        Paclitaxel에 의한 관절연골 세포의 capase-비의존적 mitotic catastrophe 유도

        Paclitaxel은 미세소관의 탈중합을 억제하는 시약으로 알려져 있다. Paclitaxel은 다양한 세포에서 세포 내 방추체를 안정화시킴으로써 유사분열 억제 및 세포사멸을 유도한다. 본 실험에서는 토끼 관절 연골세포에서 paclitaxel이 연골세포의 증식과 사멸에 미치는 효과에 대한 연구를 수행하였다. MTT assay를 수행한 결과 paclitaxel은 연골세포에서 농도 의존적으로 세포 증식을 억제한다는 것을 확인 할 수 있었으며, FACS analysis와 Western blot analysis를 수행한 결과, paclitaxel이 G2/M 정지를 유도하는 것을 확인하였다. 또한, paclitaxel이 비정상적인 세포 분열유도와 핵 단편분절 유도없이 일어나는 mitotic catastrophe 즉, caspase-3 비의존적인 세포사멸을 유도하였다. Paclitaxel을 처리한 세포에서 일어나는 이러한 mitotic catastrophe에 의한 세포 죽음은 Gl/S기의 진행을 억제하는 시약인 thymidine을 처리하는 것에 의해 억제되는 것을 확인할 수 있었다. 이러한 결과를 종합해 볼 때, paclitaxel에 의한 토끼 관절 연골 세포에서의 세포죽음은 caspase-3 비의존적인 mitotic catastrophe에 의해 일어나는 것으로 사료되어진다. Paclitaxel is known as a potent inhibitor of microtubule depolymerization. It leads to mitotic arrest and cell death by stabilizing the spindle in various cell types. Here, we investigated the effects of paclitaxel on the proliferation and cell death of rabbit articular chondroeytes. Paclitaxel inhibited proliferation in a dose- and time- dependent manner, determined by MTT assay in rabbit articular chondrocytes. We also established paclitaxel-induced G2/M arrest by fluorescent activated cell sorter (FACS) analysis. Paclitaxel increased expression of cyclin B, p53 and p2l, while reducing expression of cdc2 and cdc25C in chondrocytes, as detected by Western blot analysis. Interestingly, paclitaxel showed the mitotic catastrophe that leads to abnormal nucleus division and cell death without DNA fragmentation through activation of caspase. Cell death by mitotic catastrophe in cells treated with paclitaxel was suppressed by inhibiting G1/S arrest with 2 mM thymidine. These results demonstrate that paclitaxel induces cell death via mitotic catastrophe without activation of casepase in rabbit articular chondrocytes.

      • 요추부 퇴행성 추간판의 자기공명 영상소견에 따른 체외배양 수핵세포주의 노화 인자와의 관련성에 대한 연구

        Objectives: The authors conducted this study to investigate the relationship between signal to noise ratio in T2 weighted signal intensity of intervertebral disc (IVD) on MRI and biomarkers associated with senescence changes. Methods: Five patients (5 male) who underwent open discectomy or posterior lumbar discectomy and fusion for symptomatic herniated nucleus pulposus (NP) were included. The NP specimens were grouped according to a grading system for IVD degeneration based on the preoperative MRI and SNR was measured. The SA-b-gal activity of the NP cells was determined and the percentage of SA-b-gal-positive NP cells were calculated. The telomerase activity was analyzed and the amount of TRAP products was determined by measurement of their absorbance at 450 nm. Results: The mean patient age was 35.2 years (range: 21-52 years, SD ±17.283). There was no significant correlation between every age and SNR (Spearman’s test, r2=0.440, p=0.111). As degenerative changes became more distinct, decrement of SNR showed each senescence marker changed to the same tendency in the patient with age of fifties. The correlation between SNR and SA-b-gal was not statistically significant. However, SNR was slightly correlated with SA-b-gal. (R2 = 0.0404, p = 0.373) Despite the lack of statistical significance, there was a negative correlation between telomerase activity and SNR. (R2 = 0.3103, p = 0.165) In aged group of twenties, the means of the percentages of the SA-b-gal-positive NPCs from the patients aged 21, 21, and 26 years were 63.25%, 42.21%, and 62.27%, respectively, at passage 3 and the means of the percentages of the SA-b-gal-positive NPCs from the patients aged with fifties were 56.97% and 71.84% respectively at passage 3. There was a steady decline in the telomerase activity of the NP cells with advancing passages, irrespective of patient age. However, this tendency did not have statistical significance. (p>0.05) Conclusions: The NP cells in aging discs exhibited characteristic senescent features such as an increased SA-b-gal expression and decreased telomerase activity. SNR is associated with degenerative changes of NP, and as it increases, the number of SA-b-gal positive cells increases and telomerase activity tends to decrease.

        과학연극활동이 유아의 과학적 탐구능력 및 창의성에 미치는 영향

        본 연구의 목적은 과학연극활동이 유아의 과학적 탐구능력 및 창의성에 미치는 영향을 알아보는데 있다. 본 연구 대상은 S시에 위치한 B유치원 만 5세 유아 36명(실험집단 18명, 비교집단 18명)이다. 총 6주 동안 실험집단은 유아를 대상으로 한 과학연극활동을 실시하였다. 연구도구는 Martin(1997)의 과학적 탐구능력 평가 척도를 기초로 하여 이경민(2000)이 제작하고 서윤희(2003)가 수정 · 보완한 ‘과학적 탐구능력 검사’를 사용하였고, 유아의 창의성을 검사하기 위해 Torrance(1990)가 개발한 창의성 검사(Torrance Tests of Creative Torrence; TTCT)를 김영채(2010)가 수정 · 제작한 창의성 검사 〈한국판 TTCT(도형)검사〉를 사용하였다. 과학연극활동이 유아의 과학적 탐구능력 및 창의성에 미치는 영향을 분석한 결과는 다음과 같다. 첫째, 과학연극활동은 유아의 과학적 탐구능력 전체 및 하위개념인 ‘예측하기’, ‘관찰하기’, ‘분류하기’, ‘토의하기’의 향상에 긍정적인 영향을 미치는 것으로 나타났다. 둘째, 유아의 창의성 전체 및 하위개념인 ‘유창성’, ‘독창성’, ‘정교성’, ‘성급한 종결에 대한 저항’이 비교집단의 활동에 비해서 향상된 것으로 나타났다. 이러한 연구결과는 과학연극활동이 유아의 과학적 탐구능력 및 창의성을 향상시키는데 효과가 있음을 의미한다. 본 연구는 만 5세의 과학적 탐구능력과 창의성 향상을 위한 교수학습방법에 대한 새로운 시도로 유아교육현장에서 과학연극활동이 활성화되기 위한 기초자료를 제공할 것으로 기대된다. The purpose of this study was to examine the effect of science drama activities on young children’s scientific inquiry abilities and creativity. The subjects were 36 five-year-old young children in B kindergarten located in S city. The subjects were divided into an experiment group of 18 and a comparison group of 18. The experimental group was taught by the early childhood science drama activities utilizing picture books for 6 weeks. The checking tool used the “scientific inquiry ability test” produced by Lee Kyung Min (2000) based on Martin(1997)’s scientific inquiry ability evaluation scale. and Creativity inspection based on the ‘Torrance Tests of Creative Torrence(TTCT)’ developed by Torrance(1990)‘s ‘inspection of Korean version TTCT (figure)’ was used. The study findings are as follows: First, pre- and post-test results indicated that the scientific drama activities had a positive influence on improving young children’s scientific inquiry abilities such as prediction, observation, classification and communication. Second, creativities including fluency, originality, sophistication and hasty termination resistance are improved in comparison with the contrast group activities. This study’s findings show that the scientific drama activities are educational values as effective activities to improve young children’s scientific inquiry abilities and creativity. This study is expected to provide basic data for the scientific drama activities education in early childhood education as a new attempt to improve the teaching and learning methods of 5-year-old children.
