        Experimental Pathogenesis of Pullorum Disease with the Local Isolate of Salmonella enterica serovar. enterica subspecies Pullorum in Pullets in Bangladesh

        The research work was carried out to study the pathogenesis covering the clinical signs, gross and histopathological lesions in different organs, and reisolation and identification of the organisms after experimental infection with the local isolate of Salmonella enterica serovar. enterica subspecies (S.) Pullorum at different time interval of the experiment during the period February 2006 to December 2006. One hundred pullets (seronegative to S. Pullorum of 12 weeks age were purchased and divided into 5 (A, B, C, D and E) groups and each group consisted of 20 birds. Four groups (A, B, C and D) were infected orally with a dose of 106 CFU, 107 CFU, 2 × 107 CFU, 108 CFU of S. Pullorum, respectively, and one group (E) was treated as uninfected control. The used methods were necropsy and histopathology, culture of bacteria, staining and biochemical test of Salmonella. Five birds from each group were randomly selected and sacrificed 1st week, 2nd, 3rd and 4th weeks of post infection (PI). From all the groups, the bacteriological samples (crop, liver, lung, heart, spleen, bile duodenum, ceca and blood) were collected with pre enriched in buffered peptone water in sterile poly bags. Liver, lungs, heart, spleen, intestine, etc. were collected in 10% buffered-formalin for histopathological examination. No clinical signs, gross and histopathological lesions were found in control group and no S. Pullorum was reisolated. Clinical sign of experimentally infected with S. Pullorum in pullets were loss of appetite (100%), slight depression (75%), ruffled feathers (85%), diarrhea (60%) and loss of weight (100%) in chickens. The feed intake and body weight at different weeks after PI differed significantly (p<0.01) among the groups. Grossly, the highest recorded lesion was button-like ulcer in the ceca (80%) and the lowest was white nodules in lungs (1.25%). S. Pullorum were reisolated from crop (91.25%), liver (91.25%), lung (83.75%), heart (71.25%), spleen (87.75%), bile (33.25%), duodenum (92.50%), ceca (97.50%) and from different group of infection (61.25%). The highest microscopic findings were intestinal and cecal mucosa and submucosa exhibited infiltration of mononuclear cells and congestion (96.25%), and the lowest finding was nodule formation in the lungs (3.75%). The pattern of the disease production by local isolate of S. Pullorum in Bangladesh is almost similar with other isolates in different countries.       • Gibberellin secreting rhizobacterium, Pseudomonas putida H-2-3 modulates the hormonal and stress physiology of soybean to improve the plant growth under saline and drought conditions

        The physiological changes in tolerant soybean plants under salt and drought stress conditions with Pseudomonas putida H-2-3 were investigated. A bacterial isolate H-2-3 was isolated from soil and identified as Pseudomonas putida H-2-3 by 16S rDNA sequences. The treatment of P. putida H-2-3 significantly increased the length, fresh and dry weight of shoot and chlorophyll content in gibberellins (GAs) deficient mutant Waito-c rice seedlings over the control, it might be the presence of GA<SUB>1</SUB>, GA<SUB>4,</SUB> GA<SUB>9</SUB> and GA<SUB>20.</SUB> The soybean plant growth was retarded in salt (120 mM sodium chloride) and drought (15% polyethylene glycol) stress conditions at 10 days treatments, while P. putida H-2-3 effectively enhanced the shoot length and fresh weight of plants suffered at salt and drought stress. The chlorophyll content was lower in abiotic stress conditions and bacterial inoculant P. putida H-2-3 mitigated the stress effects by an evidence of higher quantity of chlorophyll content in plants exposed to salt and drought. The stress hormonal analysis revealed that individual treatment of P. putida H-2-3, salt and drought significantly enhanced the abscisic acid and salicylic acid content than their control. P. putida H-2-3 applied to salt and drought stressed plants showed a lower level of abscisic acid and salicylic acid and a higher level of jasmonic acid content. Under stress condition induced by salt and drought in plants expressed higher level of total polyphenol, superoxide dismutase and radical scavenging activity and no significant changes in flavonoids. The bio-inoculant, P. putida H-2-3 modulated those antioxidants by declining superoxide dismutase, flavonoids and radical scavenging activity. P. putida H-2-3 induced tolerance against abiotic stress was confirmed by a reduction of Na content in abiotic stressed plants. The results suggest that P. putida H-2-3 application reprograms the chlorophyll, stress hormones and antioxidants expression in abiotic stress affected soybean plant and improves their growth under stress environment.

        Use of Chemical Treatments to Reduce Tannins and Trypsin Inhibitor Contents in Salseed (Shorea robusta) Meal

        This study investigated the effect of chemical treatments on tannins (condensed and hydrolysable) and on the trypsin inhibitor (TI) activity in salseed meal. Triplicate samples of ground salseed meal (1 kg) were mixed with 820 ml of either distilled water (pH 5.3), 0.67 M acetic acid (pH 2.4), 0.67 M sodium bicarbonate (pH 8.2) or 2% polyvinyl-pyrrolidone (PVP) solution. The material was placed in airtight plastic containers and incubated at $37^{\circ}C$ for 0, 3, 6, 12, 24, 48 and 72 h. Samples of untreated salseed meal which had not been subjected to soaking or incubation were run through the analysis to serve as control. Addition of water, acetic acid, sodium bicarbonate and PVP solutions to salseed meal and subsequent anaerobic incubation at $37^{\circ}C$ significantly reduced chemically detectable tannins. At each incubation time, alkali solution was more effective than its counterparts. The effect of acidic solution on hydrolysable tannin was least among the treatments. All the treatments reduced TI activity of salseed meal. The reduction in TI activity by these treatments was similar and ranged between 80-84%. Treatment time effected a decrease in the contents of antinutritional substances. However, the effect of the treatment with the reagents, even for zero incubation time, was quite pronounced. It may be concluded from the present results that the treatment of salseed meal with sodium bicarbonate (0.67 M) is more effective in reducing hydrolysable and condensed tannin contents than PVP, water and acid solutions. Treatment with sodium bicarbonate solution is more economical and easier to handle than acid and PVP treatments. Incubation of the treated material for 12 h is reasonably effective, economical and safe from any mould growth.

        Development of Bovine Specific Leptin Radioimmunoassay and Relationship of Plasma Leptin with Vitamin A and Age of Wagyu

        Leptin is produced by adipocytes and its role in the regulation of lipid metabolism, feed intake, productive and reproductive performance of domestic animal species has been greatly stressed and extensively investigated in recent years. This study was conducted to develop a radioimmunoassay (RIA) for the estimation of plasma bovine leptin and to determine plasma leptin concentration in fattening Japanese Black cattle (Wagyu) and its crossbreds at commercial farms. Relationships of plasma leptin with plasma vitamin A and age of crossbred cattle were also determined. Recombinant bovine leptin (rbleptin) was produced by the E. coli overexpressed leptin as a GST (glutathione S-transferase)-fusion protein. Then antiserum against bovine leptin was obtained by its immunization in rabbits. Using this antiserum, a bovine specific RIA was developed and plasma leptin level was determined in 120 crossbred fattening cattle (WagyuHolstein, 50:50) at commercial farms. The plasma leptin level increased with the age of cattle and its level was greater in the crossbred heifers than in the steers. Plasma vitamin A level was negatively correlated with plasma leptin level in crossbred heifers and steers. This relationship was stronger in heifers than in steers. Plasma leptin was gradually increased with advancing age in fattening Wagyu cattle. In conclusion, development of a bovine specific RIA to estimate plasma leptin will contribute to better understanding of the role of leptin in cattle.

        Enhanced photo-Fenton assisted photocatalytic degradation of atenolol using a novel rGO embedded double Z-scheme nano-heterojunction: Mechanism, kinetics and toxicity studies

        In this work, rGO-based ternary dual Z-scheme heterojunction (rGO/CuFe2O4/CdS/Bi2S3 QDs) was formulatedfor the effective photo-Fenton assisted photocatalytic degradation of a b-blocker, atenolol. The catalystwas synthesised via a simple co-precipitation/hydrothermal method and was characterized usingXRD, XPS, FT-IR, SEM, TEM, PL, EIS, ESR, Raman and DRS. It exhibited about 76.5% degradation of atenololin 360 min under visible light irradiation with a rate constant of 0.004 min1 wherein the mechanism ofenhanced activity was attributed to the formation of OH, h+ and photo-Fenton reaction; a possible mechanismwas drafted. The intermediates were determined using GC–MS analysis delineating a plausibledegradation pathway, the mineralization of atenolol being confirmed by TOC analysis. The catalyst wasreusable and highly stable up to six cycles of degradation, as affirmed via XRD and XPS analysis. Further, the toxicity of the catalyst has been studied using the root cells of Allium cepa, and the degradedproduct and intermediates were assessed deploying ECOSAR software. Thus, the proposed work haspotential which can be implemented in future for the large-scale purification of wastewater.

        Host plant growth promotion and cadmium detoxification in Solanum nigrum, mediated by endophytic fungi

        <P>Current investigation conducted to evaluate the associated fungal endophyte interactions of a Cd hyper accumulator Solanum nigrum Korean ecotype under varying concentrations of Cd. Two indole-3-acetic acid (IAA) producing fungal strains, RSF-4L and RSF-6L, isolated from the leaves of S. nigrum, were initially screened for Cd tolerance and accumulation potential. In terms of dry biomass production, the strain RSF-6L showed higher tolerance and accumulation capacity for Cd toxicity in comparison to RSF-4L. Therefore, RSF-6L was applied in vivo to S. nigrum and grown for six weeks under Cd concentrations of 0, 10, and 30 mg Kg(-1) of dry sand. The effect of fungal inoculation assessed by plant physiological responses, endogenous biochemical regulations, and Cd profile in different tissues. Significant increase were observed in plant growth attributes such as shoot length, root length, dry biomass, leaf area, and chlorophyll contents in inoculated RSF-6L plants in comparison to non-inoculated plants with or without Cd contamination. RSF-6L inoculation decreased uptake of Cd in roots and above ground parts, as evidenced by a low bio-concentration factor (BCF) and improved tolerance index (TI). However, Cd concentration in the leaves remained the same for inoculated and non-inoculated plants under Cd spiking. Fungal inoculation protected the host plants, as evidenced by low peroxidase (POD) and polyphenol peroxidase (PPO) activities and high catalase (CAT) activity. Application of appropriate fungal inoculation that can improve tolerance mechanisms of hyper-accumulators and reduce Cd uptake can be recommended for phyto-stabilisation/immobilisation of heavy metals in crop fields.       • Novel Ag@TiO<sub>2</sub> nanocomposite synthesized by electrochemically active biofilm for nonenzymatic hydrogen peroxide sensor

        A novel nonenzymatic sensor for H<SUB>2</SUB>O<SUB>2</SUB> was developed based on an Ag@TiO<SUB>2</SUB> nanocomposite synthesized using a simple and cost effective approach with an electrochemically active biofilm. The optical, structural, morphological and electrochemical properties of the as-prepared Ag@TiO<SUB>2</SUB> nanocomposite were examined by UV-vis spectroscopy, X-ray diffraction, transmission electron microscopy and cyclic voltammetry (CV). The Ag@TiO<SUB>2</SUB> nanocomposite was fabricated on a glassy carbon electrode (GCE) and their electrochemical performance was analyzed by CV, differential pulse voltammetry and electrochemical impedance spectroscopy. The Ag@TiO<SUB>2</SUB> nanocomposite modified GCE (Ag@TiO<SUB>2</SUB>/GCE) displayed excellent performance towards H<SUB>2</SUB>O<SUB>2</SUB> sensing at -0.73V in the linear response range from 0.83μM to 43.3μM, within a detection limit and sensitivity of 0.83μM and ~65.2328+/-0.01μAμM<SUP>-1</SUP>cm<SUP>-2</SUP>, respectively. In addition, Ag@TiO<SUB>2</SUB>/GCE exhibited good operational reproducibility and long term stability.

      • Gibberellin-producing Serratia nematodiphila PEJ1011 ameliorates low temperature stress in Capsicum annuum L.

        We studied the effects of plant growth-promoting rhizobacteria (PGPR) on the physio-hormonal attributes of pepper (Capsicum annuum L.) plants grown under low-temperature stress. After initial screening for growth promoting effect on gibberellin (GA) mutant Waito-C rice seeds, the PGPRs were analysed for gibberellins (GA) production through advanced chromatographic and spectroscopic techniques. Among 17 bacterial isolates, a novel isolate PEJ1011 produced bioactive GA<SUB>4</SUB> (8.65 ng ml<SUP>-1</SUP>) and physiologically inactive GA<SUB>20</SUB> (6.21 ng ml<SUP>-1</SUP>) and GA<SUB>9</SUB> (1.64 ng ml<SUP>-1</SUP>). The isolate PEJ1011 was identified as Serratia nematodiphila PEJ1011 using molecular techniques. To further assess it growth promoting effects, S. nematodiphila PEJ1011 was inoculated to pepper plant, where it significantly improved the growth attributes of pepper plants, while mitigated the deleterious effects of low temperature on pepper exposed to low temperature stress of 5 <SUP>o</SUP>C. It was observed that the inoculated plants grown under normal and low temperature stress contained higher endogenous GA<SUB>4</SUB> contents. To modulate cold stress, the beneficial association of PGPR up-regulated the endogenous ABA levels in pepper plants, while reduced the endogenous jasmonic acid and salicylic acid contents. This up and down regulation of stress hormones contribute to the immediate adaptation of plants exposed to low temperature stress. Current study showed the significance of S. nematodiphila PEJ1011 association to crops grown under adverse climatic conditions, and also reports the GA producing capacity of genus Serratia for the first time.

      • Inhibition of sulfide mineral oxidation by surface coating agents: Batch and field studies

        The potential of several surface coating agents to inhibit the oxidation of metal sulfide minerals from Young-Dong coal mine and the Il-Gwang gold mine was examined by conducting laboratory scale batch experiments and field tests. Powdered pyrite as a standard sulfide mineral and rock samples from two mine outcrops were mixed with six coating agents (KH<SUB>2</SUB>PO<SUB>4</SUB>, MgO and KMnO<SUB>4</SUB> as chemical agents, and apatite, cement and manganite as mineral agents) and incubated with oxidizing agents (H<SUB>2</SUB>O<SUB>2</SUB> or NaClO). For the observed time period (8 days), Young-Dong coal mine samples exhibited the least sulfate (SO<SUB>4</SUB><SUP>2-</SUP>) production in the presence of KMnO<SUB>4</SUB> (16%) or cement (4%) while, for Il-Gwang mine samples, the least SO<SUB>4</SUB><SUP>2-</SUP> production was observed in presence of KH<SUB>2</SUB>PO<SUB>4</SUB> (8%) or cement (2%) compared to control. Field-scale pilot tests at the Il-Gwang site also showed that addition of KH<SUB>2</SUB>PO<SUB>4</SUB> decreased SO<SUB>4</SUB><SUP>2-</SUP> production from 200 to 13mgL<SUP>-1</SUP> and it also reduced Cu and Mn from 8 and 3mgL<SUP>-1</SUP>, respectively to <0.05mgL<SUP>-1</SUP> (below ICP-OES detection limits). The experimental results suggested that the use of surface coating agents is a promising alternative for sulfide oxidation inhibition at acid mine drainage sites.
