This study was carried out as a preliminary step for analyzing the vitamin D₂ and D₃ in feedstuffs using HPLC. 1. Absorbance curve following the wavelength of UV light showed that the 264㎚ gave a maximum absorbance. 2. The mixed solution of 20 ppm vitaminD₂ and D₃ was injected in HPLC with μ Bondapak C_(18) column. The vitamin D₂ was resolved at 14.8 min. and D₃ at 15.4 min. after injection when eluted with 95% methanol + 5 % H₂O. 3. Varying amount of vitamin D₂ was injected into the column of μ Bondapak C_(18), and the detector 240 showed that the least detectable level was 5ng and that 5-100ng range brought a linear calibration curve. 4. When μ Bondapak column and 440 detector were used, 10-100ng range gave a linear standard curve. 5. When μ Porasil column and 440 detector were applied, 10-100ng range showed a linear calibration curve.
자외선 조사에 의한 8주령 브로일러 병아리 볏 표피에서의 비타민 D3 및 그의 유사물질 함량 변화

This study was carried out to investigate the increment of previtamin D₃(PreD₃), lumisterol₃(L₃) and vitamin D₃(D₃) in comb epidermis of 8 weeks old broiler chicks when the epidermis was irradiated by UV light in vitro. The day-old Hubbard line broiler chicks were fed with vitamin D-deficient diet for 3 weeks and with finisher ration for 5 weeks. The UV light at 310 nm was utilized to irradiate the epidermis for up to 150 min. The lipid in UV-exposed epidermis was extracted by 60% ethyl acetate in n-hexane and PreD₃, L₃, D₃ and 7-dehydrocholesterol (7-DHC) in the lipid were analyzed by normal phase HPLC. When the comb epidermis was exposed to UV light for 0, 90 or 150 :min, the L₃ content was 525, 720 or 520 ng/㎠ respectively. The increasing rate of L₃ was 2.2 ng/㎠/min up to 90 min. The PreD₃ content was increased from original 175 to 420 ng/㎠ when exposed for 30 min and the increasing rate was 8.2 ng/㎠/min. The mean D₃ concentration in epidermis was increased from initial 50 to 140 ng/㎠ by 30 min irradiation and the rate of increment was 3.0 ng/㎠/min. The 7-DHC content was decreased from 1950 to 350 ng/㎠ when exposed to UV light for 150 min, and the decreasing rate was 10.7 ng/㎠/min.
무창 (無窓) 약등시 (弱燈時) 비타민 D3 첨가수준이 브로이러 병아리의 증체 (增體) , 영양소이용율 , 발가락무기물 및 혈청성상에 미치는 영향

This study was carried out to observe the effect of vitamin D₃ supplementation levels(0-2500 IU/㎏) on body weight gain, nutrient utilization, toe minerals and serum characteristics of broiler chicks fed cornsoya-fish meal diet in a window-less room under subduced lighting. The arginine content of starter diet was 66% of the level recommended by NRC(1977) and that of grower diet was 90%. The day-old 378 chicks (Hubbard) were allocated at triplicate 9 treatments. In respect to body weight gain an increasing figure was found up to 800 IU/㎏ vitamin D₃ treatment. A slowly increasing trend was obtained from 800 to 2500 IU/㎏ groups. The feed intakes of chicks increased when fed more vitamin D₃ up to 2500 IU/㎏ There was a highly significant correlation between body weight gains and feed intakes of chicks experimented. However, no significant difference was found among treatments in relation to feed conversion, mortality and incidence of weak legs. No significant difference was recognized among treatments in respect to the utilization efficiencies of dry matter, crude protein, and ether extract, too. An increasing figure was shown in utilization efficiencies of nitrogen-free extract when the supplemening level of vitamin D₃ increased. The utilization efficiencies of crude ash were similar among treatments, but chicks fed diet containing 200IU/ ㎏ vitamin D₃ gained lowest values. The utilization of calcium, phosphorus, magnesium, potassium, sodium, iron, zinc and copper were similar among treatments. The contents of ash and minerals in toes of broiler chicks were similar among groups tested. A significant difference was recognized among calcium contents of serum. However, neither increasing nor decreasing trend was fund as more vitamin D₃ was supplied. The similar contents of phosphorus and activities of alkaline phosphatase in serum were obtained among treatments.
HPLC 에 의한 닭 표피중의 Vitamin D3 및 그 유사물질 분석

This study was carried out to analyze the contents of vitamin D₃ and it`s analogues in chicken epidermis. The retention time and calibration curve were obtained through HPLC. The comb, wattle or dorsal epidermis of 57 week-old breeder irradiated by 310 nm UV hight was extracted by 60% ethyl acetate in n-hexane with appropriate amounts of vitamin D3 and it`s analogues, dried under mitrogen, dissolved in 0.5% isopropanol in n-hexane, analyzed by HPLC, and obtained the recoveries of the compounds. 1. The mean retention times of previtamin D₃(PreD₃3), lumisterol₃(L₃), vitamin D₃(D₃), tachysterol₃(T₃) and 7-dehydrocholesterol (7-DHC) were 6.95, 7.65, 8.90, 12.83 and 14.68 min., respectively. 2. The mean recovery of [³H]-7-DHC through HPLC was 88%. 3. The mean recoveries of PreD₃, L₃, D₃, T₃ and 7-DHC through extraction and HPLC analysis were 72, 34, 27, 13 and 90%, respectively.
사료내의 비타민 D2 및 D3 분석방법에 관한 연구 2 . 정제과정이 정상액체 크로마토그라피를 이용한 비타민 D2 및 D3 분석에 미치는 영향

This study was carried out to establish an analytical method of vitamin D₂ or D₃ in feedstuffs. The ladino clover and orchard grass hays, and calf grower ration were used as samples. The vitamin D₂ or D₃ solution was added into the samples as an internal standard. The sample and vitamin D were saponified, extracted, and cleaned up through alumina, polyethylene glycol 600-Chromosorb W and florex columns. Then Sep-Pak silica and C_(18) cartridges were utilized for further purification. The Sephadex LH-20 column and μ Hondapak C_(18) column (HPLC, reverse phase) were applied to remove the remained impurities. Finally the normal phase μ Porasil column HPLC was utilized to quantitate the vitamin. It was known that the mean recovery of internal standard was 68% (S.D. 6%). The vitamin D₂ contents in ladino clover and orchard grass hays were 1,280±83 and 1,324±79 IU/㎏, respectively. The vitamin D₃ in calf grower ration was 1,463±102 IU/㎏.
