RAW 264.7 대식세포에서 LPS로 유도된 NF-κB signaling pathway 내 Causonis japonica (Thunb.) Raf. 추출물의 항염증 효능 평가

한국생명공학연구원으로부터 분양받은 해외생물소재의 항염증 활성을 탐색 후 항염증 효능이 탁월한 CJE를 시료로 선별하였다. CJE는 세포 독성이 나타나지 않는 농도에서 LPS에 의한 iNOS의 발현과 NO의 생성을 유의적으로 억제하였으나, COX-2의 발현에는 영향을 미치지 못했다. CJE는 LPS에 의해 활성화된 IKK, p65 및 MAPKs (p38, ERK, JNK)의 인산화를 억제하였으며, 세포질에서 핵으로의 p65 전위를 억제하였다. 연구 결과를 통해 in vitro 내 CJE가 LPS 유도 iNOS/NO 생성과 NF-κB 및 MAPKs 신호전달경로의 예방을 통해 잠재적인 항염증 효능을 보였으며, 해당 결과를 바탕으로 CJE를 활용한 건강기능식품 개발을 위해서는 추가적인 지표성분의 동정 및 정량화 연구가 필요할 것으로 판단된다. We evaluated the anti-inflammatory effects of Causonis japonica (Thunb.) Raf. extract (CJE) in mouse RAW264.7 macrophage cells. The CJE significantly suppressed lipopolysaccharide (LPS)-induced inducible nitric oxide synthase (iNOS)/nitric oxide (NO) production. It also suppressed LPS-induced phosphorylation of IkB kinase (IKK), p65, the translocation of p65 from the cytosol to the nucleus, and the phosphorylation of mitogen-activated protein kinases (MAPKs). Overall, our results suggested that the CJE suppresses LPS-induced inflammatory responses by regulating the NF-κB and MAPK signaling pathways.

우리는 한국생명공학연구원에서 제공한 소재를 탐색 후 ACE를 선별하였고 이 소재의 항염증 효능을 평가하였다. ACE는 세포독성이 없는 농도에서 LPS에 의한 iNOS의 발현과 NO의 생성을 유의적으로 억제하였다. ACE는 LPS에 의해 증가한 IKK, IκB, p65의 인산화와 IL-1β의 생성을 유의적으로 억제하였다. 이 논문은 ACE가 NO/iNOS 발현 및 NF-κB 신호 전달 경로의 억제를 통해 항염증 효능을 나타냄을 밝힌 최초의 논문으로서 의의를 가진다. 또한 이 결과를 바탕으로 ACE는 항염증 기능성식품 또는 의약품소재로써 활용가치가 높을 것으로 기대된다. Argyreia capitata (Vahl) Choisy is a species of the genus Argyreia. Although many studies have analyzed the biological activity of A. nervosa, A. speciasa, and A. acuta, the anti-inflammatory effect of A. capitata extract (ACE) remains unclear. In this study, we evaluated the anti-inflammatory effect of ACE using lipopolysaccharide (LPS)-induced inflammatory markers in RAW 264.7 cells. We confirmed that the ACE inhibited the LPS-induced NO (nitric oxide) and iNOS (inducible nitric oxide synthase) expression in RAW 264.7 cells. ACE suppressed not only the LPS-induced phosphorylation of IKK, IκB, and p65 but also IL-1β expression. Collectively, these results suggest that ACE is a novel anti-inflammatory agent that suppresses iNOS expression, NO production, and the NF-κB signaling pathways.

To suggest the native plants combination for restoration of the dissected sloping area, the plant communities of thirty seven quadrats from five localities of mid-southern part of Korean peninsula were surveyed from 2007 to 2008. Their flora, vegetation structure, and dominant species based on the importance value were investigated. And the soil characters (pH, moisture content, water holding capacity, and organic matter) of each dominant species were analyzed. Also, germination tests were performed to check the stability of restored native plants using seed chips. As a result, 79 native plants were suggested for the dissected sloping area: 20 trees and subtrees for the upper layer, 18 shrubs and vines for the middle layer, and 41 herbs for the lower layer, taken together their flora, dominant species, vegetation structure, soil condition, germination test, flowering period.
