        사료내의 비타민 D2 및 D3 분석방법에 관한 연구 2 . 정제과정이 정상액체 크로마토그라피를 이용한 비타민 D2 및 D3 분석에 미치는 영향

        This study was carried out to establish an analytical method of vitamin D₂ or D₃ in feedstuffs. The ladino clover and orchard grass hays, and calf grower ration were used as samples. The vitamin D₂ or D₃ solution was added into the samples as an internal standard. The sample and vitamin D were saponified, extracted, and cleaned up through alumina, polyethylene glycol 600-Chromosorb W and florex columns. Then Sep-Pak silica and C_(18) cartridges were utilized for further purification. The Sephadex LH-20 column and μ Hondapak C_(18) column (HPLC, reverse phase) were applied to remove the remained impurities. Finally the normal phase μ Porasil column HPLC was utilized to quantitate the vitamin. It was known that the mean recovery of internal standard was 68% (S.D. 6%). The vitamin D₂ contents in ladino clover and orchard grass hays were 1,280±83 and 1,324±79 IU/㎏, respectively. The vitamin D₃ in calf grower ration was 1,463±102 IU/㎏.

        HPLC 에 의한 닭 표피중의 Vitamin D3 및 그 유사물질 분석

        This study was carried out to analyze the contents of vitamin D₃ and it`s analogues in chicken epidermis. The retention time and calibration curve were obtained through HPLC. The comb, wattle or dorsal epidermis of 57 week-old breeder irradiated by 310 nm UV hight was extracted by 60% ethyl acetate in n-hexane with appropriate amounts of vitamin D3 and it`s analogues, dried under mitrogen, dissolved in 0.5% isopropanol in n-hexane, analyzed by HPLC, and obtained the recoveries of the compounds. 1. The mean retention times of previtamin D₃(PreD₃3), lumisterol₃(L₃), vitamin D₃(D₃), tachysterol₃(T₃) and 7-dehydrocholesterol (7-DHC) were 6.95, 7.65, 8.90, 12.83 and 14.68 min., respectively. 2. The mean recovery of [³H]-7-DHC through HPLC was 88%. 3. The mean recoveries of PreD₃, L₃, D₃, T₃ and 7-DHC through extraction and HPLC analysis were 72, 34, 27, 13 and 90%, respectively.

        사료의 비타민 D2 및 D3 분석방법에 관한 연구 1 . 고성능 액체 크로마토그라프를 이용한 표준곡선 작성

        This study was carried out as a preliminary step for analyzing the vitamin D₂ and D₃ in feedstuffs using HPLC. 1. Absorbance curve following the wavelength of UV light showed that the 264㎚ gave a maximum absorbance. 2. The mixed solution of 20 ppm vitaminD₂ and D₃ was injected in HPLC with μ Bondapak C_(18) column. The vitamin D₂ was resolved at 14.8 min. and D₃ at 15.4 min. after injection when eluted with 95% methanol + 5 % H₂O. 3. Varying amount of vitamin D₂ was injected into the column of μ Bondapak C_(18), and the detector 240 showed that the least detectable level was 5ng and that 5-100ng range brought a linear calibration curve. 4. When μ Bondapak column and 440 detector were used, 10-100ng range gave a linear standard curve. 5. When μ Porasil column and 440 detector were applied, 10-100ng range showed a linear calibration curve.

        자외선 조사에 의한 브로일러 병아리 피부에서의 Vitamin D3 유사물질 합성

        Studies on the synthesis of previtamin D₃(PreD₃), lumisterol₃(L₃) and vitamin D₃(D₃) in dorsal skin of broiler chicks irradiated by ultraviolet(U.V.) light of different wavelength were carried out. The dorsal skin of 3 week-old chicks fed vitamin D-defficient diet were exposed to U.V. light of 254, 310 or 365nm for 0, 30, 60, 120 or 150 minutes. The vitamin D₃ and its analogues were extracted from epidermis of dorsal skin and analyzed by HPLC. 1. When the dorsal skin of 3 week-old chicks were irradiated by 254nm UN light, the 7-DHC contents were decreased continuosly from initial 255 to 55ng/㎠ for 150 min exposure; the decreasing amount was 200ng/㎠ and the decreasing rate was 1.3ng/㎠/min. The L₃ contents were increased from first 110 to 115ng/㎠ of 150min. irradiation ; 5ng/㎠ was synthesized, being the rate of 0.03ng/㎠/min. The PreD₃ contents were decreased from 58 to 5ng/㎠ of 150min. exposure; 53ng/㎠ of PreD₃ was decreased at the rate of 0.14ng/㎠/min. The D₃ contents were increased from original 2 to 30ng/㎠ of 150 min. irradiation ; 28ng of D₃ was produced and the production rate was 0.19ng/㎠/min. 2. As the dorsal skin of 3 week-old chicks were exposed to the 310nm U.V. light, the 7-DHC contents were decreased from initial 340 to 27ng/㎠ for 150mm. exposure ; 313ng/㎠ was disappeared at the rate of 2.1ng/㎠/min. The L₃ contents were increased from original 170 to 400ng of 60 min. irradiation ; 230ng/㎠ of L₃ was synthesized at the rate of 3.8ng/㎠/min. The D₃ contents were increased from 85 to 320ng/㎠ of 90 min. irradiation ; 235ng/㎠ of D₃ was synthesized at the rate of 2.6ng/㎠ min. The PreD₃ contents were changed from 52 to 70ng/㎠ of 60min. treatment ; 18ng/㎠ was produced at the rate of 0.3ng/㎠/min. 3. When the dorsal skin of 3 week-old chicks were irradiated by 365nm U.V. light, the 7-DHC consents were diminished from 800 to 200ng/㎠ by 150 min. exposure ; 600ng/㎠ was disappeared at the rate of 4ng/㎠/min. The PreD₃ contents were increased from initial 40 to 75ng/㎠ by 60 min. irradiation ; 35ng/㎠ were synthesized at the rate of 0.6ng/㎠/min. The L3 contents were increased from original 12 to 15ng/㎠ of 30 min. exposure ; 3ng/㎠ was produced at the rate of 0.1ng/㎠/min. The D₃ contents were increased from initial 0 to 50ng/㎠ of 90 min. exposure ; 50ng/㎠ was generated at the rate of 0.56ng/㎠/min. 4. It was verified that 310nm U.V. light would be desirable to irradiate the dorsal skin of 3 week-old broiler chicks rather than 254 or 365 nm lamp in order to sythesize D₃ and its analogues.
