Background: We investigated whether wogonin and apigenin significantly affect the epidermal growth factor receptor (EGFR) signaling pathway involved in MUC5AC mucin gene expression, and production from cultured airway epithelial cells; this was based on our previous report that apigenin and wogonin suppressed MUC5AC mucin gene expression and production from human airway epithelial cells. Methods: Confluent NCI-H292 cells were pretreated with wogonin or apigenin for 15 minutes or 24 hours and then stimulated with epidermal growth factor (EGF) for 24 hours or the indicated periods. Results: We found that incubation of NCI-H292 cells with wogonin or apigenin inhibited the phosphorylation of EGFR. The downstream signals of EGFR such as phosphorylation of MEK1/2 and ERK1/2 were also inhibited by wogonin or apigenin. Conclusion: The results suggest that wogonin and apigenin inhibits EGFR signaling pathway, which may explain how they inhibit MUC5AC mucin gene expression and production induced by EGF.
Use of Dried Carrot Meal (DCM) in Laying Hen Diet for Egg Yolk Pigmentation

Dried carrot meal (DCM) prepared from fresh carrot was found to be a good source of xanthophyll (54 mg/kg) and morderate source of protein (188.3 g/kg) and energy (2,510 kcal/kg) with low level of fibre (80 g/kg). DM, EE and NFE content of DCM were 895, 35 and 661.7 g/kg respectively. The ingredient was used in laying hen diet to evaluate its pigmenting value for egg yolk. Thirty-two laying hens from Starcross strain were divided into four groups and fed four different diets: a control diet containing 62% ground wheat, a corn-based diet (50% ground yellow corn), control + 4% DCM and control + 8% DCM. Use of DCM at 8% level in layer mash significantly improved yolk colour at 3rd, 6th and 9th week of supplementation in comparison with wheat-based control diet. This improvement was statististically similar to that obtained from yellow corn-based diet. DCM at 4% level also improved yolk colour score. Wheat-based diet significantly increased feed consumption compared to yellow corn and diet supplied with 8% DCM. Body weight gain, egg production, feed conversion were not significantly affected due to dietary addition of DCM and no mortality was observed during 63-day experimental period. Further works on DCM are suggested.

The temperature and the field dependences of the magnetization have been measured for Fe73:5-xCrxCu1Nb3Si13:5B9 (0 ≤ x ≤ 17.5) prepared by using a melt spin technique. The temperature dependence of the magnetization in the range of 5 to 300 K with an applied field of 1 Tesla for all the compositions follows the relaxation M(T) = M0(1-BT3/2 -CT5/2), which is indicative of the presence of a spin wave excitation. The saturation magnetizations at 5 and 300 K decrease linearly with Cr concentration for the entire composition range, which indicates that a simple dilution of the Fe moment takes place due to Cr substitution. The field-cooled (FC) and the zero-field-cooled (ZFC)magnetizations measured with an applied field of 1 Oe show divergences for x = 10, 12.5, 15 and 17.5, which may be attributed to the magnetic hardening at low temperatures.

We conducted this study to investigate whether berberine significantly affects MUC5AC mucin gene expression and mucin production induced by epidermal growth factor (EGF), phorbol 12-myristate 13-acetate (PMA) or tumor necrosis factor-α (TNF-α) from human airway epithelial cells. Confluent NCI-H292 cells were pretreated with varying concentrations of berberine for 30 min and then stimulated with EGF, PMA or TNF-α for 24 h. MUC5AC mucin gene expression and mucin production were measured by reverse transcription-polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assay (ELISA), respectively. Berberine was found to inhibit the expression of MUC5AC mucin gene induced by EGF, PMA or TNF-α. Berberine also inhibited the production of MUC5AC mucin protein stimulated by the same inducers. This result suggests that berberine can regulate the expression of mucin gene and production of mucin protein, by directly acting on human airway epithelial cells.

Dendroclimatological Investigation of High Altitude Himalayan Conifers and Tropical Teak In India

A wide tree-ring data network from Western Himalayan region as well as from Central and Peninsular India have been established by the Indian Institute of Tropical Meteorology (IITM), Pune, India. This includes several ring width and density chronologies of Himalayan conifers (Pinus, Picea, Cedrus, Abies)covering entire area of Western Himalaya and teak (Tectona grandis L.F.) from central and peninsular India. Many of these chronologies go back to $15^{th}$ century. Tree-ring based reconstructed pre-monsoon (March-April-May) summer climate of Western Himalaya do not show any significant increasing or decreasing trend since past several centuries. High altitude tree-ring chronologies near tree line-glacier boundary are sensitive to the winter temperature. Unprecedented higher growth in recent decades is closely associated with the warming trend over the Himalayan region. Dendroclimatic analysis of teak (Tectona grandis) from Central and Peninsular India show significant relationship with pre-monsoon and monsoon climate. Moisture index over the region indicates strong association with tree-ring variations rather than the direct influence of rainfall. It is evident that, two to three consecutive good monsoon years are capable of maintaining normal or above normal tree growth, even though the following year is low precipitation year.

We evaluated the cardioprotective effect of Aloe vera gel isoprenaline (ISO)-administered myocardial infarction in rats. ISO administration increased lipid peroxidation and oxidative stress in rats, which were ameliorated by A. vera gel supplementation. Our study also revealed that creatine kinase-MB (CK-MB) activities were increased in ISO-administered rats, while the activities of cellular antioxidants, such as superoxide dismutase and catalase, and glutathione concentration were decreased. A. vera gel lowered CK-MB enzyme activities and the glutathione concentration in ISO-administered rats, and increased antioxidant activities. Histopathological examination also revealed increases in thickness of the left ventricle myocardium, increases in mononuclear cell infiltrations, increased degeneration of focal areas of the endocardium, and increased fibrous tissue deposition in the heart of ISO-administered rats; whereas, A. vera prevented infiltration of inflammatory cells and reduced left ventricular fibrosis. In conclusion, we show that A. vera supplementation protects against development of cardiac inflammation, fibrosis, and oxidative stress in ISO-administered rats.

In the present study, we investigated whether apigenin significantly affects tumor necrosis factor-α (TNF-α)-induced production and gene expression of MUC5AC mucin in airway epithelial cells. Confluent NCI-H292 cells were pretreated with apigenin for 30 min and then stimulated with TNF-α for 24 h or the indicated periods. The MUC5AC mucin gene expression and mucin protein production were measured by reverse transcription - polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assay (ELISA), respectively. Apigenin significantly inhibited MUC5AC mucin production and down-regulated MUC5AC gene expression induced by TNF-α in NCI-H292 cells. To elucidate the action mechanism of apigenin, effect of apigenin on TNF-α-induced nuclear factor kappa B (NF-κB) signaling pathway was also investigated by western blot analysis. Apigenin inhibited NF-κB activation induced by TNF-α. Inhibition of inhibitory kappa B kinase (IKK) by apigenin led to the suppression of inhibitory kappa B alpha (IκBα) phosphorylation and degradation, p65 nuclear translocation. This, in turn, led to the down-regulation of MUC5AC protein production in NCI-H292 cells. Apigenin also has an influence on upstream signaling of IKK because it inhibited the expression of adaptor protein, receptor interacting protein 1 (RIP1). These results suggest that apigenin can regulate the production and gene expression of mucin through regulating NF-κB signaling pathway in airway epithelial cells.

Background: We investigated whether chrysin affected MUC5AC mucin production and gene expression induced by phorbol ester (phorbol 12-myristate 13-acetate, PMA) or epidermal growth factor (EGF) from human airway epithelial cells. Methods: Confluent NCI-H292 cells were pretreated with varying concentrations of chrysin for 30 minutes, and were then stimulated with PMA and EGF for 24 hours, respectively. MUC5AC mucin gene expression and mucin protein production were measured by reverse transcription polymerase chain reaction and enzyme-linked immunosorbent assay. Results: Concentrations of $10{\mu}M$ and $100{\mu}M$ chrysin were found to inhibit the production of MUC5AC mucin protein induced by PMA; A concentration of $100{\mu}M$ chrysin also inhibited the production of MUC5AC mucin protein induced by EGF; $100{\mu}M$ chrysin inhibited the expression of MUC5AC mucin gene induced by PMA or EGF. The cytotoxicity of chrysin was checked by lactate dehydrogenase assay, and there was no cytotoxic effect observed for chrysin. Conclusion: These results suggest that chrysin can inhibit mucin gene expression and the production of mucin protein by directly acting on airway epithelial cells.
