        Voxelwise lp-ntPET for detecting localized, transient dopamine release of unknown timing: Sensitivity Analysis and Application to Cigarette Smoking in the PET Scanner

        <P>The “linear parametric neurotransmitter PET” (lp-ntPET) model estimates time variation in endogenous neurotransmitter levels from dynamic PET data. The pattern of dopamine (DA) change over time may be an important element of the brain's response to addictive substances such as cigarettes or alcohol. We have extended the lp-ntPET model from the original region of interest (ROI) - based implementation to be able to apply the model at the voxel level. The resulting endpoint is a dynamic image, or movie, of transient neurotransmitter changes. Simulations were performed to select threshold values to reduce the false positive rate when applied to real <SUP>11</SUP>C-raclopride PET data. We tested the new voxelwise method on simulated data, and finally, we applied it to <SUP>11</SUP>C-raclopride PET data of subjects smoking cigarettes in the PET scanner. In simulation, the temporal precision of neurotransmitter response was shown to be similar to that of ROI-based lp-ntPET (standard deviation ∼ 3 min). False positive rates for the voxelwise method were well controlled by combining a statistical threshold (the <I>F</I>-test) with a new spatial (cluster-size) thresholding operation. Sensitivity of detection for the new algorithm was greater than 80% for the case of short-lived DA changes that occur in subregions of the striatum as might be the case with cigarette smoking. Finally, in <SUP>11</SUP>C-raclopride PET data, DA movies reveal for the first time that different temporal patterns of the DA response to smoking may exist in different subregions of the striatum. These spatiotemporal patterns of neurotransmitter change created by voxelwise lp-ntPET may serve as novel biomarkers for addiction and/or treatment efficacy.</P>

        Effects of Artemisia argyi flavonoids on growth performance and immune function in broilers challenged with lipopolysaccharide

        Objective: This research aimed to study the effects of Artemisia argyi flavonoids (AAF) supplemented in diets on the growth performance and immune function of broiler chickens challenged with lipopolysaccharide (LPS). Methods: A total of one hundred and ninety-two 1-d-old broiler chicks were assigned into 4 treatment groups, which were, respectively, fed a basal diet (control), fed a diet with 750 mg/kg AAF, fed a basal diet, and challenged with LPS, fed a diet with 750 mg/kg AAF, and challenged with LPS. Each treatment had six pens with 8 chicks per pen. On days 14, 16, 18, 20 (stress phase I) and 28, 30, 32, 34 (stress phase II), broilers were injected with LPS (500 ㎍/kg body weight) or an equivalent amount of saline. Results: The results demonstrated that dietary AAF significantly improved the body weight (d 21) and alleviated the decrease of average daily gain in broilers challenged with LPS on d 21 and d 35 (p<0.05). Dietary AAF increased bursa fabricius index, and dramatically attenuated the elevation of spleen index caused by LPS on d 35 (p<0.05). Furthermore, serum interleukin-6 (IL-6) concentration decreased with AAF supplementation on d 21 (p<0.05). Diet treatment and LPS challenge exhibited a significant interaction for the concentration of IL-1β (d 21) and IL-6 (d 35) in serum (p<0.05). Additionally, AAF supplementation mitigated the increase of IL-1β, IL-6 in liver and spleen induced by LPS on d 21 and 35 (p<0.05). This study also showed that AAF supplementation significantly reduced the expression of IL-1β (d 21) and nuclear transcription factor kappa-B p65 (d 21 and 35) in liver (p<0.05), and dietary AAF and LPS treatment exhibited significant interaction for the gene expression of IL-6 (d 21), toll like receptor 4 (d 35) and myeloid differentiation factor 88 (d 35) in spleen (p<0.05). Conclusion: In conclusion, AAF could be used as a potential natural immunomodulator to improve growth performance and alleviate immune stress in broilers challenged with LPS.

        Test and Numerical Investigation Mechanical Behavior of Cable Dome

        Cable dome structure is a kind of space steel structure system with stay cable, hoop cable, ridge cable and brace strut. In order to study the mechanical properties of cable dome with different stay (steel tie rod and steel cable), two 6-m-span cable dome structures were designed and manufactured. One is with steel tie rod (SR), other is with steel cable (SC). Meanwhile, two finite element models, which were identical with the solid model, were established to analyzed the mechanical properties of the cable dome and compared with the test results. The static mechanical properties of the cable domes (SC, SR) with Full-span load and Half- span load were analyzed by test. The results show that the internal forces of the two kinds of cable domes increased with the increased of Full-span load. The vertical displacement of the cable dome (SR) was smaller than that of the cable dome (SC). When the structure was under Half-span load, the stress of the two cable domes on the loading side was relatively complex, and the internal force of the rods on the loading side varies greatly, which indicating that the structure was very sensitive to the asymmetric load. The variation trend between the simulated value and the test value was consistent, which verifies the correctness of the finite element model. Furthermore, the natural frequencies of the structure with three different load state were tested by using the exciter excitation method. The results show that the natural frequencies of the two cables were dense, and the damping ratio of the cable dome (SC) structure was basically in the range of 0.0115–0.0238, and the damping ratio of the cable dome structure (SR) was basically in the range of 0.0081–0.0132, Which can provide reference for similar cable dome structures in practical projects.

      • Multiple damage detection of maglev rail joints using time-frequency spectrogram and convolutional neural network

        Maglev rail joints are vital components serving as connections between the adjacent F-type rail sections in maglev guideway. Damage to maglev rail joints such as bolt looseness may result in rough suspension gap fluctuation, failure of suspension control, and even sudden clash between the electromagnets and F-type rail. The condition monitoring of maglev rail joints is therefore highly desirable to maintain safe operation of maglev. In this connection, an online damage detection approach based on three-dimensional (3D) convolutional neural network (CNN) and time-frequency characterization is developed for simultaneous detection of multiple damage of maglev rail joints in this paper. The training and testing data used for condition evaluation of maglev rail joints consist of two months of acceleration recordings, which were acquired in-situ from different rail joints by an integrated online monitoring system during a maglev train running on a test line. Short-time Fourier transform (STFT) method is applied to transform the raw monitoring data into time-frequency spectrograms (TFS). Three CNN architectures, i.e., small-sized CNN (S-CNN), middle-sized CNN (M-CNN), and large-sized CNN (L-CNN), are configured for trial calculation and the M-CNN model with excellent prediction accuracy and high computational efficiency is finally optioned for multiple damage detection of maglev rail joints. Results show that the rail joints in three different conditions (bolt-loosenesscaused rail step, misalignment-caused lateral dislocation, and normal condition) are successfully identified by the proposed approach, even when using data collected from rail joints from which no data were used in the CNN training. The capability of the proposed method is further examined by using the data collected after the loosed bolts have been replaced. In addition, by comparison with the results of CNN using frequency spectrum and traditional neural network using TFS, the proposed TFSCNN framework is proven more accurate and robust for multiple damage detection of maglev rail joints.

        Compound K attenuates hyperglycemia by enhancing glucagon-like peptide-1 secretion through activating TGR5 via the remodeling of gut microbiota and bile acid metabolism

        Background: Incretin impairment, characterized by insufficient secretion of L-cell-derived glucagon-like peptide-1 (GLP-1), is a defining step of type 2 diabetes mellitus (T2DM). Ginsenoside compound K (CK) can stimulate GLP-1 secretion; however, the potential mechanism underlying this effect has not been established. Methods: CK (40 mg/kg) was administered orally to male db/db mice for 4 weeks. The body weight, oral glucose tolerance, GLP-1 secretion, gut microbiota sequencing, bile acid (BA) profiles, and BA synthesis markers of each subject were then analyzed. Moreover, TGR5 expression was evaluated by immunoblotting and immunofluorescence, and L-cell lineage markers involved in L-cell abundance were analyzed. Results: CK ameliorated obesity and impaired glucose tolerance in db/db mice by altering the gut microbiota, especially Ruminococcaceae family, and this changed microbe was positively correlated with secondary BA synthesis. Additionally, CK treatment resulted in the up-regulation of CYP7B1 and CYP27A1 and the down-regulation of CYP8B1, thereby shifting BA biosynthesis from the classical pathway to the alternative pathway. CK altered the BA pool by mainly increasing LCA and DCA. Furthermore, CK induced L-cell number expansion leading to enhanced GLP-1 release through TGR5 activation. These increases were supported by the upregulation of genes governing GLP-1 secretion and L-cell differentiation. Conclusions: The results indicate that CK improves glucose homeostasis by increasing L-cell numbers, which enhances GLP-1 release through a mechanism partially mediated by the gut microbiota-BA-TGR5 pathway. Therefore, that therapeutic attempts with CK might be useful for patients with T2DM.

        Cell-SELEX Based Identification of an RNA Aptamer for Escherichia coli and Its Use in Various Detection Formats

        Escherichia coli are important indicator organisms, used routinely for the monitoring of water and food safety. For quick, sensitive and real-time detection of E. coli we developed a 2'F modified RNA aptamer Ec3, by Cell-SELEX. The 31 nucleotide truncated Ec3 demonstrated improved binding and low nano-molar affinity to E. coli. The aptamer developed by us out-performs the commercial antibody and aptamer used for E. coli detection. Ec3(31) aptamer based E. coli detection was done using three different detection formats and the assay sensitivities were determined. Conventional Ec3(31)-biotin-streptavidin magnetic separation could detect E. coli with a limit of detection of $1.3{\times}10^6CFU/ml$. Although, optical analytic technique, biolayer interferometry, did not improve the sensitivity of detection for whole cells, a very significant improvement in the detection was seen with the E. coli cell lysate ($5{\times}10^4CFU/ml$). Finally we developed Electrochemical Impedance Spectroscopy (EIS) gap capacitance biosensor that has detection limits of $2{\times}10^4CFU/mL$ of E. coli cells, without any labeling and signal amplification techniques. We believe that our developed method can step towards more complex and real sample application.

        Influence of creep on dynamic behavior of concrete filled steel tube arch bridges

        Concrete creep, while significantly changing the static behaviors of concrete filled steel tube (CFST) structures, do alter the structures' dynamic behaviors as well, which is studied quite limitedly. The attempt to investigate the influence of concrete creep on the dynamic property and response of CFST arch bridges was made in this paper. The mechanism through which creep exerts its influence was analyzed first; then a predicative formula was proposed for the concrete elastic modulus after creep based on available test data; finally a numerical analysis for the effect of creep on the dynamic behaviors of a long-span half-through CFST arch bridge was conducted. It is demonstrated that the presence of concrete creep increases the elastic modulus of concrete, and further magnifies the seismic responses of the displacement and internal force in some sections of the bridge. This influence is related closely to the excitation and the structure, and should be analyzed case-by-case.

        Compression-Friendly Low Power Test Application Based on Scan Slices Reusing

        This paper presents a compression-friendly low power test scheme in EDT environment. The proposed approach exploits scan slices reusing to reduce the switching activity during shifting for test scheme based on linear decompressor. To avoid the impact on encoding efficiency from resulting control data, a counter is utilized to generate control signals. Experimental results obtained for some larger ISCAS’89 and ITC’99 benchmark circuits illustrate that the proposed test application scheme can improve significantly the encoding efficiency of linear decompressor.
