        Identification and Functional Analysis of RelA/SpoT Homolog (RSH) Genes in Deinococcus radiodurans<sup>s</sup>

        To identify the global effects of (p)ppGpp in the gram-positive bacterium Deinococcus radiodurans, which exhibits remarkable resistance to radiation and other stresses, RelA/SpoT homolog (RSHs) mutants were constructed by direct deletion mutagenesis. The results showed that RelA has both synthesis and hydrolysis domains of (p)ppGpp, whereas RelQ only synthesizes (p)ppGpp in D. radiodurans. The growth assay for mutants and complementation analysis revealed that deletion of relA and relQ sensitized the cells to H<sub>2</sub>O<sub>2</sub>, heat shock, and amino acid limitation. Comparative proteomic analysis revealed that the bifunctional RelA is involved in DNA repair, molecular chaperone functions, transcription, the tricarboxylic acid cycle, and metabolism, suggesting that relA maintains the cellular (p)ppGpp levels and plays a crucial role in oxidative resistance in D. radiodurans. The D. radiodurans relA and relQ genes are responsible for (p)ppGpp synthesis/hydrolysis and (p)ppGpp hydrolysis, respectively. (p)ppGpp integrates a general stress response with a targeted re-programming of gene regulation to allow bacteria to respond appropriately towards heat shock, oxidative stress, and starvation. This is the first identification of RelA and RelQ involvement in response to oxidative, heat shock, and starvation stresses in D. radiodurans, which further elucidates the remarkable resistance of this bacterium to stresses.

        Based on STC microprocessor as a center of the transmitting and receiving parts, apply the interrupt systems, timers, counters integrated. Using Code-Division infrared coding, infrared receiver distinguishes the light pulse , judges and makes the corresponding control actions to complete the all infrared remote control transmitter and receiver process. Combination of the software programs, after commissioning the system can achieve multiple dimming function, such as LED lamp PWM dimming, alternating current filament lamp dimming, multiple channel switches. The system realized the function that weak power can control not only flea power but also strong power. This system has the feature of simple hardware circuit, sophisticated software capability, high cost performance. At the same time the study of intelligent lighting control system contributes to the improvement and development of the intelligent home system.

        Fucoidan from Acaudina molpadioides Protects Pancreatic Islet against Cell Apoptosis via Inhibition of Inflammation in Type 2 Diabetic Mice

        Inflammation induces pancreatic islet cell apoptosis. Effects of fucoidan from Acaudina molpadioides (Am-FUC) on inhibition of pancreatic islet cell apoptosis and inflammation in type 2 diabetic mice were investigated. Am-FUC repaired pancreatic islet cells, decreased serum C-reactive protein (CRP), macrophage inflammatory protein 1 (MIP-1), interleukin 1β (IL-1β), IL-6, and tumor necrosis factor-α (TNF-α) levels, and increased the IL-10 level. Am-FUC also reduced TNF-α, CRP, MIP-1, IL-1β, and IL-6 mRNA expressions, and increased IL-10 mRNA expression in epididymal adipose tissues. Am-FUC reduced Bid, Bax, cytochrome c, caspase 9, and caspase 3 mRNA expressions, and increased Bcl-2 and Bcl-xL mRNA expressions. Am-FUC down-regulated t-Bid, Bax, cytochrome c, and caspase 9 activities, cleaved caspase 3 proteins, and up-regulated Bcl-2 and Bcl-xL proteins. Thus, an Am-FUCblocked mitochondrial pathway was the suppression mechanism in pancreatic islet cell apoptosis via regulation of inflammatory cytokines providing dietary intervention in type 2 diabetes and inflammation-induced pancreatic islet apoptosis.

        Influence of the insemination method on the outcomes of elective blastocyst culture

        Objective: The aim of this study was to explore the effects of the insemination method on the outcomes of elective blastocyst culture. Methods: We retrospectively analyzed the outcomes of elective blastocyst culture performed between January 2011 and December 2014. Results: There were 2,003 cycles of conventional in vitro fertilization (IVF) and 336 cycles of intracytoplasmic sperm injection (ICSI), including 25,652 and 4,164 embryos that underwent sequential blastocyst culture, respectively. No significant differences were found in the female patients' age, basal follicle-stimulating hormone level, basal luteinizing hormone level, body mass index, number of oocytes, maturity rate, fertilization rate, or good-quality embryo rate. However, the blastocyst formation rate and embryo utilization rate were significantly higher in the conventional IVF group than in the ICSI group (54.70% vs. 50.94% and 51.09% vs. 47.65%, respectively, p<0.05). The implantation/pregnancy rate (IVF, 50.93%; ICSI, 55.10%), miscarriage rate (IVF, 12.57%; ICSI, 16.29%), and live birth rate (IVF, 42.12%; ICSI, 44.08%) were similar (p>0.05). No cycles were canceled due to the formation of no usable blastocysts. Conclusion: Although the fertilization method had no effect on clinical outcomes, the blastocyst formation rate and embryo utilization rate in the ICSI group were significantly lower than those observed in the conventional IVF group. Therefore, more care should be taken when choosing to perform blastocyst culture in ICSI patients.

        Weighted Average Consensus of Second-order Multi-agent Systems with Various Intelligence Levels via Sampled-data Control

        In this paper, weighted average consensus of second-order multi-agent systems (MASs) via sampleddata control is investigated. Specially, since the intelligence degree of each agent may be different in practical applications, the concept of intelligence level is introduced to characterize the intelligence degrees of agents in this paper. A novel distributed continuous-time control protocol with various intelligence levels is put forward based on sampled data in order to reduce the updating rates and computational burden of the controller. A necessary and sufficient condition is derived to achieve weighted average consensus under the proposed protocol. Moreover, according to the acquired result, the authors also give the choosing approaches of the feedback gains and sampling period. Finally, several numerical simulations are provided to illustrate the reliability of the theoretical results.

        Robust Charging and Discharging Coordination of Electric Vehicles with State Dynamics Uncertainties

        This article studies a class of electric vehicle (EV) charging and discharging coordination problems considering specific uncertainties involved in the dynamics of EV’s state of charge (SOC). A robust strategy is explored for EVs such that it is always feasible with respect to all the uncertainties. More specifically, based on the so-called robust positive invariant (RPI) set introduced by Bertsekas et al. to describe the effect of the disturbances, a deterministic optimization problem is formulated by removing the uncertainty part in the state dynamics. Then, based upon the implemented optimal solution to the formulated deterministic problem, design a decentralized coordination strategy which is shown to be feasible for all the uncertainties and guarantee the robust optimality. Furthermore, in order to reduce the conservatism and improve the robust optimality, a sequence of robust disturbance sets, instead of the RPI set, is applied for the underlying coordination problem with a fixed terminal time. Numerical simulations are presented to verify the developed results in this work.

        Novel Mutations in CYP51B from Penicillium digitatum Involved in Prochloraz Resistance

        Green mold caused by Penicillium digitatum is one of themost serious postharvest diseases of citrus fruit, and it isubiquitous in all citrus growing regions in the world. Sterol14α-demethylase (CYP51) is one of the key enzymes of sterolbiosynthesis in the biological kingdom and a prime targetof antifungal drugs. Mutations in CYP51s have been foundto be correlated with resistance to azole fungicides in manyfungal species. To investigate the mechanism of resistance toprochloraz (PRC) in P. digitatum, the PRC sensitivity wasdetermined in vitro in this study to assess the sensitivity of78 P. digitatum isolates collected in Hubei province. Theresults showed that 25 isolates were prochloraz-resistant(PRC-R), including six high-resistant (HR) strains, twelvemedium-resistant (MR) and seven low-resistant (LR) strains. A sequence analysis showed no consistent point mutationsof PdCYP51A in the PRC-R strains, but four substitutionsof CYP51B were found, Q309H in LR strains, Y136H andQ309H in HR strains, and G459S and F506I in MR strains,which corresponded to the four sensitivity levels. Based onthe sequence alignment analysis and homology modelingfollowed by the molecular docking of the PdCYP51B protein,the potential correlation between the mutations and PRC resistance is proposed.

        Esophagogastric Junction Contractility Integral Reflect the Anti-reflux Barrier Dysfunction in Patients with Gastroesophageal Reflux Disease

        Background/Aims Anti-reflux barrier dysfunction is one of the primary mechanisms in gastroesophageal reflux disease (GERD) pathogenesis. The esophagogastric junction contractile integral (EGJ-CI) is a new metric adopted to evaluate the EGJ contractility, which implies the antireflux barrier function. The aim of the current study was to validate this new metric in patients with GERD and its correlation with the esophageal acid exposure, as well as the efficacy of proton pump inhibitor treatment. Methods Ninety-eight patients with GERD and 21 healthy controls were included in the study. Upper endoscopy, high-resolution manometry (HRM) and 24-hour multichannel intraluminal impedance-pH monitoring were performed in all patients. Three respiration cycles were chosen at the initial HRM resting frame and the value computed with distal contractile integral tool was then divided by the duration of the cycles to yield EGJ-CI. All the patients were treated with esomeprazole 20 mg twice-daily for 8 weeks. Results EGJ-CI was lower in the patients with GERD than that of the controls (P < 0.05). For patients with GERD, EGJ-CI was lower in those with hiatal hernia (P < 0.05). The new metric correlated with esophageal acid exposure in the supine position (P < 0.05), and it also negatively correlated to the total reflux episodes (P < 0.05). There was no significant difference on EGJ-CI between patients with and without response to the esomeprazole treatment (P = 0.627). Conclusions EGJ-CI reflected the dysfunction of the anti-reflux barrier in patients with GERD, but it had little impact on the esomeprazole response. (J Neurogastroenterol Motil 2017;23:27-33)

        Fatigue life prediction model of Ti-6Al-4V alloy forgings based on forging process parameters

        Forging processing parameters have an important impact on the fatigue resistance of metal in the process of plastic forming, which directly threatens the safety and reliability of the metal’s service. Taking Ti-6Al-4V alloy as the research object, this study investigates the effect of processing parameters, including forging temperature and deformation degree on the Young's modulus, ultimate tensile strength, and reduction of fracture area on the basis of the mechanical property testing of Ti-6Al-4V alloy forgings and obtains the regression equations at the same time. Combined with the existing stress fatigue life prediction formula, the fatigue life prediction based on forging process parameters is realized. Results show that forging temperature and deformation degree have a significant effect on fatigue life. The forging processing parameters of Ti-6Al-4V alloy with optimum fatigue life are as follows: 985 °C-990 °C forging temperature and 46 %-50 % deformation degree.
