        MAP3Kθ1 is Involved in Abscisic Acid Signaling in Drought Tolerance and Seed Germination in Arabidopsis

        Mitogen-activated protein kinase cascades play pivotal roles in mediating environmental stress responses and plant development. In this study, a loss-of-function mutant of Arabidopsis, map3kθ1, exhibited wider stomatal openings, reduced root elongation, and increased seed germination rate compared with its wild type under exogenous abscisic acid (ABA) treatment. MAP3Kθ1 encodes a MAP kinase kinase kinase (MAP3K) with unknown function. Two overexpression lines of MAP3Kθ1 exhibited inhibited seed germination and narrowed stomata, which were aggravated by ABA treatment. Upregulation of MAP3Kθ1 also resulted in stronger drought tolerance, whereas map3kθ1 was more sensitive to water defciency, partially due to diferences in the water-holding capacity of leaves. The MAP3Kθ1-overexpressing lines also showed a greater ability to maintain root elongation under exogenous ABA. Expression of MAP3Kθ1 was inhibited by ABA, H2O2, and methyl viologen treatments in roots. The MAP3Kθ1-overexpressing lines accumulated more ABA by promoting its biogenesis and inhibiting its catabolism, whereas the map3kθ1 mutant accumulated less ABA, compared with wild-type plants. These fndings indicate that MAP3Kθ1 promotes ABA accumulation to regulate stomatal movement, root elongation, and seed germination, while the ABA–H2O2 signaling module inhibits MAP3Kθ1 expression through feedback regulation.

      • Effect Analysis of Speed Guidance Control at Urban Expressway Intersection

        As the backbone of metropolitan transportation, urban expressway shares a large proportion of long distance traffic flow. With the motorization processing, traffic congestion is becoming more and more serious. Intersections are the key interrupted nodes of continuous traffic flow which are the direct inducement of freeway congestion. As an important method of active traffic management, speed guidance control was adopted to improve the road efficiency and safety level. Firstly, speed guidance mathematical model was established based on the traffic flow data and control strategy was also designed for the intersection. Then, online simulation system was built. Microscopic simulation platform was used to simulate the real world traffic. The macro dynamic traffic flow model was established and real-time data were exchange through the application programming interface among the three parts of the simulation system. Next, control effects were analyzed in detail. The results show that vehicle travel time and traffic delay greatly decreased. Emissions of pollutants also have been significantly reduced. Finally, this paper puts forward suitable proposals for the implementation of speed guidance control in China.

        A Numerical Study of Droplet Splitting using Different Spacers Spacers in EWOD Device

        In this paper, we investigated the droplet splitting using different spacers in the air. The droplets were manipulated by the EWOD force and the level set method was used for our numerical implementation. The results show that the property of spacer has the effect on the droplet splitting. Specifically, we showed that the re-merging occurred when split daughter droplets pass the sub-channels for the short spacer and unequal droplet splitting can be achieved for the off-centered obstacle. In the study of the thickness, the thicker spacer made the splitting process unstable. Furthermore, we found that the triangle spacer was optical for the separation from the consideration of pressure. Our task can provide some help for the design of the spacer size in the future.

        The Cement Content Measurement of Cement Mixing Piles with EDTA Titration Method

        The uniformity of cement mixing piles is visually tested on the drilled core samples, significantly relying on experience and extremely inaccurate. The cement content of core samples is an important parameter to quantify the uniformity of cement mixing piles. However, there were no well-accepted methods of testing the cement content of core samples. EDTA titration method is widely used as an effective way to test the cement content of cement-stabilized soil in highway subgrade. However, the cement mixing piles cannot be drilled until 28 days after construction, which causes remarkable change of water content and cement hydration during this period. In addition, factors affecting the EDTA titration results should be investigated before standard EDTA titration curves are constructed. In this paper, effects of different factors including curing method, curing period, soil type, content of water glass, and cement type, on EDTA titration testing results were investigated. It is found that cement type and soil type will significantly affect the test result. It is critical to keep the curing environment and soil unsieved before testing. Water glass will reduce the EDTA solution consumptions at early stages of curing. The EDTA titration results are verified by the proposed density estimation method.

        A Rapid and Effective Colony PCR Procedure for Screening Transformants in Several Common Mushrooms

        In the post-genomic era, gene function analysis has attracted much attention. Transformation is often needed to investigate gene function. In this study, an easy, rapid, reliable, and cost-effective colony polymerase chain reaction (PCR) method for screening mushroom transformants was developed: picking up a suitable amount of transformant’s tissue (1-10 lg) to 20 ll 0.25% Lywallzyme solution, and vortexing for 10 s followed by incubation at 34 ℃ for 15 min. Finally, 2 ll of the suspension was used as templates to perform PCR and single target bands were successfully amplified from respective transformants of Tremella fuciformis, Pleurotus ostreatus, and Pleurotus tuber-regium. This procedure could be widely employed for screening transformants in mushroom transformation experiments.

        Comparing the sugar profiles and primary structures of alkali-extracted water-soluble polysaccharides in cell wall between the yeast and mycelial phases from Tremella fuciformis

        To gain insights into dimorphism, cell wall polysaccharides from Tremella fuciformis strains were obtained from alkaliextracted water-soluble fractions PTF-M38 (from the mycelial form), PTF-Y3 and PTF-Y8 (from the yeast form) of T. fuciformis strains were used to gain some insights into dimorphism study. Their chemical properties and structural features were investigated using gel permeation chromatography, gas chromatography, UV and IR spectrophotometry and Congo red binding reactions. The results indicated that the backbones of PTF-M38, PTF-Y3 and PTF-Y8 were configured with α-linkages with average molecular weights of 1.24, 1.08, and 1.19 kDa, respectively. PTF-M38 was mainly composed of xylose, mannose, glucose, and galactose in a ratio of 1:1.47:0.48:0.34, while PTF-Y3 and PTF-Y8 were mainly composed of xylose, mannose and glucose in a ratio of 1:1.65:4.06 and 1:1.21:0.44, respectively. The sugar profiles of PTF-M38, PTF-Y3 and PTF-Y8 were also established for further comparison. These profiles showed that all three polysaccharides contained the same sugars but in different ratios, and the carbon sources (xylose, mannose, glucose, and galactose) affected the sugar ratios within the polysaccharides.

        An Enzymolysis-Assisted Agrobacterium tumefaciens-Mediated Transformation Method for the Yeast-Like Cells of Tremella fuciformis

        Agrobacterium tumefaciens-mediated transformation (ATMT), as a simple and versatile method, achieves successful transformation in the yeast-like cells (YLCs) of Tremella fuciformis with lower efficiency. Establishment of a more efficient transformation system of YLCs is important for functional genomics research and biotechnological application. In this study, an enzymolysis-assisted ATMT method was developed. The degradation degree of YLCs depends on the concentration and digestion time of Lywallzyme. Lower concentration (_0.1%) of Lywallzyme was capable of formation of limited wounds on the surface of YLCs and has less influence on their growth. In addition, there is no significant difference of YLCs growth among groups treated with 0.1% Lywallzyme for different time. The binary vector pGEH under the control of T. fuciformis glyceraldehyde-3-phosphate dehydrogenase gene (gpd) promoter was utilized to transform the enzymolytic wounded YLCs with different concentrations and digestion time. The results of PCR, Southern blot, quantitative real-time PCR (qRT-PCR) and fluorescence microscopy revealed that the T-DNA was integrated into the YLCs genome, suggesting an efficient enzymolysis-assisted ATMT method of YLCs was established. The highest transformation frequency reached 1200 transformants per 10<sup>6</sup> YLCs by 0.05% (w/v) Lywallzyme digestion for 15 min, and the transformants were genetically stable. Compared with the mechanical wounding methods, enzymolytic wounding is thought to be a tender, safer and more effective method.

        An Improved Total RNA Extraction Method for White Jelly Mushroom Tremella fuciformis Rich in Polysaccharides

        An improved method for extracting high quality and quantity RNA from a jelly mushroom and a dimorphic fungus―Tremella fuciformis which is especially rich in polysaccharides, is described. RNA was extracted from T. fuciformis mycelium M1332 and its parental monokaryotic yeast-like cells Y13 and Y32. The A260/280 and A260/230 ratios were both approximately 2, and the RNA integrity number was larger than 8.9. The yields of RNA were between 108 and 213 μg/g fresh wt. Downstream molecular applications including reverse transcriptional PCR and quantitative real-time PCR were also performed. This protocol is reliable and may be widely applicable for total RNA extraction from other jelly mushrooms or filamentous fungi rich in polysaccharides.

        <P>The ability to tune material properties using gating by electric fields is at the heart of modern electronic technology. It is also a driving force behind recent advances in two-dimensional systems, such as the observation of gate electric-field-induced superconductivity and metal-insulator transitions. Here, we describe an ionic field-effect transistor (termed an iFET), in which gate-controlled Li ion intercalation modulates the material properties of layered crystals of 1T-TaS2. The strong charge doping induced by the tunable ion intercalation alters the energetics of various charge-ordered states in 1T-TaS2 and produces a series of phase transitions in thin-flake samples with reduced dimensionality. We find that the charge-density wave states in 1T-TaS2 collapse in the two-dimensional limit at critical thicknesses. Meanwhile, at low temperatures, the ionic gating induces multiple phase transitions from Mott-insulator to metal in 1T-TaS2 thin flakes, with five orders of magnitude modulation in resistance, and superconductivity emerges in a textured charge-density wave state induced by ionic gating. Our method of gate-controlled intercalation opens up possibilities in searching for novel states of matter in the extreme charge-carrier-concentration limit.</P>
