      • Association of MDR1 Gene Polymorphisms with Susceptibility to Hepatocellular Carcinoma in the Chinese Population

        Objective: The objective of this study was to evaluate the association of MDR1 gene polymorphisms with susceptibility to hepatocellular carcinoma (HCC). Methods: A total of 689 HCC patients and 680 cancer-free subjects were enrolled. Human MDR1 gene polymorphisms were investigated by created restriction site-polymerase chain reaction (CRS-PCR) and DNA sequencing methods. Multiple logistic regression models were applied to estimate the association between MDR1 gene polymorphisms and susceptibility to HCC. Results: We detected a novel c.4125A>C polymorphism and our findings suggested that this variant was significantly associated with susceptibility to HCC. A significantly increased susceptibility to HCC was noted in the homozygote comparison (CC versus AA: OR=1.621, 95% CI 1.143-2.300, ${\chi}^2$=7.4095, P=0.0065), recessive model (CC versus AC+AA: OR=1.625, 95% CI 1.167-2.264, ${\chi}^2$=8.3544, P=0.0039) and allele contrast (C versus A: OR=1.185, 95% CI 1.011-1.389, ${\chi}^2$=4.4046, P=0.0358). However, no significant increase was observed in the heterozygote comparison (AC versus AA: OR=0.995, 95% CI 0.794-1.248, ${\chi}^2$=0.0017, P=0.9672) and dominant model (CC+AC versus AA: OR=1.106, 95% CI 0.894-1.369, ${\chi}^2$=0.8560, P=0.3549). Conclusions: These findings suggest that the c.4125A>C polymorphism of the MDR1 gene might contribute to susceptibility to HCC in the Chinese population. Further work will be necessary to clarify the relationship between the c.4125A>C polymorphism and susceptibility to HCC on larger populations of diverse ethnicity.

        Anti-Oxidation of Cultured Cordyceps militaris Growing on Silkworm Pupa

        Cordyceps, one of the well-known traditional Chinese medicines, consists of the dried fungus C. sinensis growing on the larva of caterpillar. It is commonly used as a tonic of restoring vital body functions. In order to evaluate the pharmacological effects of C. militaris, the anti-oxidation and antiaging effect of cultured C. militaris growing on pupa of silkworm Bombyx mori, were investigated. The results showed! that the contents of lipofusin in myocardium were reduced and the SOD activity in the liver were elevated in a dose-dependent manner while no effect was observed for blood GSH-Px activity and liver MDA content in mice perfused with the cultured C. militaris compared to those untreated mice. Furthermore, the average life span, maximum life span and 50% death time of the fruit flies treated with cultured C. militaris were markedly prolonged. The results suggest that the cultured C. militaris growing on pupa of silkworm possesses anti-oxidation and delay senescence activity.

        Identification a novel de novo RUNX2 frameshift mutation associated with cleidocranial dysplasia

        Background: Cleidocranial dysplasia (CCD) is a rare genetic disorder affecting bone and cartilage development. Clinical features of CCD comprise short stature, delayed ossification of craniofacial structures with numerous Wormian bones, underdeveloped or aplastic clavicles and multiple dental anomalies. Several studies have revealed that CCD development is strongly linked with different mutations in runt-related transcription factor 2 (RUNX2) gene. Objective: Identification and functional characterization of RUNX2 mutation associated with CCD. Methods: We performed genetic testing of a patient with CCD using whole exome sequencing and found a novel RUNX2 frameshift mutation: c.1550delT in a sporadic case. We also compared the functional activity of the mutant and wild-type RUNX2 through immunofluorescence microscopy and osteocalcin promoter luciferase assay. Results: We found a novel RUNX2 frameshift mutation, c.1550delT (p.Trp518Glyfs*60). Both mutant RUNX2 and wild-type RUNX2 protein were similarly confined in the nuclei. The novel mutation caused abrogative transactivation activity of RUNX2 on osteocalcin promoter. Conclusions: We explored a novel RUNX2 deletion/frameshift mutation in a sporadic CCD patient. This finding suggests that the VWRPY domain may play a key role in RUNX2 transactivation ability.

        Ringing Frequency Extraction Method Based on EMD and FFT for Health Monitoring of Power Transistors

        Condition monitoring has been recognized as an effective and low-cost method to enhance the reliability and improve the maintainability of power electronic converters. In power electronic converters, high-frequency oscillation occurs during the switching transients of power transistors, which is known as ringing. The ringing frequency mainly depends on the values of the parasitic capacitance and stray inductance in the oscillation loop. Although circuit stray inductance is an important factor that leads to the ringing, it does not change with transistor aging. A shift in either the inside inductance or junction capacitance is an important failure precursor for power transistors. Therefore, ringing frequency can be used to monitor the health of power transistors. However, the switching actions of power transistors usually result in a dynamic behavior that can generate oscillation signals mixed with background noise, which makes it hard to directly extract the ringing frequency. A frequency extraction method based on empirical mode decomposition (EMD) and Fast Fourier transformation (FFT) is proposed in this paper. The proposed method is simple and has a high precision. Simulation results are given to verify the ringing analysis and experimental results are given to verify the effectiveness of the proposed method.

        Strain induced hardening and softening behaviors of deformed Cu and Cu-Ge alloys

        <▼1><B>Abstract</B><P/></▼1><▼2><P>Herein, Cu and Cu-Ge alloys with different stacking fault energies (SFEs) are prepared via rolling at room temperature (RTR) and via a combination of high-pressure torsion (HPT) and RTR (HPT + RTR). The x-ray diffraction measurements reveal that the grain size, dislocation density, and twin density vary with the strain and SFEs. The tensile tests indicate that the strength of materials with medium SFEs increases initially and then slightly declines, while the ductility is enhanced by increasing the strain via HPT. In contrast, for low-SFE materials, enhanced strength and improved ductility may be achieved simultaneously through increasing the strain to a high level. The variation of strength with respect to strain is primarily dependent on the solute concentration and SFE. The underlying mechanisms governing the effect of strain and SFE on the microstructures and mechanical properties of the metals are also discussed.</P></▼2>

        Experimental study on vibration characteristics of double-helical gearbox with isolators

        A power closed double-helical gear test rig with isolators was created to investigate the dynamic response characteristics of the double-helical gearbox. Experimentally, the vibration accelerations of the bearing seat, machine foot, and base were tested under various working conditions. Experiments validated the validity of the generalized finite element theorybased dynamic model of the double-helical gear system. The study found that the unloaded vibration of the gearbox is greatly caused by the shaft frequency excitation due to assembly/manufacturing errors. As the load increases, the assembly/manufacturing errors are compensated by the system deformation. In the acceleration spectrum of the bearing seat, it is found that the tooth surface modification can reduce the amplitude of the mesh frequency, but the bearing outer ring frequency components are increased. That is, the axial movement of double-helical gears is intensified. Furthermore, the closer the excitation frequency is to the natural frequency of the base, the worse the vibration isolation effect is.

        The Rapid and Efficient Synthesis of Bromohydrins from Olefins under HBr/H2O2 System by Visible Light Induced

        A simple and safe method has been presented for conversion of olefins into bromohydrins using hydrogen bromide and hydrogen peroxide as bromide source by visible light induced within a very short time to get high yield bromohydrins along with a little mount dibromo-product. In this paper, cyclohexene is firstly carried out as the model substrate and investigated the bromination under HBr/H2O2 system using 150 W incandescent light irradiated in CCl4 within short time to get good yield of 2-bromocyclohexanol along with a little mount of 1,2-dibromocyclohexane; then, a series of alkenes are brominated to corresponding bromohydrins using the same protocol.

        Ectopic Over-expression of Oncogene Pim-2 Induce Malignant Transformation of Nontumorous Human Liver Cell Line L02

        In order to prove that ectopic over-expression of Pim-2 could induce malignant transformation of human liver cell line L02, three groups of cells were set up including human liver cell line L02 (L02), L02 cells transfected with Pim-2 gene (L02/Pim-2) and L02cells transfected with empty-vector (L02/Vector). Pim-2 expression levels were detected. The morphology, proliferation level, apoptosis rate and migration ability of the cells were detected respectively. Then the cells were subcutaneously inoculated into athymic mice and the microstructures of the neoplasm were observed. Compared with the controls, Pim-2expression levels were significantly higher in L02/Pim-2 cells (P<0.05), and their morphology had obvious malignant changes. They also showed a significantly increased proliferation rate (P<0.05) and migration capacity (P<0.05), as well as a significantly decreased apoptosis rate (P<0.05). Only the athymic mice inoculated with L02/Pim-2 cells could generate neoplasm, and the morphology of the neoplasm coincided with that of the hepatoma. The results manifest that ectopic Pim-2 gene could be stably expressed in L02/Pim-2 cells. Both the morphological and biological changes of L02/Pim-2 cells demonstrate the trend of malignant transformation. L02/Pim-2 cells could generate hepatoma in athymic mice. In conclusion, Pim-2 could induce malignant transformation of human liver cell line L02.
