Depression is a psychiatric syndrom consisting of dejected mood, psychomotor retardation, insomnia and weight loss, sometimes associated with irrational guilt feeling And it is also similiar to Hwa-byung(火病) symptom in oriental medicine. But it is difficult to diagnose with objective method. Digital Infrared Thermographic Imaging(D.I.T.I) is one of diagnotic method that measure the changes of skin temperature in body. Specially we checked the skin temperature on depressed patients by using D.I.T.I and compared with Bell`s palsy patients and normal persons group. The results are as follows; Average body temperature of the depressed patient group is 36.68±0.43˚C and that of the control group is 36.73±0.40˚C. So there is no meaningful difference. The depressed patient group has higher temperature than the control group by △T〉1.0˚C at the following acupuncture points in these body parts - upper and lower, left and right, anterior and posterior. When acupuncture points temperature was compared superior and inferior part of the body, depressed patient group have meaningful difference at the GV-4(Myung-moon) and also in the control group. When acupuncture points temperature was compared left and right part of the body, depressed patient group have no meaningful difference and also in the control group. When acupuncture points temperature was compared in the anterior and posterior part of the body, depressed patient group have meaningful difference at the GV4(Myung-moon) and also in the control group. From this study, we think that D.I.T.I could be used to diagnose objectively on the depressed patients and useful to another psychoneurogenic diagnosis in oriental medicine.

In previous studies, heat shock has been reported to induce the apoptosis or programmed cell death through the activation of caspase-3. 1 investigated the effect of juniper pure essential oil on the heat shock-induced apoptosis in human astrocyte cell line CCF-STTGI. Treatment of the astrocytes with heat shock markedly induced apoptotic cell death. However, pretreatment of the astrocytes with juniper oil ingibited the heat shock-induced apoptosis. To determine whether juniper inhibits the heat shock-induced activation of these apoptotic proteases, activation of CPP32 was assessed by Western blotting. Consistent with flow cytometry. DNA fragmentation and giemsa staining, heat shock-induced activation of CPP32 was blocked by juniper oil. Poly(ADP-ribose) polymerase (PARP), cysteine protease substrates were fragmented as a consequence of apoptosis by heat shock. Juniper oil inhibited the PARP fragmentation. This juniper oil also inhibited the heat shock-induced activation of caspase-3. These results suggest that juniper oil may modulate the apoptosis through the activation of the interleukin-1-converting enzyme-like protease.

Objectives: The oriental medicine Jangwonhwan originally described in the Korean medical text, DonguiBogam(amnesia chapter). Recently, a modified formula of Jangwonhwan(LMK02-Jangwonhwan), was shown to reduce β-amyloid deposition in the brain of Tg-APPswe/PS1dE9 mouse model for Alzheimer`s disease. This experiment aimed to investigate the acute oral toxicity of LMK02 in SD rats by up-and-down procedure determinations. Methods: Quality control of tablet form of LMK02 was established by estimating indicative components, Ginsenoside Rg3 of Red Ginseng and Decursin of Angelicagigas Nakai. The toxicity of LMK02 was investigated in 6 week old, specific pathogen free(SPF), Sprageu-Dawley rats. 3 female rats received 5,000 mg/10 ml/kg of test substance and their death rate, clinical sings, weight changes and autopsy findings had been observed for 2 weeks. Results: Any specific symptoms or death were resulted in this experiment. No significant changes in rats` weight. No significant differences in atopsy. Conclusions: The minimum lethal dose(MLD) of LMK02 for female Sprauge-Dawley rats were more than 5,000mg/kg in this experiment.

We investigated the effects of lemon pure essential oils on the heat shock-induced apoptosis in human astrocyte cell line CCF-STTGI. In previous studies, heat shock has been reported to induce the apoptosis or programmed cell death through the activation of caspase-3. Treatment of CCF-STTGI cells with heat shock markedly induced apoptotic cell death as determined by flow cytometry. Interestingly, pretreatment of CCF-STTGI cells with lemon pure essential oils inhibited the heat shock-induced apoptosis. Lemon also inhibited the heat shock-induced apoptosis in primary cultured rat astrocytes. To determine whether lemon inhibits the heat shock-induced activation of these apoptotic proteases, activation of CPP32 was assessed by Western blotting. Consistent with flow cytometry. DNA fragmentation and giemsa staining, heat shock-induced activation of CPP32 was blocked by lemon pure essential oil. PARP, cysteine protease substrates were fragmented as a consequence of apoptosis by heat shock. Lemon oil inhibited the PARP fragmentation. This essential oil also inhibited the heat shock-induced activation of caspase-3. These results suggest that lemon pure essential oils may modulate the apoptosis through the activation of the ICE-like caspases.

To study the effects of Ramulus et Uncus Uncariae (REUU) on oxygen free radical-mediated damage by hydrogen peroxide (H-2O-2) on cultured spinal sensory neurons, in vitro assays such as MTT assay, NR assay, neurofilament enzymeimmuno assay (EIA), sulforhodamine B (SRB) assay, assay for lactate dehydrogenase (LDH) activity and assay for lipid peroxidation were used in cultured spinal dorsal root ganglion neurons derived from mice. Spinal dorsal root ganglion neurons were cultured in media containing various concentrations of H-2O-2 for 5 hours, after which the neurotoxic effect of H-2O-2 was measured by in vitro assay. The protective effect of the herb extract, Ramulus et Uncus Uncariae (REUU) against H2O2-induced neurotoxicity was also examined. The results are as follows. 1. In NR assay and MTT assay, H-2O-2 sigruficantly decreased the cell viability of cultured mouse spinal dorsal root ganglion neurons according to exposure concentration in these cultures. An additional time course study was done on these cultures. 2. Cultured spinal dorsal root ganglion neurons which were exposed to various concentrations of H-2O-2 showed a quantitative decrease of neuronal cells by EIA and of total protein by sulforhodamine B (SRB) assay, while they showed an increase of both lipid peroxidation and LDH activity. 3. The effect of Ramulus et Uncus Uncariae (REUU) on H-2O-2 induced neurotoxicity showed a quantitative increase in both neurofilament and total protein, but showed a decrease of lipid peroxidation and LDH activity. These results suggest that H-2O-2 has a neurotoxic effect on cultured spinal dorsal root ganglion neurons from mice and that the herb extract, Ramulus et Uncus Uncariae (REUU), was very effective in protecting H-2O-2 induced neurotoxicity by decreasing lipid peroxidation and LDH activity. “This work was supported by the Brain Korea 21 Project.”

Objectives: By analyzing and comparing indications, components and their doses of herb medicines, which were recorded in insomnia chapters of medical books, we examined historical changes and made clear documentary ground of medications. Methods: 1. 10 books which were noted in insomnia part of 『The Eastern Medical Textbook of neuropsychiatry』 and 13 books were mentioned in same part of 『Uibujeonrok』 were selected as reference. 2. We searched article database such as KISS, riss4U and journals issued by the Korean society of oriental neuropsychiatry. Finally, 39 books which were noted in 3 literature reviews, were chose as reference. 3. In reference books, we collected the herb medicines in chapters related to insomnia. 4. In case of no section associated with insomnia in books, we found prescriptions related to insomnia in whole chapters. 5. If the chief virtue of medication was not primarily for insomnia, we excluded applied or added herb medicines. 6. We classified prescriptions into small groups which had same name, and then rearranged them in chronological order. Results and Conclusions: 1. Banha-tang( banxiatang) of 『Hwangjenaegyeong·Youngchu』 was the first-recorded prescription used for insomnia. 2. The chapter of insomnia appeared in 『Chunkeumyobang』 at first. 3. Ondam-tang(Wendantang) was most cited prescription in medical books. It was recorded totally 19 books from 『Chunkeumyobang』 to 『Hyeoljeungron』. 4. We suspects indications, components, dosage of medication of insomnia in Donguibogam were revised according to author`s clinical experience and circumstances.

Substance P(SP) can stimulate production of tumor necrosis factor-α(TNF-α) from astrocytes stimulated with lipopolysaccharide(LPS). The objective of the current study was to determine the effect of Taraxacum officinale(TO) on the production of TNF-α from primary cultures of rat astrocytes. TO(100 & 1000㎍/ml) significantly inhibited the TNF-α production by astrocytes stimulated with LPS and SP. Interleukin-1(IL-1) has been shown to elevate TNF-α production from LPS-stimulated astrocytes while having no effect on astrocytes in the absence of LPS. We therefore examined whether IL-1 mediated inhibition of TNF-α production from primary astrocytes by TO. Treatment of TO(100 and 1000㎍/ml) to astrocytes stimulated with both LPS and substance P decreased IL-1 production significantly. Moreover, the production of TNF-α by LPS and substance P in astrocytes was progressively inhibited with increasing amount of IL-1 neutralizing antibody. These results suggest that TO may inhibit TNF-α production by inhibiting IL-1 production and that TO has an antiinflammatory activity in the central nervous system.
