<P><B>Abstract</B></P> <P>To appraise the effects and involved mechanism of <I>Dendropanax morbifera</I> leaves extract (DLE) on diabetes and retinal degeneration, in this study, male C57BLKS/J-m+/db and five groups of db/db mice were treated with 0.5% CMC or metformin or three doses of DLE for 6 weeks. Administration of DLE stimulated AMP-activated protein kinase in the liver, skeletal muscle and adipose tissue, leading to the improvement of gene and protein expression related with insulin resistance and glucose homeostasis in db/db mice. Morphological changes and insulin depletion of pancreatic islets were also attenuated by treatment of DLE. Moreover, DLE treatment prevented the deterioration of retina by regulating not only the gene expression related with apoptosis and neovascularization, but also the level of activator protein-1 in retina. Investigation of a double shrouded probe for particle sampling in high velocity airflows

<P><B>Abstract</B></P> <P>An accurate forecast of the concentration of fine dust in the atmosphere is critical because of the negative public health impacts associated with high concentrations of particulate matter. To achieve an accurate forecast, large volumes of data need to be collected over a wide range of regions to act as forecast model boundary conditions. Therefore, the concentration of fine particles should be measured at both fixed observatories and from a range of moving monitoring stations, including cars, trains, and aviation vehicles. To accurately record particulate concentrations at flow velocities up to 200 km h<SUP>−1</SUP>, this study proposed a double-shrouded probe design based on the widely used single-shrouded probe. Using the double-shrouded probe, isokinetic sampling for PM<SUB>2.5</SUB> was achieved at velocities up to 200 km h<SUP>−1</SUP>, and for PM<SUB>10</SUB> at velocities up to 75 km h<SUP>−1</SUP>. When flow velocities exceeded these values and particle sizes increased, particulate concentrations were overestimated. Downregulation of APE1/Ref-1 Is Involved in the Senescence of Mesenchymal Stem Cells

<P>The senescence of human mesenchymal stem cells (hMSCs) causes disruption of tissue and organ maintenance, and is thus an obstacle to stem cell-based therapies for disease. Although some researchers have studied changes in the characteristics of hMSCs (decreases in differentiation ability and self-renewal), comparing young and old ages, the mechanisms of stem cell senescence have not yet been defined. In this study, we developed a growth curve for human bone marrow derived MSCs (hBMSCs) which changes into a hyperbolic state after passage number 7. Senescence associated beta-galactosidase (SA beta-gal) staining of hBMSCs showed 10% in passage 9 and 45% in passage 11. We detected an increase in endogenous superoxide levels during senescence that correlated with senescence markers (SA beta-gal, hyperbolic growth curve). Interestingly, even though endogenous superoxide increased in a replicative senescence model, the expression of APE1/Ref-1, which is sensitive to intracellular redox state, decreased. These effects were confirmed in a stress-induced senescence model by exogenous treatment with H(2)O(2). This change is related to the p53 activity that negatively regulates APE1/Ref-1. p21 expression levels, which represent p53 activity, were transiently increased in passage 9, meaning that they correlated with the expression of APE1/Ref-1. Overexpression of APE1/Ref-1 suppressed superoxide production and decreased SA beta-gal in hBMSCs. In conclusion, intracellular superoxide accumulation appears to be the main cause of the senescence of hBMSCs, and overexpression of APE1/Ref-1 can rescue cells from the senescence phenotype. Maintaining characteristics of hBMSCs by regulating intracellular reactive oxygen species production can contribute to tissue regeneration and to improved cell therapy.</P>

<P><B><I>Background.</I></B> Recently isolated <I>Vibrio parahaemolyticus</I> strains have displayed multiple antibiotic resistance. Alternatives to conventional antibiotics are needed, especially for the multiple-antibiotic–resistant <I>V. parahaemolyticus</I> pandemic strain.</P><P><B><I>Methods.</I></B> A bacteriophage, designated pVp-1, showed effective infectivity for multiple-antibiotic–resistant <I>V. parahaemolyticus</I> and <I>V. vulnificus</I>, including <I>V. parahaemolyticus</I> pandemic strains. The therapeutic potential of the phage was studied in a mouse model of experimental infection using a multiple-antibiotic–resistant <I>V. parahaemolyticus</I> pandemic strain. We monitored the survivability and histopathological changes, quantified the bacterial and phage titers during phage therapy, and observed the immune response induced by phage induction.</P><P><B><I>Results.</I></B> Phage-treated mice displayed protection from a <I>V. parahaemolyticus</I> infection and survived lethal oral and intraperitoneal bacterial challenges.</P><P><B><I>Conclusions.</I></B> To the best of our knowledge, this is the first report of phage therapy in a mouse model against a multiple-antibiotic–resistant <I>V. parahaemolyticus</I> pandemic strain infection.</P>
