분할운동 훈련에 따른 흰쥐 골격근내 TNF-α, IKKα/β, NFκB의 변화

The purpose of this study is to investigate the effect of split routine and prolonged endurance training on expression of TNFα, IKKα/β, NFκB protein in rat skeletal muscle. Male SD rat(n=18) were randomly placed in one of three exercise groups(CON, split routine(SE), prolonged exercise(PE)). The rat ran the treadmill running for the intensity of 1H at VO₂max 70% 5days/week during 8 weeks. The split routine endurance training composed as 30min running period and 6H of exercise interruption between each session. soleus and extensor digitorum longus muscle exercised after given exercise and western blotting analysis performed for the TNFα, IKKα/β, NFκB protein. We derived the conclusion that The expression of TNFα decreased in SE group however PE was not. IKKα/NFκB significantly increased in SE however PE decreased. This study can be concluded that split routine endurance exercise can be ameliorate inflammatory cytokine in skeletal muscle and promote the myogenic cytokines via the Ikk control signal.

The purpose of this study was to investigate the effect of hot and cold temperature on the expression of nitric oxide related signal factor in the rat skeletal muscle during acute exercise. Male Sprague-Dawley rats (n=60) were randomly assigned to one of three temperature groups (normal=20℃, hot=34℃, cold=4℃). The Rats ran the treadmill running for the intensity of 90min at 26M/min with slope degree 10°. Solues and extensor digitorum longus muscle were exercised during the given exercise(30, 60, 90min). The western blotting and ELISA analysis were performed for the expression of nitirc oxide(NO), eNOS, GC, and PKG protein. The NO expression was not significantly decreased in cold and hot temperature. However, eNOS, GC, and PKG protein were significantly decreased in cold and hot temperature. In this study, eNOS, GC, and PKG protein expressions were affected by cold and hot temperature. Cold and hot temperature can affect the vasodilation of vascular muscle cells through the changes in NO related vasodilation signal factors.

The purpose of this study was to determine to the effect of 6weeks training and vinegar drinks(SCFA) supplement on blood immune fuction response during exercise and recovery period in swimmer. We investigated the effect of vinegar drinks on serum GPR43, cellular immune cell and CK in human blood. For this, 24swimmer was recruited and control group(CON, N=8), Placebo group(PS, N=8), and vinegar group(VS, N=8). All group 6weeks training(HRmax 70~80%), and Blood collected pre-exercise, after-exercise, and 12H recovery period. The first results of the experiment showed the VS group significantly decreased serum CK and immune cell activation than CON and PS group, and second results of the experiment showed the VS group significantly increased GPR43. We derived the conclusion that vinegar drinks supplement on exercise and recovery period is effective on decrease inflammatory response. The more requirement experiment of variety exercise intensity, type and amount of Vinegar drinks supplement.
일회성 운동 및 회복기 흰쥐 골격근 내 HSP27과 관련된 Atypical MAPK 발현 경향

This study was to investigate the effects of Atypical MAPK signaling pathway expression on HSP27 expression which was small heat shock protein (HSP) related on damage and recovery of skeletal muscle. Generally, acute or high intensive exercise can elicit skeletal muscle injuries, cellular inflammatory response. Mitogen activated protein kinase (MAPK) highly involves exercise induced cell damage and recovery which have regulatory function on small HSP expression. MAPK is stimulated by stress and reative oxygen species induced by exercise. As a expect, MAPK may stimulates HSP27 which has protectional function of skeletal muscle cytoskeleton and is involved in protein synthesis and recovery. However exact regulatory mechanism between MAPK and HSP 27 not elucidated. To investigate the relation and regulatory function between Atypical MAPK and HSP27 during exercise and recovery, we did acute exercise and examined protein expression in gastrocnemius of ICR mouses by western blotting. Muscle harvest was various which divided by 5 time point (exercise: 15mins, 60mins / recovery: 6hours, 12hours), and control groups. As a result, the proteins expression of MAPK sub factors, PAK2, ERK3, and MK5, showed similar tendency to aspects of HSP27 revelation during exercise and recovery period. However, PAK1 was seen an inverse tendency different from other MAPK sub factors during exercise and recovery period. PAK1, in general, was seen a similar tendency to HEK293, PC12 cell, and had a same tendency to the result of preceding studies on the importance of signaling pathway to maintain the cell homeostasis. Thus, Atypical MAPK signaling pathway was important to prevent from skeletal muscle injuries caused by exercise stress.

The purpose of this study is to determine the expression of a acute endurance exercise both in cold(2℃) and hot(34℃) temperatures on the skeletal muscle. The main purpose of this study is to the effects of cold and hot temperatures to the heat shock protein 70, heat shock protein 27 and blood lactate, plasma CK using rats as the exercise. Adult male Sprague-Dawley rats were randomly placed in one of three temperature groups (normal, hot, cold). The treadmill run with an intensity of 26m/min, 10°intensity and soleus(SOL) and extensor digitorum longus(EDL) muscle extracted at 30, 60, 90 min during the exercise. The western blotting analyzed the expression of HSP70 and HSP27 during the exercise, the increased HSP27 protein in the SOL and EDL tissues of the cold, hot and normal temperatures were observed. However, it was observed that only SOL tissues increased HSP70 protein in both cold and hot temperatures. These results implied that increased apoptotic factor by cold and hot temperatures could cause the expression of HSP70 and HSP27 which involve cell damage in early stage of exercise and due to temperatures.
일회성 운동 및 회복기 흰쥐 골격근내 Autophagy factor Beclin1과 LC3의 발현 변화

Purpose : This study is to investigate the effect of acute endurance exercise and recovery on expression Autophagy factor Beclin1 and LC3 in rat skeletal muscle Method : The male Sprague-Dawley rats(n=65) were randomly placed in one of three groups(40% low intensity, 60% the middle intensity, 80% high intensity). The Rat in the exercised groups ran the treadmill running for the intensity 60min 7~26M/min with degree slope 10°. Soleus and Extensor digitorum longus muscle exercised 30,60min and during recovery 6, 12, 24h. The western blotting analysis performed for the expression of Beclin1 and LC3 protein. Results : The expression of Beclin1 in Soleus middle and high intensity than low intensity. And the middle intensity 12h, high intensity 6h significantly increased than other. The expression of Beclin1 in Extensor digitorum longus gradually increased in follow during recovery . The expression of LC3-Ⅰ in soleus during acute exercise gradually decreased in intensity, however LC-Ⅱ middle intensity during exercise 30min significantly increased the other group. The expression of LC3-Ⅰ in soleus during recovery different of group and intensity and LC3Ⅱ gradually increased time. The expression of LC3-Ⅰ in Extensor digitorum longus all group increased in during exercise 30min, and LC3-Ⅱ displayed similar type of middle and high intensity group. The expression of LC3-Ⅰ in Extensor digitorum longus during recovery gradually increased in follow during time, and LC3-Ⅱ significantly increased high intensity recovery 12h. Conclusion : This study can be concluded that acute exercise has positive effects on autophagy factor Beclin1 and LC3. this factor displayed different skeletal muscle type and intensity.
온도별 트레드밀 걷기운동에 따른 중년여성의 혈중 Nitric Oxide 및 혈압, 심박수의 변화

The purpose of this study was to investigate the effect of treadmill exercise in different temperature on changes of blood nitric oxide(BNO), blood pressure(BP) and heart rate(HR) in middle-aged women. Subjects were 7 women in the 50’s, treated by the different temperature of 3 groups, as normal temperature(23±1˚C), cold temperature(6±1˚C), hot temperature(32±1˚C) during treadmill exercise and effects on changes of BNO, BP and HR have been evaluated. For statistical significance, we use repeated measured ANOVA. Main results of the present study were as followings: 1)BNO did not show significance among normal, cold and hot group. 2) The effects of on elderly women’s systolic blood pressure(SBP) was a significant difference by exercise time(p〈.05). 3) Diastolic blood pressure(DBP) in all groups decreased at post-exercise more than pre-exercise and increased at recovery. Also, DBP was a significant difference by exercise time(p〈.05) and between groups(p〈.05). 4) HR was a significant difference by exercise time(p=〈.05) and between groups(p=〈.05), and the interaction was significant at the level of p=〈.05. To conclude, BNO contributes to blood endothelial cell relaxation, vasodilation and inhibition of pressor effect and it has a high relation with high blood pressure. These results may suggest that temperature has an influence on the BNO production and therefore leads to changes of BP and HR.
운동 강도별 일회성 운동에 따른 흰쥐 골격근 내 MK5(MAPKAPK5)와 관련된 신호체계 발현

The Aim of this study was to investigate the effect of acute endurance exercise on expression of MK5 related protein kinases in SD rat skeletal muscle. The male SD rat(N=20) was ran the treadmill running with various running intensity(VO₂max 40~80%) and soleus and extensor digitorum longus muscle was harvested for analysis. Western blooting was performed for the expression of HSP27 and p38 MAPK phosphorylation, ERK3 and MK5. The skeletal muscle’s cAMP changes was analysed by ELISA. The cAMP level was not significantly increased by exercise intensity increase however the MK5 and HSP27 expression level was significantly increased. Also p38 MAPK level was significantly increased in solues but EDL was not. importantly, novel MK5 activator, ERK3 was significantly increased coinside with the MK5 and HSP27 expression. As results, We derived the conclusion that ERK3 is a novel MK5 activator in skeletal muscle when exercise. However, a downside of these experiments in that the reason and process for direct evidence for ERK3 activation is still unknown.
