A Yellow Pigmented Soil Bacterium Producing a Polysaccharide of High Viscosity

A strictly aerobic bacterium forming yeller pigment and a highly viscous polysaccharide was isolated. The bacterium was identified as Pseudomonas mendocia. The polysaccharide was presumed to be ${\beta}-glucan$ with o-acetyl group in its structure and the constituent sugar components were glucose and rhamnose in the molar ratio of 2.1: 1.0. The intrinsic viscosity was 64.73dl/g. The apparent viscosity of 1% aqueous solution was 428 mPa.s. at $42\;sec^{-1}$ and the yield stress of the solution was 8.89Pa. The polysaccharide did not have thermal stability but show pH and salt stability. 산업용 다당류의 개발을 목적으로 고점도의 다당류생산 미생물을 토양으로부터 분리하였다. 본 세균은 동정한 결과 Pseudomonas mendocina로 생각되었다. 본 세균이 생산하는 다당류는 o-acetyl기를 함유한 ${\beta}-glucan$으로 추정되며 구성당으로서는 포도당과 람노오스가 몰비로 2.2 : 1.0 구성으로 되어 있고 수용액은 의가소성 유체이다. 1% 용액은 428mPa.s의 점도를 나타내며 항복치는 8.89Pa이다. 이 용액은 열안정성은 없으며 pH 안정성과 염안정성은 있었다. 유동활성화에너지는 6.57 Kcal 이다.

재래식 간장의 향미를 개선시키기 위하여 Zygosacharomyces rouxii H-62균주를 이용하여 염농도별, 염수 용량별, 발효 온도별 발효시험을 실행하였다. 효모를 발효시 첨가함으로서 관능적 품질을 향상시킬 수 있었으며 발효는 메주를 12등분하여 메주 1개(1.2㎏)당 15.5%의 소금물을 5L 넣어 30℃에서 60일간 발효하는 것이 적합하다고 판단되었다. 이와 같은 공정으로 간장을 제조하면 성분으로서는 총 질소 함량이 0.80%, 순 추출물 6.0%이상인 기존의 재래식 간장보다 저염화된 우수한 품질의 간장을 제조할 수 있었다. The traditional kanjang has been prepared by mixing meju and 18% saline solution, and fermenting for 60 days. The traditional kanjang is very salty and inferior in flavor and taste comparing with commercial fermented soy sauce. To improve the quality of traditional kanjang, Zygosaccharomyces rouxii H-62, a flavor-related mutant, was inoculated during fermentation. It was found that the addition of Z rouxii helped to improve the organoleptic quality of traditional kanjang. The optimal condition for preparing traditional kanjang was to use 5L of 15.5% saline solution per meju. Meju must be cut into 12 pieces to get a proper total nitrogen and pure extract content. The optimal fermentation temperature was 30℃. The prepared kanjang contained over 0.8% total nitrogen and 6.0% pure extract after 60 days of fermentation.
