        유색 보리 종실의 품종별 색소 특성

        유색보리 4품종과 보통보리 4품종 종실의 종피에 함유되어 있는 색소의 특성을 분석하여 해당작물의 신품종 육성시 우수계통 선발에 유용한 자료를 제공코자 하였다. 유색(검정)쌀보리인 Ab2231을 표준시료로 하여 실험한 결과 에탄올의 농도가 80%일 때 색소추출 효율이 양호하였다. 80% 에탄올에 1.0% 염산을 첨가한 것이 추출효율이 가장 좋았다. 추출시간에 따른 추출효율성을 조사한 결과 1시간 추출이면 충분하였다. 추출된 색소의 안정성을 유지하기 위하여 첨가시키는 산의 적정농도를 알아본 결과 1.0% HCl을 첨가한 농도에서 가장 낮은 색소량 감소율이 나타났다. 각 보리 품종 시료에서 추출한 색소의 흡광스펙트럼을 조사하였던 바, Ab2231의 그것은 535㎚에서 최대흡광파장을 나타내어 안토시아닌계 색소로 판단되었으며, 호계4호의 그것은 안토시아닌계 이외의 색소를 주로 함유하는 것으로 보였다. 한편, 추출된 색소의 특성에 따라 유색보리의 품종은 Ab2231와 모찌무기로 대표되는 쌀보리 그룹과 Lion과 호계4호로 대표되는 겉보리 그룹으로 분류되었다. The characteristics of the pigments extracted from grains of the colored barley varieties Ab2231. Mozzimugi, Lion, and Hogye 4 were studied. Extraction of the pigments from powder (80 mesh) of the barley grains was most efficient in 80% ethanol containing 1.0% HCI for 1 hr at 60℃. The pigments were also stable in 80% ethanol containing 1.0% HCI. Depending on the absorption spectra, maximal peak wavelength, and the ratio of A_(440)/Anu_(max), the varieties were separated into two groups, colored naked barley (Ab2231 and Mozzimugi) and colored covered barley (Lion and Hogye 4).

        Screening of 1-deoxynojirimycin (DNJ) producing bacteria using mulberry leaf

        1-Deoxynojirimycin (DNJ) has been extensively investigated for its applications as an a-glucosidase inhibitor in postprandial hyperglycemia, and has been applied to nutraceuticals and medicines to prevent or delay the progression of type 2 diabetes. However, the amount of DNJ in mulberry leaves is low (approximately 0.1%), therefore, a more effective extraction method is needed. In this study, microbial DNJ production was developed as an alternative to chemical methods. We identified fermented sericultural products and bacteria that produce DNJ in large quantities using high performance liquid chromatography and thin layer chromatography. The inhibition of a-glucosidase activity was examined with respect to DNJ production or non-production. Crude DNJ from the isolated strains exhibited greater than 70% a-glucosidase activity. An investigation of the effect of mulberry leaf powder concentration (1~5%), using high DNJ producing bacteria, provided evidence for microbial mass production of DNJ

        Effects of Physiological Active Substance Extracted from Silkworm Fece

        Silkworm (Bombyx mori ) feces have long been used in the pharmaceutical and food industriesas a natural colorant. However, there is limited data on the bioactive compounds thatconstitute silkworm feces. This research emphasizes the antioxidant activity of different solventand flavonoid extracts of silkworm feces. The solvents were ethanol, butanol, and water, whilethe methods utilized included ultrasonification, stirrer, reflux, and reflux after ultrasonificationextraction. Results showed that butanol ultrasonification extraction (BUE) yield the lowestextraction (1.75%), while the other methods yielded 7 to 14%. The total polyphenol contentutilizing BUE was 3.3 mg TAE/g, while water ultrasonification extraction (WUE) yielded thehighest extraction rate with 51.6 mg TAE/g. The total flavonoid content was significantly higherusing ethanol reflux extraction (EUE) at 266.8 mg QRE/g BUE, which was 158.3 and 151.3mg QRE/g. Both DPPH radical scavenging activity and SOD-like (superoxide dismutase)activity, showed significant antioxidant effects. Finally, all other extracts except for BUE hada-glucosidase inhibition at 60%. Therefore, an effective extraction method for physiologicallyactive substances must be selected.

        Quantitative Analysis of 1-Deoxynojirimycin Content Using Silkworm Genetic Resources

        1-Deoxynojirimycin(1-DNJ or DNJ), a component in silkworm powder, prevents glucose frombeing absorbed into the bloodstream from the small intestine by inhibiting a-glucosidaseactivity. This study compared the functional components of 1-DNJ from various silkwormspecies using a gene database with those of 1-DNJ produced by silkworms bred throughcross-combinations. We utilized comparisons of geographical origins and species ofsilkworms using a gene database and discovered that 1-DNJ activity was ranked in thefollowing order by species, Japanese (SK-1) > Chinese (C48) > European (Rock191). 1-DNJconstituted varying percentages of silkworm organs in the following order, blood > epithelialtissue > body fat > silk glands. With regard to sex, 1-DNJ, levels were higher in males thanfemales. However, 1-DNJ levels with respect to various genetic traits (blood color, silk color,and egg color) were consistent. In order to study 1-DNJ changes that occurred during crossbreeding of the silkworm gene, we bred cross-combinations utilizing SK-1 and C48 silkworms. In conclusion, in order to provide information about the constituents of functional materialscontained in silkworm powder, it is imperative that silkworm cross breeding occurs so that thedatabase of functional materials extracted from silkworms will expand.

        Anti-atopic Effects of the Silkworm Feces Extracts in the NC/Nga Mice

        This study was evaluated the antiatopic activity of silkworm feces extracts in DNCBinducedNC/Nga atopy mice. It was found that each level of IgE and histamine in blood wassignificantly decreased in the silkworm feces extracts treatment groups, compared with theDNCB-induced atopy control group. When the silkworm feces extracts was applied to theatopy mice, it could be observed that its skin recovered to normal condition with the skinsurface being clean and smooth without any tissue. The results suggest that the application ofsilkworm feces extracts effect on atopic model through a inhibition of histamine emissions withreducing the levels of blood IgE in NC/Nga atopy mice.

        Investigation of mulberry farm’s soil properties and mulberry leaf nutritive components in local areas of Korea

        Mulberry is a hardy, perennial, deep-rooted plant capable of thriving under diverse agroclimatic conditions. The selection of suitable land and appropriate variety can help the sustainable mulberry field. However, no conclusive and comprehensive investigation has been conducted on the mulberry soil properties and nutritional composition of mulberry cultivars from Korea local areas in previous studies. In our study, soil properties and mulberry leaf components of Korea local mulberry farms were briefly investigated. In result, the soil organic matter (OM) content was significantly high in Buan (6.81%) and Jangseong (6.14%). In contrast, available phosphate (P2O5) was different in each local area. To investigate relationship between soil property and nutritive component of mulberry leaf, Cheongil leaf samples from 8 local areas were analyzed. Among the macrominerals (K+ , Ca2+, Na+ , and Mg2+), the concentration of K varied from (1884±9.36) mg/100 g to (2685±11.5) mg/100 g. The potassium (K+ ) of Cheongil leaf in Sangju was the highest at 2685 mg/100 g. Besides macrominerals, flavonoids, total dietary fiber contents and moisture of Cheongil leaf samples were studied in the 8 local areas. In terms of these contents, the variation was largely depending on the local areas. This study provides a possible industrial use of mulberry, and holds promise to enhance the overall profitability of sericulture.

        Flavonoids analysis about mulberry fruit of Korean mulberry cultivar, ‘Daeshim’

        Mulberry fruit is a new income product in Korea sericulture due to the increase of fruit consumption. However, flavonoids of Korean mulberry cultivar for fruit production did not reported yet. In this study, the typical mulberry cultivar, ‘Daeshim’ was analyzed using ultrahigh performance liquid chromatography coupled with diode array detection and quadrupole time-of-flight mass spectrometry (UPLC-DAD-QTOF/MS) technique for flavonoids analysis. Nine flavonoids were isolated and analyzed from Daeshim using UPLC-DAD-QTOF/ MS chromatogram. According to quantitative analysis, rutin (66.1 mg/100g DW) and quercetin 3-O-(6”-O-malonyl) glucoside (26.7 mg/100g DW) were abundant in mulberry fruit. Our results might be used as basic information for mulberry consumption

        Quantitative analysis of rutin content using silkworm genetic resources

        Rutin is an important bioflavonoid that is consumed in the daily diet. This study compared the functional components of rutin from various silkworm species using a gene database with those of rutin produced by silkworms selectively bred through cross-combinations. We made comparisons between the geographical origin and species of silkworm using a gene database and discovered that rutin activity was ranked in the following order by species, Chinese (C5)> miscellaneous varieties (Jamsaeng 1 Ho) >Japanese (Jam 115) > European (E58) >Korean (Sun 3 ho). However, rutin levels with respect to various genetic traits (blood color, silk color, and egg color) were consistent. In order to study rutin changes that occurred during the cross breeding of the silkworm gene, we bred cross-combinations utilizing Jam 115 and the 4051 silkworms. In conclusion, in order to provide information about the constituents of functional materials contained in silkworm powder, it is imperative that silkworm cross breeding occurs so that the database of functional materials extracted from silkworms will expand

        Comparison of optimal temperature and time conditions for highest a-glucosidase inhibitory activity from various of Korea mulberry teas

        The influence of commonly used steeping times and temperatures about mulberry tea on a-glucosidase inhibitory activity were studied. The effects of product and preparation variables on the in-cup chemical composition of mulberry tea extracts is of interest because the appearance and taste characteristics and the possible health effects of a tea liquor arise from the chemical components extracted from the leaf during mulberry tea preparation. A comprehensive study was therefore undertaken to determine the contributions of product and provide a basic data for development of high quality mulberry tea products against diabetes. Specific eight mulberry tea were collected from each region of Korea (A-I) and when it was extracted on 85°C for 2min, a-glucosidase activity was best for 98-102%. According to various temperature and time extraction methods, this study was carried out to optimize teamaking conditions for maximal DNJ extraction from Korean mulberry tea and and it would be of particular interest for people who drink mulberry tea to control blood glucose levels.
