Background : The aim of this study is to determine prognostic factors of breast cancer in Korean patients who underwent curative mastectomy. Methods : Medical records of 181 patients who underwent curative mastectomy were reviewed. Relapse-free survival and overall survival were documented for each patient. Factors influencing survival were analyzed using Cox proportional hazard model. Results : Overall 5-year survival rate was 82.0%, and 8-year survival rate was 74.7%. Multivariate analysis indicated that multiple axillary lymph node involvement (≥4), postmenopausal status, and negative estrogen receptor were independent adverse prognostic factors. The adjusted relative risks of multiple axillary lymph node involvement (≥4), postmenopausal status, and negative estrogen receptor were estimated 2.60 (95% Confidence Interval (CI): 1.28-5.30), 2.64 (95% CI: 1.46-4.79), and 2.27 (95% CI: 1.19-4.35), respectively. Conclusion : Multiple axillary lymph node involvement (≥4), postmenopausal status, and negativstrogen receptor were independent adverse prognostic factors in Korean breast cancer patients after curative mastectomy.(Korean J Med 58:446-452, 2000)$quot;

<P>Accumulation of expanded polyglutamine proteins is considered to be a major pathogenic biomarker of Huntington disease. We isolated SCAMP5 as a novel regulator of cellular accumulation of expanded polyglutamine track protein using cell-based aggregation assays. Ectopic expression of SCAMP5 augments the formation of ubiquitin-positive and detergent-resistant aggregates of mutant huntingtin (mtHTT). Expression of SCAMP5 is markedly increased in the striatum of Huntington disease patients and is induced in cultured striatal neurons by endoplasmic reticulum (ER) stress or by mtHTT. The increase of SCAMP5 impairs endocytosis, which in turn enhances mtHTT aggregation. On the contrary, down-regulation of SCAMP5 alleviates ER stress-induced mtHTT aggregation and endocytosis inhibition. Moreover, stereotactic injection into the striatum and intraperitoneal injection of tunicamycin significantly increase mtHTT aggregation in the striatum of R6/2 mice and in the cortex of N171-82Q mice, respectively. Taken together, these results suggest that exposure to ER stress increases SCAMP5 in the striatum, which positively regulates mtHTT aggregation via the endocytosis pathway.</P>

The activity-guided fractionation upon the MeOH extract of the aerial parts of Sorbaria sorbifolia var. stellipila led to the isolation of two cucurbitacin-compounds, cucurbitacin D and cucurbitacin F, as active principles. Two compounds were shown to exhibit significant cytotoxicity against cultured human tumor cell lines, A-549, SK-OV-3, SK-MEL-2, XF-498, and HCT 15.

<P>Paclitaxel (PTX) is commonly used to treat urothelial carcinoma (UC) after platinum-based chemotherapy has failed. However, single-agent taxane therapy is not sufficient to inhibit tumor progression and drug resistance in advanced UC. Epithelial-to-mesenchymal transition (EMT) induced by fibroblast growth factor receptor (<I>FGFR</I>)1 signaling has been proposed as a mechanism of PTX resistance, but it is unclear whether this can be overcome by <I>FGFR1</I> inhibition. The present study investigated whether <I>FGFR1</I> overexpression contributes to PTX resistance and whether <I>FGFR</I> inhibition can enhance PTX efficacy in UC. The effects of PTX combined with the <I>FGFR</I> inhibitor BGJ398 were evaluated in UC cell lines by flow cytometry; Western blot analysis; cell viability, migration, and colony forming assays; and RNA interference. PTX+BGJ398 induced cell cycle arrest and apoptosis in UC cells with mesenchymal characteristics was accompanied by downregulation of <I>cyclin D1</I> protein and upregulation of gamma-histone 2A family member X and cleaved poly(ADP-ribose) polymerase. Additionally, PTX+BGJ398 synergistically suppressed UC cell migration and colony formation via regulation of EMT-associated factors, while <I>FGFR1</I> knockdown enhanced the antitumor effect of PTX. These findings provide a basis for development of effective strategies for overcoming PTX resistance in UC through inhibition of <I>FGFR1</I> signaling.</P>

Annual variation of water quality in Pal`tang Reservoir in Korea is described. Water samples were collected monthly from surface layer at the one station of Pal`tang reservoir, Korea from March 1988 to May 1992. Dissolved oxygen, turbidity, conductivity, pH and chlorophyll-α concentration in water column was high compared with that of 1970`s. Recently, the frequency and intensity of phytoplankton blooms(over 15㎍ chlorophyll-α1^-1) appear to be increasing. Total nitogen concentration was high compared with that of 1970`s. But, it decreased gradually from 4.76mg/l to 0.51mg/l on January 1991 as it goes. Total phosphorus concentration in water column has increased more and more from 0.004mg/l to 0.074mg/l since 1988. Therefore, N/P ratio by atoms reduced from 500 to 10∼20 because of the increasing of phosphorus and decreasing of nitrogen. This result suggests that phosphorus limited phytoplankton growth in Pal`tang Reservoir. However, if N/P ratio reduced continuously like this pattern, dominat phytoplankton species might change due to nitrogen limitation from diatom assemblages to blue green algae.

Purpose: The guidelines for pathological evaluation of early gastric cancer (EGC) recommend wider section intervals for surgical specimens (5-7 mm) than those for endoscopically resected specimens (2-3 mm). Studies in surgically resected EGC specimens showed not negligible lymph node metastasis risks in EGCs meeting the expanded criteria for endoscopic submucosal dissection (ESD). Materials and Methods: This retrospective study included 401 EGC lesions with an endoscopic size of ≤ 30 mm detected in 386 patients. Pathological specimens obtained by ESD or surgery were cut into 2-mm section intervals for reference. Submucosal or lymphovascular invasion (LVI) was evaluated arbitrarily in 4- or 6-mm section intervals. McNemar's tests compared the differences between submucosal and LVI. Results: Submucosal invasion was detected in 29.2% (117/401) and LVI in 9.5% (38/401) at 2-mm interval. The submucosal invasion detection rates in 4-mm intervals decreased to 88.0% or 90.6% (both P<0.001), while the LVI detection rates decreased to 86.8% or 57.9% (P=0.025 and P<0.001, respectively). In 6-mm intervals, the submucosal and LVI detection rates decreased further to 72.7-80.3% (P<0.001 for all three sets) and 55.3-63.2% (P<0.001 for all three sets), respectively. Among 150 out-of-indication cases at 2-mm interval, 4-10 (2.7%-6.7%) at 4-mm intervals, and 10-17 (6.7%-11.3%) at 6-mm intervals were misclassified as lesions meeting the curative resection criteria due to the underestimation of submucosal or LVI. Conclusions: After ESD, the 2-mm wide section interval was suitable for the pathological evaluation of focal submucosal or LVI. Thus, if an EGC lesion meets the expanded criteria for the ESD specimen pathological evaluation, it could be safely followed up.

This paper presents FPGA implementation for programmable digital FIR/IIR filter. Sine wave generation and detecting frequency of the sine wave signals have been proposed. Simulation results show that the system has the ability to provide a better frequency response for various cut-off frequencies. The design has been implemented in Altera, cyclone II family EP2C70F89618 device using Quartus software.
The Korea Cohort Consortium: The Future of Pooling Cohort Studies

Objectives: We introduced the cohort studies included in the Korean Cohort Consortium (KCC), focusing on large-scale cohort studies established in Korea with a prolonged follow-up period. Moreover, we also provided projections of the follow-up and estimates of the sample size that would be necessary for big-data analyses based on pooling established cohort studies, including population-based genomic studies. Methods: We mainly focused on the characteristics of individual cohort studies from the KCC. We developed “PROFAN”, a Shiny application for projecting the follow-up period to achieve a certain number of cases when pooling established cohort studies. As examples, we projected the follow-up periods for 5000 cases of gastric cancer, 2500 cases of prostate and breast cancer, and 500 cases of non-Hodgkin lymphoma. The sample sizes for sequencing-based analyses based on a 1:1 case-control study were also calculated. Results: The KCC consisted of 8 individual cohort studies, of which 3 were community-based and 5 were health screening-based cohorts. The population-based cohort studies were mainly organized by Korean government agencies and research institutes. The projected follow-up period was at least 10 years to achieve 5000 cases based on a cohort of 0.5 million participants. The mean of the minimum to maximum sample sizes for performing sequencing analyses was 5917-72 102. Conclusions: We propose an approach to establish a large-scale consortium based on the standardization and harmonization of existing cohort studies to obtain adequate statistical power with a sufficient sample size to analyze high-risk groups or rare cancer subtypes.
