In this study, a strategy was proposed for making galacto-oligossaccharides (GOS), a high valueadded product, from a byproduct of the dairy industry,cheese whey, using a commercial β-galactosidase from Kluyveromyces lactis (Lactozym® 3000L). The effects of the substrate concentration, temperature, and enzyme dosage were statistically studied and their optimum combinations were determined using response surface methodology. The increase in lactose concentration, temperature, and enzyme concentration favored a transgalactosylation reaction. The maximum values for GOS concentration (119.8 mg/mL)and yield (29.9%) in a 4 h process were obtained in the reaction system, composed of 400 mg/mL of lactose and 10 U/mL of enzyme at 40°C. Under these conditions, the lactose conversion was 68.7%. The maximum value for lactose conversion (87.8%) was observed at the same temperature and enzyme concentration, although the lactose level was 20%.

The yeast Phaffia rhodozyma is known for producing carotenogenic pigments, commonly used in aquaculture feed formulation as well as in cosmetic,pharmaceutical, and food industries. Despite the high production of carotenoids from microorganisms by biotechnology, their use has limitation due to the cell wall resistance, which constitutes a barrier to the bioavailability of carotenoids. Therefore, there is a need to improve carotenoids recovering technique from microorganisms for the application of food industries. This study aimed to compare mechanical, chemical, and enzymatic techniques of cell disruption for extracting carotenoids produced by P. rhodozyma NRRL Y-17268. Among the techniques studied, the highest specific concentration of carotenoids (190.35 μg/g) resulted from the combined techniques of frozen biomass maceration using diatomaceous earth and enzymatic lysis at pH of the reaction medium of 4.5 at 55oC, with initial activity of β-1,3 glucanase of 0.6 U/mL for 30 min.

Green reduction of graphene oxide (GO) using drained water from soaked mung beans (Phaseolus aureus L.) has been demonstrated. In comparison to the toxic and hazardous reducing chemicals, the drained water from soaked mung beans (P. aureus L.) is completely green reducing agent, the reduction process is very simple and cost effective. The removal of oxygen containing functional groups of GO has been confirmed by UV-vis, Fourier transform infrared and X-ray photoelectron spectroscopy analysis. Morphological characterization of rGO has been performed by atomic force and transmission electron microscopy analysis. Electrochemical performances of rGO have been evaluated by cyclic voltammetry (CV), charge-discharge and electrochemical impedance spectroscopy techniques. The specific capacitance (SC) of rGO has been found to be 137Fg<SUP>-1</SUP> at a current density of 1.3Ag<SUP>-1</SUP>. The retention in SC is more than 98% after 1000 charge-discharge cycles suggesting long-term electrochemical cyclic stability as supercapacitor electrode materials.

Introduction: The nuclear factor ${\kappa}B$ (NF-${\kappa}B$) is a super family of transcription factors which plays important roles in development and progression of cancer. The present investigation concerns NF-${\kappa}B$ /p65 activity in human breast cancers with overexpression of ER, PR, HER-2/neu, as well as the significance of p65 expression with regard to menopausal status, stage, grade, tumor size, nodal status, and NPI of invasive ductal carcinomas in Eastern India. Materials and Methods: In this hospital based study 57 breast cancer patients attending a Breast Clinic of a reputed institute of Eastern India were assessed for p65 protein expression in breast tumor tissue samples by Western blotting. ER, PR and HER-2/neu expression was determined by immunohistochemistry. Results: NF-${\kappa}B$/p65 was significantly associated with advanced stage, large tumor size (${\geq}5$ cm), high grade, negative ER, negative PR, and positive HER-2/neu. High NF-${\kappa}B$/p65 expression was more frequent in patients with a high NPI ($NPI{\geq}5.4$, 84.6%) compared with low NPI (<5.4, 44.4%) and this association was statistically significant (p = 0.002). Conclusion: NF-${\kappa}B$/p65 overexpression was associated with advanced stage, large tumor size, high grade, and high NPI which are poor prognostic factors linked to enhanced aggressiveness of the disease. NF-${\kappa}B$/p65 expression implies aggressive biological behavior of breast cancer and this study validates significant association of NF-${\kappa}B$ /p65 overexpression with negative estrogen and progesterone receptor status and overexpression of HER-2/neu oncoprotein. In our good clinical practice, patients with NF-${\kappa}B$ positive tumors need to be treated aggressively.

Felodipine, a calcium channel blocker has been widely used for the treatment of hypertension and cardiovascular diseases; but the frequent dosing is needed for its poor solubility and variable bioavailability. In present study an attempt has been made to overcome the problems through nanoparticulate delivery system using poly (D, L-lactic-co-glycolic acid) polymer keeping in the view to get better sustainability of the formulation. The nanoparticles were prepared by single emulsion solvent evaporation technique and the physico-chemical characterization of prepared nanoparticles confirmed the particles were nanosize range with smooth and spherical morphology. Further, the compatibility of drug-polymer combination was analyzed by FTIR and DSC study. The in vitro drug release study of PLGA nanoparticles showed longer duration of drug release with reduced burst release compared with pure felodipine. The in vitro drug release data were fitted with various mathematical models to establish the drug release mechanism from the nanoparticles and found to follow mixed order kinetics. The in vivo toxicity study in albino mice showed no noticeable change in biochemical parameters and histopathology of organs. Hence, the developed felodipine nanoparticles were prepared, characterized and could possibly be advantageous for prolonged drug release and improving the antihypertensive effect.
The Origin of Minus-end Directionality and Mechanochemistry of Ncd Motors

<▼1><P>Adaptation of molecular structure to the ligand chemistry and interaction with the cytoskeletal filament are key to understanding the mechanochemistry of molecular motors. Despite the striking structural similarity with kinesin-1, which moves towards plus-end, Ncd motors exhibit minus-end directionality on microtubules (MTs). Here, by employing a structure-based model of protein folding, we show that a simple repositioning of the neck-helix makes the dynamics of Ncd non-processive and minus-end directed as opposed to kinesin-1. Our computational model shows that Ncd in solution can have both symmetric and asymmetric conformations with disparate ADP binding affinity, also revealing that there is a strong correlation between distortion of motor head and decrease in ADP binding affinity in the asymmetric state. The nucleotide (NT) free-ADP (φ-ADP) state bound to MTs favors the symmetric conformation whose coiled-coil stalk points to the plus-end. Upon ATP binding, an enhanced flexibility near the head-neck junction region, which we have identified as the important structural element for directional motility, leads to reorienting the coiled-coil stalk towards the minus-end by stabilizing the asymmetric conformation. The minus-end directionality of the Ncd motor is a remarkable example that demonstrates how motor proteins in the kinesin superfamily diversify their functions by simply rearranging the structural elements peripheral to the catalytic motor head domain.</P></▼1><▼2><P><B>Author Summary</B></P><P>Proteins belonging to the kinesin superfamily are responsible for vesicle or organelle transport, spindle morphogenesis, and chromosome sorting during cell division. Interestingly, while most proteins in kinesin superfamily that share the common catalytic motor head domain have plus-end directionality along microtubules, kinesin-14 (Ncd) exhibits minus-end directionality. Despite the several circumstantial evidences on the determining factors for the motor directionality in the last decade, a comprehensive understanding of the mechanism governing the Ncd minus-end directionality is still missing. Our studies provide a clear explanation for this minus-end directionality and the associated mechanochemical cycle. Here, we modeled an Ncd motor by employing structural details available in the literature to simulate its conformational dynamics. Simulations using our structure-based model of Ncd assert that the dynamics due to a simple rearrangement of structural elements, peripheral to the catalytic motor domain, is the key player in determining both the directionality and mechanochemistry unique to Ncd motors. Although Ncd has a different directionality, it uses a similar strategy to kinesin-1 of structural adaptation of the catalytic motor domain. Therefore using the same physical principle of protein folding and very similar structural elements, motors in the kinesin superfamily are able to achieve a variety of biological function.</P></▼2>

In recent years, several interesting families of generating functions for various classes of hypergeometric functions were investigated systematically. In the present paper, we introduce a new family of extended Wright type hypergeometric function and obtain several classes of generating relations for this extended Wright type hypergeometric function.
