 (83.6%), H03 (82.6%), H08 (81.1%), H44 (81.4%), H02 (80.0%), H45 (79.1%), H17 (78.7%), H19 (77.7%), H34 (77.0%), H15 (76.9%), and H42 (76.3%). The relative injury rate of pollen germination in heat stress condition was low in H06 (59.5%), H14 (74.4%), H44 (85.3%) and H03 (90.2%) compared to a control, H37 (94.9%). Based on the results of seed germination, marketable fruit yield and pollen germination, H03, H04, H06, H14, H17, H22, H44, H45 were selected for heat tolerance in tropical climate.
