Clostridium bifermentans strain DPH-1 has already been found to dechlorinate perchloroethylene (PCE) to cis-dichloroethylene (cis-DCE) via trichloroethylene (TCE). In this study, our investigation on different culture conditions of this DPH-1 strain was extended to find a more efficient and cost effective growth medium composition for this DPH-1 strain in bioremediation practices. Temperature dependency of strain DPH-1 showed that the growth starting time and PCE degradation at $15^{\circ}C$ was very slow compared to that of $30^{\circ}C$, but complete PCE degradation occurred in both cases. For the proper utilization of strain DPH-1 in more cost effective bioremediation practices, a simpler composition of an effective media was studied. One component of the culture medium, yeast extract, had been substituted by molasses, which served as a good source of electron donor. The DPH-1 strain in the medium containing molasses, in the presence of $K_{2}HPO_4\;and\;KH_{2}PO_4$, showed identical bacterial multiplication (0.135 mg protein $mL^{-1}h^{-1}$) and PCE degradation rates ($0.38\;{\mu}M/h$) to those of the yeast extract containing medium.

Cancer is a multi-factorial disease and variation in genetic susceptibility, due to inherited differences in the capacity to repair mismatches in the genome, is an important factor in the development of gastric cancer (GC), for example. Epigenetic changes, including aberrant methylation of 5/CpG islands in the promoter regions of mismatch repair (MMR) genes like hMLH1, have been implicated in the development of various types of GC. In the present study we evaluated the role of hMLH1 promoter hypermethylation in Kashmiri GC patients and controls, and assessed correlations with various dietary and lifestyle factors. The study included 70 GC patients (56 males and 14 females; age ($mean{\pm}S.D$) $50{\pm}11.4$ years). Distinction between methylated and unmethylated was achieved with MS-PCR and DNA band patterns. The Chi-square test was applied to assess the risk due to promoter hypermethylation. We found a strikingly high frequency of promoter hypermethylation in GC cases than in normal samples (72.9% (51/70) in GC cases vs 20% (14/70) in normal samples (p=0.0001).We also observed a statistically significant association between methylated hMLH1 gene promoter and smoking, consumption of sundried vegetables and hot salted tea with the risk of GC. This study revealed that hMLH1 hypermethylation is strongly associated with GC and suggested roles for epigenetic changes in stomach cancer causation in the Kashmir valley.

Objectives : This comparative study investigated various methods of advanced oxidation processes (AOPs) that were separately conducted for treating raw petroleum refinery effluent regarding chemical oxygen demand (COD) removal. Methods : Fenton, photo-Fenton, TiO₂, ZnO, TiO₂/Ultra violet (UV), and ZnO/UV were performed individually for measuring the effect of light irradiation, treatment time, pH, catalysts dosage, and light source on the profile of COD values. Results and Discussion : The experimental data of this work showed that the dependency on the light exposure in heterogeneous photo-catalytic reaction using TiO₂ and ZnO is higher than that of homogeneous photo-Fenton technique. The optimum operating conditions in heterogeneous system occurred at 100 min of oxidation time, pH5, and catalyst dosage 1 g/L that resulted in 21.8, 20.68, 60.9, and 55.17% of COD removal for TiO₂, ZnO, TiO₂/UV, and ZnO/UV, respectively. In contrast, both Fenton and photo-Fenton experienced their highest performance at pH 4 by obtaining 44.2 and 59.77% of COD removal, respectively. Eventually, kinetic study indicated that COD degradation can be well expressed by second-order pattern that reached higher correlation coefficient values by 0.999 and 0.998 for TiO₂/UV and TiO₂, respectively. Conclusions : Overall, it could be assumed that AOPs are reliable techniques to purify raw and complex raw industrial effluents.
Hypermethylation Status of E-Cadherin Gene in Gastric Cancer Patients in a High Incidence Area

Gastric cancer (GC) is the fourth most prevalant cancer and the second leading cause of cancer-related mortality worldwide. As in other cancers gastric carcinogenesis is multifactorial involving environmental, genetic and epigenetic components. Epigenetic silencing due to hypermethylation of tumour suppressor genes is one of the key events in gastric carcinogenesis. This study was aimed to analyse the hypermethylation status of the E-Cadherin (CDH1) gene promoter in GCs in the ethnic Kashmiri population. In this study a total of 80 GC patients were recruited. Hypermethylation in tumour tissue was detected by methylation specific PCR (MS-PCR). Hypermethylation of CDH1 promoter was observed in 52 (65%) of gastric carcinoma cases which was significantly much higher than adjacent normal tissue [$p{\leq}0.0001$]. Further the frequency of CDH1 promoter methylation was significantly different with intestinal and diffuse types of gastric cancer [55.7% vs 82.1%; p<0.05]. Moreover females and cases with lymph node invasion had higher frequencies of CDH1 hypermethylation [$P{\leq}0.05$]. Thus the current data indicate a vital role of epigenetic alteration of CDH1 in the causation and development of gastric cancer, particularly of diffuse type, in our population.

The supratrochlear foramen (STF) is an important and relatively common anatomic variation in the lower end of the humerus in humans. The present study on south Indian population is an attempt to highlight the incidence, morphological features and clinical importance of STF. The study was conducted on dried human humeri of unknown sex and free of pathological changes. The presence of a STF was noted for its shape and divided into three types (oval, round, and irregular). In bones where the foramen was absent the translucency of the septum between the coronoid and the radial fossa was noted by placing the lower end of the humerus against the X-ray view box. Out of the 142 humeri studied, 72 humeri (50.7%) were right sided and 70 humeri (49.2%) were left sided. In these 142 humeri, 38 humeri (26.7%) showed the presence of STF. The majority of STF were round shaped in 47.37%, followed by oval shaped in 42.11% and 10.53% were irregular in shape. The STF was absent in 36 humeri (25.35%) and 68 humeri (47.89%) showed the translucency of septum. The mean transverse diameter on right side was 4.50±3.183 mm and 3.32±3.222 mm on left side. The mean vertical diameter was 3.88±2.391 on right side and 3.68±3.532 mm on left side. Its existence is important to the orthopaedician in the preoperative planning of nailing fractures of the distal humerus and to the radiologist for differentiating it from an osteolytic or cystic lesion.

Background: Acute promyelocytic leukemia (APML) is characterized by the reciprocal translocation t(15;17) (p22;p12) resulting in the PML-$RAR{\alpha}$ fusion gene. A dual diagnostic and follow up approach was applied including cytogenetic demonstration of the t(15;17) translocation and detection dg PML-$RAR{\alpha}$ chimeric transcripts by molecular means. Purpose: Conventional cytogenetics involving bone marrow is beset with high probability of poor metaphase index and was substituted with phytohemagglutinin (PHA)-induced peripheral blood culture based cytogenetic analysis as a diagnostic & follow up modality in APML patients of Kashmir (North India). Both qualitative (RT-PCR) and quantitative (Q-PCR) tests were simultaneously carried out to authenticte the modified cytogenetics. Materials and Method: Patient samples were subjected to the said techniques to establish their baseline as well as follow-up status. Results: Initial cytogenetics revealed 30 patients (81%) Positive for t(15;17) whereas 7 (19%) had either cryptic translocation or were negative for t(15;17). Two cases had chromosome 16q deletion and no hallmark translocation t(15;17). Q-PCR status for PML-$RAR{\alpha}$ was found to be positive for all patients. All the APML patients were reassessed at the end of consolidation phase and during maintenance phase of chemotherapy where 6 patients had molecular relapse, wherein 4 also demonstrated cytogenetic relapse. Conclusions: It was found that PHA-induced peripheral blood cytogenetics along with molecular analysis could prove a reliable modality in the diagnosis and assessment of follow up response of APML patients.

This study investigated the restorative effect of Leea macrophylla ethanol root extract (LMERE) in carbon tetrachloride (CCl4) induced hepatic injury. It also tried to unfold the underlying mechanism through ligand-receptor interactions. Prior to conduct the CCl4 induced animal model study, the in vitro antioxidative capacities of LMERE were investigated. Gas chromatography mass spectroscopy (GC–MS) was accomplished to identify the prevalent bioactive compounds. The molecular docking was performed using Schrödinger Suites 2017-1. Results showed the promising antioxidative potentials of LMERE in in vitro models. Upon treatment of CCl4 intoxicated animals with LMERE, serum ALT and AST were found to be significantly (p < 0.05) reduced compared to the CCl4 control while LMERE50 was noted as the best dose in restoring the hepatic markers. Serum lipids and total protein were significantly restored compared to control. Remarkable changes of cell necrosis, apoptosis and sinusoidal dilution were noticed in histopathological assay of liver tissue. mRNA expression for superoxide dismutase (SOD1) and catalase was multifold increased which are statistically significant compared to reference drug, silymarin. In docking study, octadecanoic acid showed the lowest binding energy and highest binding affinity with the protein (ID: 1VKX) which is a crystallized structure of NF-κB p50/p65 heterodimer involved in cytokine production. The findings demonstrate that LMERE restores the hepatic damage by the mRNA expression of antioxidative enzymes while LMERE50, at a glance, seems the most suitable dose.
