Cervical cancer is one of the leading causes of female death from cancer worldwide with about 500,000 deaths per year. A strong association between certain human papilloma viruses (HPV types 16 and 18) and cervical cancer has been well known. An extract of natural products, named as Somatid, has been used to investigate whether this agent has the ability of inhibiting the oncogenes E6 and E7 of HPV type 16. This Somatid inhibited the proliferation of human cervical cancer cell lines (C-33A, SiHa, CaSki) and HaCaT keratinocytes in a dose response manner. In vitro binding assay and ELISA showed that Somatid inhibited the in vitro biding of E6 and E6AP which are essential for the binding and degradation of the tumor suppressor p53. In addition, Somatid inhibited the in vitro binding of E7 and Rb which is essential tumor suppressor for the control of cell cycle. The levels of mRNA for E6 and E7 were also decreased by Somatid. Our data suggested that Somatid inhibited the oncogenecity of E6 and E7 of HPV 16 type, thus can be used as a putative anti-HPV agent for the treatment of cervical carcinomas caused by HPV.
산화적 스트레스에 의한 N'-methyl-N'-nitroguanidine의 유전독성증가

To investigate the possible enhancement of genotoxicity in stress environment, we examined the effect of genotoxic material in oxidative stress-induced condition using human cell line. Human lymphoblast cell line, TK6 was treated with hydrogen peroxide (H₂O₂) for induction of oxidative stress, and treated with N’-methyl-N’-nitroguanidine (MNNG), as a genetoxic material. We carried out MTS assay to set treatment doses. TK6 was treated with H₂O₂ as 6.75 (low dose) or 13.5 μM (high dose) for 2 h, and treated with MNNG as 0.117 (low dose), 0.234 (middle dose), 0.468 μM (high dose) for 2 h. As results, a treatment of MNNG induced DNA damage as dose dependently. And TK6 treated with H₂O₂ at low as well as high dose followed by MNNG treatment showed higher DNA damage compared to MNNG alone treated groups. Malondialdehyde, as a marker of lipid peroxidation was increased in H₂O₂ and MNNG treated groups. Real-time RT-PCR analyses for expression sion of several antioxidative enzymes showed that catalase mRNA and glutathione peroxidase 1 mRNA expression were decreased in H₂O₂ and MNNG treated groups. Taken together, we conclude that genotoxicity induced by MNNG is enhanced in a condition of oxidative stress induced by H₂O₂ and it suggests that it should be associated with induction of lipid peroxidation and decrease of antioxidant enzymes.

The purpose of this study is to suggest basic information and valuable methods for effectively managing the family stress. Based on a variety of literatures relating to the family stress, this study was carried out by examining the concepts, theories and solutions to problems of the family stress, and by suggesting the system framework necessary to manage the family stress. The summarized result of this study is as follows. First, stressor implies the event and demend resulting in stress. The family stress is a physical, emotional and psychological response of a person or family to these stressors. Second, integration and adaptability in stress management are resources of the famiily system that interact with social support systems to promote adaptation and balance in family life. Personal resources are particulary useful in coping with stress, including health and cognitive resources that facilitate realistic stress perception and problem-solving skills. Third, one goal of coping with stress is to produce a balance between the integration and organization of a family group and the growth and development of its individual members. Fourth, the system factor to manage the family stress includes input, throughput, output and feedback. The input in managing stress implies demands and resources. Therefore, stress tends to be caused by the demand beyond resources. And alse, the output is obtained by the systematic activities of the throughput making the family coping possible. Then, there is occurred the feedback in all the system, where useful strategies are again input into the given system.
