Proliferative Effect of Food Plant Species from Brazil's Cerrado on Cultured Fibroblasts

Brazilian Cerrado is the second largest biome in South America and contains many unstudied valuable plant species rich in bioactive substances. In this study we investigated the phenolic content and proliferative effects on cultured fibroblasts of 32 extracts of different polarities prepared from 11 plants found in Cerrado regions. Eight extracts from six species increased cell proliferation and significantly induced ATP production by the cells. Four of these extracts were obtained from plants used as food, specifically from its fruits or seeds. A high phenolic content for these eight extracts, which directly correlated with the induction of cell proliferation, was corroborated by mass spectrometry analysis. We suggest that the bioactive substance content of these species shows an interesting potential use in cosmetic and food industry, which can contribute to the conservation and sustainable development of this region.
Cytogenotoxic study of aqueous fruit extract of Morinda citrifolia in Wistar albino rats

The use of herbal products and/or their derivatives as an alternative or complementary medicine becomes globally popular. To date, various biological activities have been reported in Morinda citrifolia Linn, especially antioxidants, antimicrobial, anti-infammatory and antitumor efects. This study aimed to investigate the genotoxicity and DNA repair capacity of the aqueous fruit extract of M. citrifolia (AEMC). Additionally, we checked the mutagenic capacity of AEMC by the comet assay and micronucleus tests in rats. For this, rats (n=10; 5 males and 5 females) were divided into fve groups as: negative control (distilled water); positive control (25 mg/kg of cyclophosphamide, i.p.); and three groups for AEMC (2.5, 5 and 10 mg/kg, i.p.). The results suggest that, the AEMC induced genotoxicity in white blood cells, but a DNA repair capacity was also observed after 72 h of AEMC exposure. AEMC also showed cytotoxicity and mutagenicity in liver and kidney cells of rats at 5 and 10 mg/kg doses. In conclusion, AEMC exhibited a dose-dependent cytotoxicity, genotoxicity and mutagenicity along with a time-dependent DNA repair capacity in rats. Further studies are necessary regarding the safe use of this medicinal plant for human consumption.

The present work reports the anti-inflammatory and antinociceptive activities of the ethanol extract obtained from the stem bark of Sterculia striata A. St.-Hil. & Naudin (Ss-EtOH) in the experimental models of edema induced by carrageenan, dextran, or histamin and nociception induced by chemical stimuli, such as acetic acid, formalin, capsaicin, or glutamate. The Ss-EtOH (50 mg/kg) promoted a marked inhibition on the hind paw edema induced by carrageenan or dextran (30% and 73%, respectively). Besides, Ss-EtOH (25 mg/kg) exhibited a slight activity (30%) on the hind paw edema induced by histamin. The Ss-EtOH (12.5 and 25 mg/kg) showed the antinociceptive activity on chemical stimuli induced by acetic acid (65.59% and 38.37%, respectively), formalin, in the initial (35.08% and 31.5%, respectively) and late phases (44.09% and 83.57%, respectively), capsaicin (43.77% and 51.31%, respectively), or glutamate (36.6% and 52.12%, respectively). Regarding the possible mechanism involved in the antinociceptive effect, Ss-EtOH (12.5 mg/kg) showed a decrease in the antinociceptive effect (65.8%) in the acetic acid model after pretreatment with naloxone. Thus, opioid mechanisms might be underlying this response.
DSM Design of Cold-Formed Steel Columns Failing in Distortional Modes at Elevated Temperatures

This paper aims at investigating the structural behaviour, strength and Direct Strength Method (DSM) design of cold-formed steel columns failing in distortional modes at elevated temperatures. The results concern pin-ended and fi xed-ended columns displaying four cross-section shapes, with various dimensions, subjected to 8 temperatures. It is shown that the current DSM distortional design is unable to handle adequately distortional failures at elevated temperatures. Then, a modifi ed DSM design approach is proposed: it consists of incorporating a temperature-dependent reduction factor ratio, based on the EC3- 1.2 model, in the existing strength curve—the modifi ed design curves are shown to yield adequate failure load predictions for the set of columns under consideration. This fi nding provides motivation and encouragement to continue the ongoing search for an effi cient DSM-based design approach for columns failing in distortional modes under elevated temperatures.
Muscle-specific color stability in fresh beef from grain-finished Bos indicus cattle

Objective: To investigate the color and oxidative stabilities of longissimus lumborum (LL) and psoas major (PM) muscles from grain-finished Bos indicus cattle in Brazil. Methods: The LL and PM muscles were obtained 24 h post-mortem from eight (n = 8) Nellore bull carcasses, fabricated into 1.5-cm steaks, aerobically packaged, and stored at 4°C for nine days. Steaks were analyzed for myoglobin concentration, pH, instrumental color, metmyoglobin reducing activity (MRA) and lipid oxidation. Results: The LL steaks exhibited greater (p<0.05) redness, color stability, and MRA than their PM counterparts on days 5 and 9. The LL and PM steaks demonstrated similar (p>0.05) lightness and yellowness on days 0, 5, and 9. On the other hand, PM steaks exhibited greater (p<0.05) myoglobin concentration, pH, and lipid oxidation than their LL counterparts. Conclusion: These results indicated that muscle source influenced the color and oxidative stabilities of beef from grain-finished Bos indicus animals. These results highlighted the necessity of muscle-specific strategies to improve the color stability of beef from grain-fed Bos indicus cattle.
