      • 水萄 F₂世代에서 水熱病 抵抗性의 分離와 他形質 分離와의 關係

        水稻 品種間 組合에서 稻熱病 抵抗性의 分離와 他形質分離와의 關係를 檢討할 目的으로 稻熱病에 抵抗性인 長稈 메벼 品種과 罹病性인 短稈 찰벼 品種間 組合, 抵抗性인 普通葉 系統과 罹病性인 圓滑葉 系統間 組合 및 맑은 쌀의 抵抗性 品種과 罹病性 찰벼 品種間 組合을 供試하여 F2 世代에서 形質의 分離를 調査하고 그 해당個體의 F3 種子로 稻熱病 檢定苗垈에서 稻熱病 抵抗性 分離를 後代檢定하였다. 그 結果를 要約하면 다음과 같다. 1. 供試 組合中 同一한 罹病性 母本과의 組合이라도 抵抗性 母本에 따라 抵抗性의 分離 程度에 差가 있었다. 2. Tadukan과 Tetep으로부터 由來된 抵抗性을 가진 系統들을 母本으로한 組合에서는 Tadukan과 Tetep을 直接 母本으로한 組合에서 보다 抵抗性이 적게 分離 되었다. 3. 毛茸이 없는 圓滑葉特性은 毛茸이 있는 普通葉特性에 對하여, 찰은 메에 對하여, 맑은 쌀(clear)은 心腹白쌀에 對하여 單純劣性으로 分離되었다. 4. 本 試驗에 供試된 組合에 있어서 稻熱病 抵抗性의 分離는 稈長의 分離, 圓滑葉特性의 分離, 찰메의 分離 및 米粒心腹白의 分離와 各各 相互 獨立的인 것으로 나타났다. Investigations were made on the relationship between blast resistance and other agronomical traits such as culm length, glabrousness, endosperm characters for F2 generations in the crosses of eight resistant varieties and two susceptible varities to blast disease of rice plant in the farm nursery. Significant differences were observed in segregation of resistant lines to blast depending on the crosses. In the progeny of the crosses with the original resistant parents of Tetep and Tandukan to susceptible parents the resistant lines were more found than in the crosses between the resistant lines deriving from Tetep and Tadukan and susceptiple parents. Resistant lines to blast in the farm nursery was independently segregated with culm length, glabrousness, waxiness and chalkiness, respectively, in this experiment.

        Quantitative Analysis and Varietal Difference of Cyanidin 3-glucoside in Pigmented Rice

        The cyanidin 3-glucoside (C3G) extracted from pigmented rice seeds in 0.5% TFA (Trifluoro acetic acid) -95% ethanol was separated by High Performance Liquid Chromatography (HPLC). A HPLC system using a Develosil ODS-5 column and 0.1 % TFA-$H_2O$~0.1 % TFA-$CH_3CN$ gradient elution was selected for separation and quantitative determination of C3G. Regression equation obtained for the standard content of C3G pigment was as Y=21.95293$^*$X-14.726771 (r=0.99$^{**}$). Using this method, 326 domestic and introduced collections were evaluated for the C3G content. The Korean bred cultivar 'Heugjinjubyeo', showed highest C3G content (552 mg/100g seed) among the tested cultivars. Among the pigmented rice cultivars ten cultivars were selected for containing a high content of C3G. The content of C3G per 100g seeds was in high order as follows: Heugjinjubyeo (552mg)>Cheng Chang (321mg)>Kilimgeugmi (240mg)>PI160979-2 (224mg)>Hong Shei Lo (221mg)>Heugnambyeo (191 mg)>Mitak =PIl60979-1 (186mg)>Suwon425 (163mg)>Sanghaehyanghyeolla (108mg). The C3G pigment was not detected in the common white rice cultivars.

        벼 雄性細胞 培養에 의한 trisomicsdml 移轉과 培養細胞의 再分化에 미치는 過剩染色體의 影響

        This study was carried out to investigate the transmission frequency of the extra chromosomes in rice trisomic plants through male and female gametes including selfed progeny, and to know the effect of extra chromosomes on the male gametic cell culture of rice. the extra chromosomes were transmitted to next generation at the rate of 0.0% to 9.7% through male gamete and 11.3% to 35.7% through female gamete, although the rate was 27.7% to 40.1% in selfed progeny of each trisomic type. Trisomic plant showed different culturability for callus induction, plant regeneration and green tissue appearence depending on the trisomic types in anther culture. Callus induction rates were ranged from 4.7% to 27.3% and plant regeneration rates 3.0% to 22.0%. In anther culture the highest transmission rate of extra chromosomes was 27.8% in trisomic 4(E), while the extra chromosomes of trisomic 12(A), 8(D) and 7(F) were not transmitted through anther culture in this study. The regenerated plants varied in chromosome numbers.

        水稻品種 “統一”과 “統一찰”의 遺傳背景 Ⅰ. Polyacrylamide gel 等電點電氣泳動에 의한 種子 esterase isozyme의 band 類型

        Polyacrylamide gel isoelectric focusing was used to analyze the grain esterase isozymes of rice varieties, Tongil, Tongilchal and their related parents. Six major bands were revealed in the gel part of high pH and observed different banding patterns. Esterase isozyme banding patterns in grains were separated into three types, namely Est-Ⅰ, Est-Ⅱ and Est-Ⅲ in this experiment. Of the varieties analyzed, Japonica showed Est-Ⅰ and Indica was both Est-Ⅰ and Est-Ⅱ, while Japonica×Indica showed Est-Ⅲ, respectively. Esterase isozyme banding patterns of Tongil, Tongilchal, IR8, IR262, IR833, IR1317 and Peta were identical. It was concluded that the source of esterase isozymes genotype in grains of Tongil and Tongilchal was Indica variety Peta.

        水稻 遺傳子 分析用 遺傳標識基株 育成 Ⅱ. japonica벼 遺傳檢定親으로서 三染色體植物의 育成

        Eleven primary trisomics of the japonica group, developed and made available by lwata and Omura, were crossed as females with Korean bred Seolakbyo. From the F₁, F₂ and BC₁F₁ of each crosses 11 types, of trisomics were identified on the basis of their distinct morphological characters. Leaf character and plant type were useful criteria for the identification of A, E, F and O type trisomics, while B, C, D, G, H, L and N type trisomics were classified by the panicle and glume characters. The female transmission frequency of the extra chromosome of each trisomics ranged from 8.3 to 35% in F₁, from 16.7 to 50% in F₂ and from 14.3 to 50% in BC₁F₁, respectively in this study.

        C3G색소 고함유 벼품종의 자유라디칼 소거작용 및 항염효과

        국내육성 흑자색미(흑진주벼, 흑남벼, C3GHi벼)와 일반미(일품벼, 추청벼, 동진벼) 품종의 메탄을 추출물을 대상으로 항산화 항혈전 항염증활성을 검토하였으며, 그 결과는 다음과 같다. 1. 0.4 mg/mL농도에서 C3GHi벼와 흑진주벼 추출물이 73.25%, 50.38%의 우수한 자유라디칼 소거능을 나타내었다. 2. 메탄올추출물의 혈액응고 저해활성을 측정한 결과, C3GHi 벼는 258.76%, 흑진주벼는 243.52%의 강한 트롬빈 저해활성을 보였다. 그러나 대조약물로 사용한 아스피린보다(398%) 약하게 나타났다. 3. 공시한 흑자색미와 일반미 품종들은 COX-1과 COX-2에 대한 활성이 나타나지 않았으나, C3GHi벼는 PD2에 대한 항염활성을 보였다. 4. H. pylori균에 의해서 나타나는 AGS cell line에 대한 독성을 조사한 결과 흑자색미 추출물은 $250{\mu}g$과 $500{\mu}g$ 처리군에서 모두 H. pylori균의 AGS cell line에 대한 부착을 억제하는 활성이 있는 것으로 판명되었으며 일반미 추출물은 H. pylori 균에 대한 저해작용은 없는 것으로 나타났다. 5. Zone assay에 의한 항균실험결과 흑자색미 추출물은 $200{\mu}g/mL$의 농도에서 ATCC 43504 및 COO1 균주에 대해서 clear zone이 검출되어 항균활성이 있는 것으로 판명되었으나 SEO 균주에 대해서는 항균활성이 검출되지 않았다. Free Radical Scavening and inflammatory of the methanol extracts, which were prepared from 6 difffrent bran of rice, were evaluated to investigate bioactive substances. Among them, the extract of C3GHibyeo and Heugjinjubyeo showed strong DPPH scavening activities (73.25% and 50.38% at 0.4 mg/ml, respectively). The extract of C3GHibyeo and Heugjinjubyeo showed strong thrombin inhibition activities (258.76% and 243.52% inhibition at 5 mg/ml, respectively). The result of antibacterial activity by Zone assay showed that C3GHi rice extract $(250{\mu}g\;and\;500{\mu}g)$ inhibited attachment of Helicobactor pylori on the ATCC48504 and COO1 cell line. But no effect on the SEO cell line. Cytotoxicity of blackish purple rice extract on the H. pylori doesn't showed. These result support a functional superiority of rice-base livelihood, and suggest that the development of healthy food using functional ingredients of rice is possible.

        국문초록 없음 This experiment was carried out to clarify the property of leaf appearance of rice varieties. The 14 remote crossed and the 5 Japonica varieties were tested at the three seeding date: March 26, April 11, and April 25. The results are summarized as follows; 1) Days until turning period of leaf appearance were the same in the remote crossed and Japonica varieties at the first seeding date (March 26), but the days of Japonica varieties were longer than remote crossed varieties at the second and the third seeding dates. Days until turning period was shortened as delay of seeding date in all the tested varieties. 2) The total leaves on main stem ranged from 19.55 to 19.72 in the remote crossed varieties, and from 19.26 to 19.36 in Japonica varieties. 3) The leaf appearance interval (days) until turning period on main stem ranged from 4.75 to 5.51 days in remote crossed varieties, and from 4.96 to 5.75 in Japonica varieties. The leaf appearance interval of remote crossed varieties was more rapid than Japonica varieties. 4) High positive correlation coefficients were obtained between total leaves on main stem and days to heading, total leaves on main stem and no. of leaves at turning point. The correlation between total leaves on main stem and leaf appearance interval, number of leaves at turning point and leaf appearance interval were negatively significant.

        Antioxidant Activity of Blackish Purple Rice

        Anthocyanin in blackish purple rice is composed of cyanidin, peonidin, malvidin, pelargonidin and delphinidin flavylium ion and their glucosides, which shows antioxidant activity similar to a-tocoperol. It has been demonstrated that cyanidin-3-glucoside(C3G) of anthocyanin has a high-potency antioxidant activity, and is a prominent in anticancer and antithrombotic activity. In analysis of content of anthocyanin from the genetic source of blackish purple rice, the level of C3G was approximately 80% of total contents of anthocyanins, and showed the highest content in Heugjinjubyeo, of which contains an amount of 500 mg by weight of 100g brown rice in comparison that most of varieties has less than 50mg by weight of 100g brown rice. We found that C3G pigment in blackish purple rice was considerably affected by cultural season, cultivation area, ripening temperature, and weather condition, etc. as well as genetic properties. In cross combination between Heugjinjubyeo and Suwon425, intermediate parent F9 generation which was three times higher or more in C3G content than that of Heugjinjubyeo was bred, of which it contained an amount of 1678 mg by weight of 100g brown rice, and we called it “C3GHi” Rice. And, we found that the C3GHi rice was more excellent than that of the existing Heugjinjubyeo in anticancer and antithrombotic activity. Methanol extract from Heugjinjubyeo was fractionated by organic solvents in order of n-hexane, CH2Cl2 and n-BuOH, and then oryzafuran, quercetin, vanillic acid and protocatechuic acid and their structures from the n-BuOH fraction were ascertained. Oryzafuran of these compounds was a natural compound found firstly in nature. Quercetin and protocatechuic acid besides new compound oryzafuran showed excellent antioxidant ability to vitamin C. These results suggest that blackish purple rice has very high value as a source of various functional food as well as staple food.
