        백색부후균에 의한 침출수처리 공정의 최적화

        In this study a process for the leachate treatment has been optimized. A packed column reactor system was used for the treatment of leachate. The cells of white rot fungi are immobilized on the glass beads in the reactor. The cells of Phanerochaete chrysosporium could grow up in a leachate solution without additional substances and produce extracelluar enzymes. The decrease of COD value and ammonia concentration relied on the cell growth. After 7 days COD, BOD and ammonia were removed 30%, 82% and 67%, respectively. After reaction kinetics there is a suggestion that a leachate with high content of ammonia can be well adaptable for microbial treatment. In the reactor most of ammonia were degraded by microbial cells and higher enzyme concentration could increase removal rate. For a higher efficiency a packed column system with microbial cells was tested. In the packed column reactor immobilized with the white rot fungi cells more than 80% of BOD and almost of ammonia were removed, but COD under 30%. The glass beads may not the best carrier for fungi cells or leachate treatment. A further study will be necessary about variable carriers for the optimization of the system.

        김치 유래 유산균의 항균적 특성과 이용

        Earlier in Korea Kimchi was made in every family and every province has own taste and specialties. These days almost of the Kimchis are manufactured. We collected variable Kimchis, which were made for private use and isolated microorganisms. Some interesting micobial cells were identified and studied for its application as food and drinks. One of them was identified as Lactobacillus sakei KJ123. This strain is known as producing interesting aromatic components during Sakei fermentation like Kimchi in variable conditions. We tried to develop a health beverage with fermentation process. The Cucurbita maxima has been known as a traditional healthy food and variable positive effects on the human body were already reported. In this study we tried to develop a production process for a healthy fermented drink on this substrate with strains originated from Kimchi. Many kinds of lacctobacilli species existed in the fermented food cannot survive in the acidic conditions like human stomach. So we selected resisting strains in this conditions. The survival rate of Lactobacillus sakei cells in the artificial gastric juice and bile acid and other physiological characteristics at the variable conditions have been tested. After fermentation process some sensory tests on the product with panels were tried.

        단백질 분해효소 생산 균주 분리

        영양 성분을 함유하고 있는 유기성 폐기물은 미생물에 의해 처리되어, 유용한 물질로 전환될 수 있다. 이러한 생물학적 공정에서 미생물 세포와 효소는 원료 물질인 기질과 함께 중요하다. 대규모화 공정에서도 미생물 세포와 효소는 공정 최적화에서 필수적인 요소이다. 본 연구에서는 이러한 생물학적 공정의 효율성을 높이는 목적으로 다량의 아미노산과 단백질을 함유하고 있는 많은 종류의 부패가 진전된 유기성 폐기물과 발효 식품에서 단백질 분해효소를 생산하는 미생물을 분리하였다. 단백질 분해 효소의 활성, 온도와 산도등 활성 조건과 활성 정도를 확인하여 선택된 균주들을 동정하였다. 산업적으로 저온에서 단백질을 분해하는 효소는 유기성 폐기물을 저온에서 처리할 수 있다. 저온에서 처리가 가능하다는 것은 폐기물의 처리온도를 낮은 상태로 유지할 수 있어 그 만큼의 열(steam)비용을 줄일 수 있다. 또한 이 단백질 분해효소를 이용하여 단백질을 분해 후 다량의 아미노산을 생산할 수 있으므로 아미노산 생산 공정에도 적용이 가능하다. 이렇게 유기 폐기물을 처리하여 다양한 용도로 사용할 수 있으므로, 폐기물의 가치를 높일 수 있다. 다양한 활성 조건에서 단백질 분해효소를 다량으로 생산하는 균주를 분리하여 동정하고, 균주 배양 조건, 효소 생산의 최적 조건에 대한 연구를 수행하였다. Protease producing microorganisms were isolated from many kinds of food waste and fermented foods, which contains high amount and variable kinds of degraded substances. Several microorganisms were identified by 16S rRNA full sequencing analysis methods. The activity of protease was analyzed and identified in variable conditions for the application. For industrial use for bio waste treatment some proteases were isolated, identified and selected from microbial cells. And the tests were carried for the further use. The protein degrading activity at low temperature is useful for the treatment of organic waste, which contains much proteins. By the protein degradation process the organic waste can be utilized in variable fields, for example from feedstuff supplement to fertilizer for agriculture. Bacterial cells with protease activity at low temperature were isolated and identified. The optimal conditions for microbial cultivation and protease production were studied.

        기술논문(技術論文) : 옥상정원 활성화에 유용한 토질영양제에 대한 연구

        Recently the roof gardening has become very popular and even on the apartment-veranda and on the roofs in high buildings many kinds of vegetables and berries are cultivated. And the demand of the nutritional supplement for the effective plant growth is also increasing. The general urban conditions are to be adapted for plant growth. A different hygroscopic and temperature-conditions after regions, sunshine and wind have strong influences on the plant growth and usually it is not optimal enough. It is because why a nutritional condition in soil for plant growth so important and essential. The usual compound-soil or -fertilizer cannot offer enough quantities of nutrients for plant growth and additional soil conditioner becomes more necessary. There are many kinds of soil conditioners like hydrogel in the market and we studied on Geohumus, Montigel and Geko, which are widely used in Europe and other countries. Water absorption and microbial immobilization with effective microorganisms were tested and compared. The EM solution was identified as bacteria, fungi and azotobacter etc. and they were immobilized at the soil conditioners at first. And the cultivated and immobilized at the soil conditioners EM-solution was added to the plant soil. 1 g of the soil conditioners absorbed ca. 20 g of water. The plant grew 10 cm more, got 3 times more branches and 2 times more fruits in the soil with soil conditioners immobilized with microorganisms. With water addition the plant with both conditions in the soil could stay fresh in comparison to without soil conditioners.

        두충나무, 황벽나무 등을 포함하는 수목추출물의 항여드름 및 항아토피 효과

        본 연구에서는 두충나무, 황벽나무 등을 포함하는 수목추출물의 소재특성을 시험하여 화장품 및 피부질환 치료소재로서의 응용 가능성을 조사하였다. 수목추출물의 항산화력은 DPPH 자유라디칼 소거법으로 측정되었으며 저농도에서는 항산화력이 낮았으나, 1,000 ug/ml의 고농도에서는 우수한 항산화제인 비타민 C와 유사하였다. 항균력은 원판확산법으로 측정되었으며, 수목추출물은 피부상재균인 Staphylococcus epidermidis에 대해서는 낮은 항균력을 보이지만, 여드름 유발균인 Propionibacterium acne에 대해서는 높은 항균력을 보여주는 선택성 항균력이 있어, 여드름소재로서 매우 우수한 특성을 나타내었다. 무모생쥐를 이용한 항아토피 시험결과, 수목추출물은 DNCB에 의해 유도된 피부염증을 14일 째 거의 정상상태로 회복시켰으며, 피부내 항체인 IgE의 농도도 대조군에 비해 16% 낮추어주어 항아토피 효과가 우수한 것으로 나타났다. 따라서 수목추출물은 항여드름효과 및 항아토피 효과가 뛰어나서 화장품 혹은 피부질환치료소재로서 응용 가능성이 매우 높음을 알 수 있었다. In this study, plant extracts including Eucommia ulmoides Oliv, and Phellodendron amurense were studied to test possible application for cosmetics and skin related medicine. Anti-oxidation effect of plant extracts was measured by DPPH free radical scavenging activity and it was insignificant at low concentration, however, it was as good as vitamin C, excellent anti-oxidation agent, at 1,000 ug/ml. Anti-bacterial effect was tested by disc diffusion method, and plant extracts showed mild anti-bacterial effect for normal skin flora, Staphylococcus epidermidis while it indicated strong anti-bacterial effect for acne inducing Propionibacterium acne. Therefore it had powerful potential for anti-acne material because of selectivity. Anti-atopic dermatitis effect was tested by hairless mouse and plant extracts recovered damaged skin to near normal condition after 14 days of treatment. IgE concentration in treated mouse was decreased 16% compared with control. From the research, plant extracts indicated strong anti-acne and anti-atopic dermatitis effect, and showed strong potential for cosmetics and skin related medicine.
