        Genome-wide identification and analysis of Subtilisin-like serine protease gene family in banana (Musa accuminta L.) and their expression under abiotic stresses

        Subtilisin-like serine proteases (SBT), a serine proteolytic enzymes play an important role in plant growth function and during different stresses responses. The systematic analysis of the SBT gene family in Musa acuminate (MaSBT) has been done and their responses to abiotic stresses in banana variety cv. G-9 were also analyzed. Total of 67 MaSBT genes were identified and based on phylogeny these were grouped into five districted subgroups. Cis-acting element analysis indicated that almost all MaSBT promoters contain regulatory elementary related to growth and development, hormonal regulation, and stress responses. The gene structure and domain analysis showed a maximum of seventeen exons and four functional domains in MaSBT. The 42 orthologous genes, 07 MaSBT paralogous genes were also identified through synteny analysis. The Ka /Ks study indicated that four MaSBT paralogous gene pairs were tandemly duplicated, while the other three were segmental duplications. Further, the expression pattern via RNA-seq data revealed that MaSBTs exhibited differential expres- sion specifically in response to the abiotic stress of low nitrogen. and also during the flowering time. The MaSBT-1.7 gene was found involved in the response to salt stress and flowering. These findings establish a cornerstone for future research on banana's salt stress mechanism. The study offers valuable insights into SBT encoding genes, shedding light on their roles in growth, development, and abiotic stress responses

        Study of fertility restoration and genetic diversity of drought-tolerant breeding lines for hybrid rice (Oryza sativa L.) development

        The identifcation of fertility restoration and genetic diversity of drought-tolerant breeding lines will be useful for the development of promising drought-tolerant rice hybrids. The extent of genetic variation of 204 drought-tolerant breeding lines based on the coefcient of parentage (COP) and test crossing with IR 58025A is studied. The linked molecular markers to Rf genes were screened on identifed maintainers and restorers. The average value of COP for 204 drought-tolerant breeding lines was 0.21, which indicates genetic divergence in breeding lines. Four major groups comprising of 81 IR 64 type, 37 Apo type, 28 Vandana type, 6 WAB type and 52 others were identifed. Out of 204 breeding lines, 37 (18.14%) maintainers, 55 (26.96%) partial maintainers, 63 (30.88%) partial restorers and 49 (24.02%) restorers were identifed. Rf genes linked molecular markers RM7466 and RM443 on chr. 1; RM6344 on chr. 7 and RM6100, RM1108, RM216, RM171 and RM244 on chr. 10 were found associated with restorers and maintainers. These Rf genes linked molecular markers might be furthermore used in marker-assisted restorer selection in cytoplasmic male sterile-WA system from large source nurseries to avoid usual test crosses in the hybrid rice development programme. The identifed restorer and maintainers may be used for the development of high-yielding drought-tolerant rice hybrids.

        Use of Gamma-Irradiated Amniotic Membrane for the Healing of Split Skin Graft Donor Site

        The management of split skin graft donor site is targeted towards promoting the healing process, while minimizing adverse effects and complications. The aim was to study the efficacy of gamma-irradiated amniotic membrane dressing for the healing of split skin graft donor site. Amniotic membranes isolated from the placenta were processed and sterilized by gamma radiation. Thirty patients with split skin graft donor site were included in the study. The average age of the patients was 33.33±16.45 years in study group and 38.93±18.52 years in control group. The average ulcer size was 92.8±66.2 cm2 in study group and 82.5±56.0 cm2 in control group. Processed amniotic membranes were applied just after taking split skin graft. Split skin graft donor site was assessed on day 8and 12 for epithelialization, pain during change of dressing, discharge from donor site, status of healing and any adverse reactions. Significant (p < 0.0001) reduction in pain during change of amniotic membrane dressing as compared to conventional dressing was observed. The average per day rate of epithelialization on split skin graft donor site was 10.31% in the study group and 7.31% in the control group (p > 0.05). Significant (p < 0.001) enhancement in the healing of donor site was observed with amniotic membrane dressing as compared to the paraffin gauze dressing. The patients with processed gamma-irradiated amniotic membrane experienced no pain during change of dressing and improved epithelialization.

      • Exploring the Utility of Vague Concept for Uncertainty and Hesitation Management

        In the realistic situation, there are many areas which contain imprecisely specified data. This imprecise data indicates the presence of vagueness, incompleteness and uncertainty which causes the problem during important decision-making task. The present paper focuses on the problem of mining important inference from supermarket basket data (in the presence of vagueness). The paper specifically studies the usefulness of vague set theory for the exploration of hesitation information and vague association rules. The hesitation information of an item plays a vital role in making selling strategies for the exhilaration of business. For this purpose, the vague set concept can be used as an important tool which can assist in the identification of hesitated item. The vague set theory with its two membership function provides more intuitive way to interact with the vague situation that causes the hesitation for any item. The effectiveness of the hesitated pattern and rule provide advanced decision-making capabilities that transform ‘almost sold’ items to ‘sold items’.

        Performance enhancement and environmental analysis of vapor compression refrigeration system with dedicated mechanical subcooling

        The primary focus of this study is on the energy, exergy, and environmental (3E) analysis of a dedicated mechanical subcooled vapor compression refrigeration (DMS-VCR) system for applications involving commercially available water chillers that employ R134a (in both subcooler and main cycle). For a cooling capacity of 100-kW water chillers, the mathematical model of the DMS-VCR system is built to determine the performance parameter of the system. The DMS-VCR system reduces electricity usage by 15.52% and increase in COP by 9.5%, which results in a significant reduction in CO 2 emissions of about 15.55%. When compared to equivalent vapor compression refrigeration system (VCRS), the system’s exergetic efficiency is also increased by 8%. Since the computer simulation results will undoubtedly give design engineers a better option, the subcooling and superheating of the vapor compression refrigeration system become alluring in this study. Consequently, the DMS-VCR system performs better as per the combined 3E study.         Metagenomics Analysis of Thrombus Samples Retrieved from Mechanical Thrombectomy

        Purpose: The purpose of this study was to assess the microbiota in middle cerebral artery thrombi retrieved in mechanical thrombectomy arising out of symptomatic carotid plaque within 6 hours of acute ischemic stroke. Thrombi were subjected to next-generation sequencing for a bacterial signature to determine their role in atherosclerosis.Materials and Methods: We included 4 human middle cerebral artery thrombus samples (all patients were male). The median age for the patients was 51±13.6 years. Patients enrolled in the study from Pacific Medical University and Hospital underwent mechanical thrombectomy in the stroke window period. All patients underwent brain magnetic resonance angiography (MRA) and circle of Willis and neck vessel MRA along with the standard stroke workup to establish stroke etiology. Only patients with symptomatic carotid stenosis and tandem lesions with ipsilateral middle cerebral artery occlusion were included in the study. Thrombus samples were collected, stored at –80 degrees, and subjected to metagenomics analysis.Results: Of the 4 patients undergoing thrombectomy for diagnosis with ischemic stroke, all thrombi recovered for bacterial DNA in qPCR were positive. More than 27 bacteria were present in the 4 thrombus samples. The majority of bacteria were <i>Lactobacillus, Stenotrophomonas, Pseudomonas, Staphylococcus</i>, and <i>Finegoldia</i>.Conclusion: Genesis of symptomatic atherosclerotic carotid plaque leading to thromboembolism could be either due to direct mechanisms like acidification and local inflammation of plaque milieu with lactobacillus, biofilm dispersion leading to inflammation like with pseudomonas fluorescence, or enterococci or indirect mechanisms like Toll 2 like signaling by gut microbiota.
