Toll-like receptors (TLRs) recognize pathogen-associated molecular patterns (PAMPs) and regulate the activation of innate immunity. All TLR signaling pathways culminate in the activation of NF-κB, leading to the induction of inflammatory gene products such as cyclooxygenase-2 (COX-2). Triptolide (TP), a natural component of Tripterygium wilfordii Hook. F, has been used as folk remedies to treat many chronic diseases for many years. In the present report, we present biochemical evidence that TP inhibits the NF-κB activation induced by polyriboinosinic polyribocytidylic acid (Poly[I:C], TLR3 agonist) and lipopolysaccharide (LPS, TLR4 agonist). TP also inhibits COX-2 expression induced by Poly[I:C] and LPS. These results suggest that TP can modulate the immune responses regulated by TLR3 and TLR4 signaling pathways.

Toll-like receptors (TLRs) play an important role for host defense against invading pathogens. TLR4 has been identified as the receptor for lipopolysaccharide (LPS), which is a cell wall component of gram-negative bacteria. The activation of TLR4 signaling by LPS leads to the activation of NF-κB and the expression of pro-inflammatory gene products such as cytokines, inducible nitric oxide synthase (iNOS), and cyclooxygenase-2 (COX-2). To evaluate the therapeutic potential of (E)-1-(2-(2-nitrovinyl)phenyl)pyrrolidine (NVPP), previously synthesized in our laboratory, NF-κB activation and iNOS and COX-2 expression induced by LPS were examined. NVPP inhibited the activation of NF-κB induced by LPS. NVPP also suppressed the iNOS expression induced by LPS but it did not suppress COX-2 expression induced by LPS. These results suggest that NVPP has the specific mechanism for anti-inflammatory responses.
교원 정보화 원격교육 효과성에 영향을 미치는 요인 분석 연구

이 연구에서는 원격교육 효과성과 효과성에 영향을 미치는 요인 분석을 위해 SPSS(version 12.0) 통계 패키지 프로그램과 구조방정식모형(Structural Equation Model)을 통해 분석하였다. 이 연구를 수행하기 위해서 제주도를 제외한 전국 15개 시ㆍ도에서 2003년부터 2005년 2월까지 정보화 원격교육과정을 이수한 교원 983명의 설문조사 응답지를 분석에 사용하였다. 원격교육의 효과성은 하위영역별로 모두 보통수준 이상으로 나타났다. 하위영역 중 자기개발 기여도(3.58)가 가장 높은 수준으로 나타났고, 그 다음으로 정보기술활용능력 향상도(3.36), 현장적용도(3.18) 순으로 나타났다. 이 연구에서 매개변인으로 설정한 정보화 원격교육 학습참여도는 보통수준 이하(2.43)로 나타났다. 원격교육 효과성에 영향을 미치는 요인은 학습자 요인, 운영자 요인, 학습참여도 요인이 유의미한 직접적인 영향을 미쳤고, 원격교육 학습참여도에는 학습자 요인만 유의미한 직접적인 영향을 미쳤다. 학습자 요인은 학습참여도에 영향을 미침으로써 간접적으로도 원격교육 효과성에 영향을 미치는 것으로 나타났다. The purpose of this study was to examine the effectiveness of distance education and what affected its effectiveness by employing SPSS(Ver. 12.0) program and a structural equation model. The subjects in this study were 983 teachers who had took courses in distance information education in between 2003 and February of 2005 in 15 provinces and large cities across the nation, except Jeju-do. After a survey was conducted, their answer sheets were analyzed. The effectiveness of distance education turned out to be above the average in its every sub-area. Among the sub-areas, that made the best contribution to improving their self-development(3.58), which was followed by information literacy(3.36) and field application(3.18). Its contribution to boosting their participation in distance information education, which was selected as a parameter in the study, was below the average(2.43). The factors of learner, operator and learning participation were identified as what had a significantly firsthand impact on the effectiveness of distance education. The participation in distance education was under the significantly firsthand influence of the learner factor only. The learner factor exercised a secondhand influence on the effectiveness of distance education, as it affected learning participation.

N/A This study was conducted to develop site index equations by main species grown in Gyunggi and Chungcheong provinces using environmental factors obtained from a digital forest site map. For this, 28 environmental factors were regressed on site index by species. Four to five environmental factors by species were selected as independent variables in the best site index equations (coefficients of determination greater than 0.91). For these site index equations, three evaluation statistics, mean difference, standard deviation of difference, and standard error of difference, were applied to the data set. Site index equations by species relationships developed in this study effectively estimate forest productivity in the study area. However, the site index equation of Larix leptolepis showed a larger than expected bias between the estimated and the measured site index. The reason is not clear in this situation, but might be because of the small sample set. It will be necessary, therefore, to conduct more studies to determine the exact reason. It is also expected that the site index equations with a few environmental factors as independent variables could provide valuable information about species well suited to given site conditions. Site index equations for other species should be developed to establish a rational policy about the selection of best species for site conditions.

Fasiglifam (TAK-875) a G-protein coupled receptor 40 (GPR40) agonist, significantly improves hyperglycemia without hypoglycemia and weight gain, the major side effects of conventional anti-diabetics. Unfortunately, during multi-center Phase 3 clinical trials, unexpected liver toxicity resulted in premature termination of its development. Here, we investigated whether TAK-875 directly inflicts toxicity on hepatocytes and explored its underlying mechanism of toxicity. TAK-875 decreased viability of 2D and 3D cultures of HepG2, a human hepatocarcinoma cell line, in concentration-(>$50{\mu}M$) and time-dependent manners, both of which corresponded with ROS generation. An antioxidant, N-acetylcysteine, attenuated TAK-875-mediated hepatotoxicity, which confirmed the role of ROS generation. Of note, knockdown of GPR40 using siRNA abolished the hepatotoxicity of TAK-875 and attenuated ROS generation. In contrast, TAK-875 induced no cytotoxicity in fibroblasts up to $500{\mu}M$. Supporting the hepatotoxic potential of TAK-875, exposure to TAK-875 resulted in increased mortality of zebrafish larvae at$25{\mu}M$. Histopathological examination of zebrafish exposed to TAK-875 revealed severe hepatotoxicity as manifested by degenerated hypertrophic hepatocytes with cytoplasmic vacuolation and acentric nuclei, confirming that TAK-875 may induce direct hepatotoxicity and that ROS generation may be involved in a GPR40-dependent manner.

Generally, the digital circuit card can be tested by automatic test equipment using LASAR(Logic Automated Stimulus and Response). This paper proposes the ROM data extracting algorithm which can test the digital circuit card that consists usually ROMs. We are implemented of the proposed fault detecting program by LabWindow/CVI 8.5 and the digital automatic test instrument with NI-VXI(National Instrument -Versa Bus Modular Europe eXtentions for Instrumentation) card. We also make an interface circuit board connecting the digital test instrument and the digital circuit card. It shows the good performance of getting the data from ROMs.
