        항노화 화장품 소재로서 대추 추출물에 관한 연구

        This study was to investigate characterization as cosmetic substances of extracts of Jujube using water with in vitro anti-aging and antioxidant activity. Generally, antioxidant activities are dependent on their phenolic constituents. We found that the various jujube extracts dose-dependently increased in vitro antioxidant activities(DPPH radical, ABTS+ radical and Superoxide anion radical scavenging activity). DPPH, ABTS+ and Superoxide anion radical scavenging activity of jujube ethylacetate fraction at concentration of 1,000 μg/mL were 84.5%, 78.5% and 54.9% respectively. In vitro elastase and collagenase inhibition effects of NN-L at concentration of 1,000 μg/mL was 42.8% and 55.3% respectively.All the tested extracts DPPH and ABTS free radical scavenging activity was depend on their concentration. Especially the ethylacetate fractions presented higher inhibitory effect on elastase (65.2% at 10000 μg/mL). so that it showed in vitro anti-wrinkle activity. Finally, the jujube ethyl acetate fractions showed Astringent activity. These results suggest that the jujube extracts can be used for cosmetic industry.

        찔레나무뿌리(Rosa multiflora root)의 항산화 및 항염증효과

        찔레나무뿌리를 열수, 에탄올, 메탄올, 아세톤에서 추출하여 항산화 및 항염증 효능효과를 확인하였다. 그 결과 아세톤추출물에서 가장 많은 67.05±0.56 ㎎/g의 폴리페놀 함량을 확인하였으며, SOD 유사활성능을 제외한 DPPH, ABTS, superoxide anion 라디컬소거능 확인 결과 우수한 항산화 효과를 확인하였다. 찔레나무뿌리 추출물의 HAase 저해능을 측정한 결과 에탄올, 메탄올, 아세톤 추출물 500 ㎍/㎖ 에서 60% 이상의 높은 HAase 저해 효과를 확인할 수 있었다. NO 생성량을 확인한 결과, RAW 264.7 cell에서 LPS에 의해 유도된 NO 생성을 농도 의존적으로 뚜렷하게 감소시키는 것을 확인하였으며, 에탄올 추출물에서 NO 생성억제 효과가 가장 우수한 것을 확인할 수 있었다. 위의 사실에 기초하여 NO 생성저해의 기전을 알아보기 위하여 iNOS의 발현을 분석한 결과, 에탄올추출물 100 ㎍/㎖에서 40%의 iNOS protein 발현 감소를 확인하였으며, iNOS의 발현억제가 NO생성억제와 유사한 경향을 나타냄으로 NO생성억제는 iNOS의 발현저해를 경유한 것임을 확인할 수 있었다. 이러한 결과들은 찔레나무뿌리 추출물이 항산화 및 항염증 연구의 기초 자료로 활용될 것으로 예상된다. 또한 추후 산업적 응용도 가능함으로 기능성화장품의 가능성을 제시하고 있다. Rosa multiflora thunberg belonging to Rosaceae is widely distributed in East Asia including Korea and Japan, and has been reported to have tormentic acid and rosamultin. To develop a new natural anti-inflammatory agent for cosmetics, we investigated the inhibitory effects of inflammation in Rosa multiflora root (R. multiflora root). The biological activity and anti-inflammatory effects were investigated by water, ethanol, methanol and acetone extracts of R. multiflora root. The measurements of polyphenol content from R. multiflora root were highest in water and acetone extracts, at 57.48 ± 0.88 ㎎/g and 67.05 ± 0.56 ㎎/g, respectively. The result of DPPH, ABTS and superoxide anion radical scavenging effects showed over 50% efficacy at 50 ㎍/㎖ in ethanol, methanol and acetone extracts. Hyaluronidase inhibition effect showed over 60% efficacy at 500 ㎍/㎖ in ethanol, methanol, and acetone extracts. Nitric oxide radical inhibition effect of R. multiflora root ethanol extracts showed over 30% efficacy at 500 ㎍/㎖. We investigated the effect of R. multiflora root extracts on nitric oxide (NO) production of inducible nitric oxide synthase (iNOS) in LPS-induced RAW 264.7 macrophage cells. The result showed that R. multiflora root extracts have an inhibitory effect on NO production and iNOS expression and also can be used as an anti-inflammatory agent. These antioxidant and anti-inflammatory effects of R. multiflora root show applicant potential application as a functional cosmetic material.

        LPS로 자극된 Raw 264.7 대식세포에서 오미자 씨앗오일의 항염증 효과

        Objectives : This study was designed to investigate of the anti-inflammatory effects of Schisandra chinensis seed oil(SSO) on the production of pro-inflammatory substances in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. Methods : SSO was measured the production of pro-inflammatory factor (NO, PGE2, IL-1β iNOS and, COX-2) in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. we used the following methods : cell viability assay, Griess reagent assay, enzyme-linked immunosorbent assay, Western blotting analysis. Results : The cell viability of SSO(0∼500 μl/mL) processing group was 96.9% and the processing of SSO didn``t have an effect on the cytotoxicity. The inhibitory effect of the nitric oxide (no) production of SSO(500 μg/mL, 50 μg/mL, 10 μg/mL) was each 70.3%, 37.6% and 26.5%. IL-1β production inhibition ability of SSO(500 μg/mL, 100 μg/mL) was each 49.88% and 48.8%. PGE2 production inhibition ability of SSO(500 μg/mL, 100 μg/mL) was each 49.88% and 73.1%, 70.5%. By using SSO, it experimented about iNOS protein expression inhibition ability, that is the NO production enzyme. iNOS protein expression increased in the group processing LPS independently. iNOS protein expression decreased in the group processing SSO together. The expression of the COX-2 protein decreased 89.6%, 81.8% in the group processing SSO. The significance was in the relationship with NO formation inhibition with the relationship with the PGE2 formation inhibition and iNOS protein, it confirmed in SSO with the COX-2 protein. Conclusions : Stimulation of the RAW 264.7 cells with LPS caused an elevated production of nitric oxide (NO), IL-1β and PGE2 which was markedly inhibited by the pretreatment with SSO without causing any cytotoxic effects. The reduced expressions of iNOS protein were consistent with the reductions in NO production in the culture media. SSO may be useful for the treatment of various inflammatory diseases.

        하엽(荷葉) 추출물이 항산화 효능 및 피부노화에 미치는 영향

        Objective: The purpose of this study was to investigate anti-aging and antioxidant effects of extracts of Nelumbo nucifera leaves (NN-L) using ethanol on skin . Methods : Each part of leaves(NN-L), flowers(NN-F) and stem(NN-S) was extracted with 70% ethanol. We performed radical scavenging assay(DPPH, ABTS+, Superoxide anion radical), elastase inhibition assay, collagenase inhibition assay. NN-L extracts were tested for cell viability(MTT assay), MMP-1 inhibition and MMP-1 protein expression on CCD-986sk cells (human fibroblast line). Results : Recently, many studies have reported that elastin is also involved in inhibiting or repairing wrinkle formation, although collagen is a major factor in the skin wrinkle formation. We measured its free radical scavenging activity, elastase inhibitory activity and expression of MMP-1 (matrix metalloprotease-1) in human fibroblast cells. Among the parts of Nelumbo nucifera , NN-L showed the highest antioxidant activities and in radical scavenging. DPPH, ABTS+ and Superoxide anion radical scavenging activity of NN-L at concentration of 1,000 μg/mL were 91.43%, 99.31% and 73.7% respectively. In vitro elastase and collagenase inhibition effects of NN-L at concentration of 1,000 μg/mL was 42.8% and 55.3% respectively. The ethanol extract of NN-L showed cell viability of 95.4% in 50 μg/mL concentration. In addition, The results from Western blot assay showed that NN-L decreased the expression of MMP-1 protein in a dose-dependent manner (by up to 35.0% at 50 μM). Conclusion : The findings suggest that the NN-L great potential as a cosmeceutical ingredient with antioxidant and anti-wrinkling effects.

        LPS로 자극된 Raw 264.7 대식세포에서 한약재복합 추출물의 세포독성 및 항염증 효과

        Amur Corktree, Paeonia lactiflora Pallas, Dictamni Radicis Cortex, nemarrhena asphodeloides Bunge, Inula helenium L have been used as a traditional medicine to treat inflammatory diseases in Korea. This study was designed to investigate the effects of Mixed Extracts from Korean Medicinal Herbs that inhibits lipopolysaccharide(LPS)-induced inflammatory responses in Raw 264.7 macrophages. Raw 264.7 macrophages were treated with various concentrations of the Medicinal Herbs complex (APDAL : Amur Corktree, Paeonia lactiflora Pallas, Dictamni Radicis Cortex, nemarrhena asphodeloides Bunge, Inula helenium L.). Nitric oxide PGE2, IL-6 inhibition effect of APDAL ethanol extracts was above 50% at a 50 μg/mL concentration. The inhibitory effects against the expression of inducible NO syntase (iNOS) and cyclooxygenase-2 (COX-2) were 65.3%, 42.3% in APDAL ethanol extracts at 50 μg/mL concentration. APDAL ethanol extracts inhibited inducible nitric oxide synthase (iNOS), COX-2 protein level on the LPS-indcued Raw 264.7 cells. In order to evaluate the effect of APDAL ethanol extracts on a proliferation of RAW 264.7 cells, the MTT assay was carried out. results showed that APDAL ethanol extracts has not cytotoxic at 50 μg/mL concentration. Thus, the anti-inflammatory reactions by APDAL ethanol extracts were not affected by the cytotoxic effect. The results suggest the usefulness of developing functional materials using anti inflammatory.

        망개떡의 저장성 향상을 위한 청미래덩굴 뿌리 추출물의 첨가

        The antimicrobial activities of Smilax china L. against spoilage bacteria isolated from Mang-gae rice cake were investigated and the storage stability of the Mang-gae rice cake was enhanced. Spoilage bacteria, which cause Mang-gae rice cake to rot, were isolated from commercial Mang-gae rice cake, and most of the isolated strains were identified as Bacillus sp. After the leaves, roots, shoots, and stalks of the Smilax china L. were extracted using 50% ethanol as the solvent, their antimicrobial activities were investigated using the paper disc method by treating them with 50 μL of Bacillus cereus, which is known as a major pathogenic micro-organism in foods that contain starch, as the test organism. The antimicrobial activities of the extracts were compared according to the size of the clear zones around the paper discs. The root extract showed significant antimicrobial activities. When red beans, which are used as stuffing for Mang-gae rice cake, were treated with the root extract of the Smilax china L., the viable cell count of the Mang-gae rice cake was 5.04 Log CFU/g after 48-hr storage, and the cake showed significantly slower growth of bacteria than with commercial products. These results show that treatment of red beans with Smilax china root extract could improve the storage stability of Mang-gae rice cake.
