        The conclusion from experiments in the characteristic of yeast strain for selecting the appropriate yeast strain according to the variation of wort plato was as follows ; 1.CB8l, CB175 and CB89 yeast strain showed 73.3%, 72.6% and 73.0% in its limit fermentation, respectively. CB89 yeast strain showed the good result of 46.3% in the fermentation power test, also CB8l and CB175 yeast strains showed 33.1% and 24.0%, respectively. 2.In the yeast cell size CB8l, CB175 and CB89 yeast strain showed 8.4㎛, 8.8㎛ and 8.0㎛, respectively, and in the flocculation test those sedimentation content showed 0.6ml, 0.4ml and 8.5 ml, respectively after 10 minutes. CB89 yeast strain with high flocculation showed on appropriate result for brewing yeast. 3.Measured reducing sugar of each yeast after a day were observed as follows; CB8l, CB175 and CB89 yeast stain used up 33.3%, 42.6%, 61.1% of reducing sugar , respectively. 4.In ending fermentation, V. D. K(Vicinal Diketones) content and suspension yeast cell number of CB8l yeast strain showed 0.51-0.65ppm and 20-32×106cells/ml, respectively. Those of CB175 yeast strain showed 0.42-0.48ppm and 12-20×106cells/ml, respectively. CB89 yeast strain showed 0.58-0.69ppm and 34×106 cells/ml. 5.In ending fermentation, alcohol content of CB89 yeast was produced 4.55% and 5.05% was higher than that of CB8l and CB175 yeast. 6.CB89 yeast strain also showed about 300% of yeast recovery rate and 5.2% of dead cell rate which were good results.

        채소연부병균 Erwinia herbicola의 생육억제균 분리 및 특성

        Erwinia herbicola(EH) 채소 연부병 방제를 위하여 충북 청원군, 진천군 및 충남 연기군 일원의 채소 경작지 토양으로부터 형광성을 나타내는 Pseudomonas속 1,196균주를 선발하고, 연부병균 Erwinia herbicola에 대한 생육억제력과 생물학적 검정으로 발병억제력이 강한 2균주를 선발하여 Pseudomonas fluorescens S4(PS4)와 Pseudomonas fluorescens S65 (PS65)로 동정하였다. 배춧잎을 이용하여 분리길항균의 연부병균에 대한 생물학적 억제력을 확인한 결과 상당한 발병억제력을 확인하였다. 이들 두 균주가 가장 높은 길항력을 나타내는 조건은 배양 초기배지의 pH를 7.0으로하여 25℃에서 3일간 배양했을 때였다. 화학농약 및 항생물질에 대해 2균주 모두 benomyl, propamocab hydrochloride, fosethyl-Al-folpet, vancomycin, penicillin 및 lincomycin에 내성을 나타냈고, PS4균주는 erythromycin에 대해서도 내성을 나타냈다. For the selection of powerful antagonistic bacterium for biological control of Erwinia sp. causing vegetable soft rot, two excellent strains(S4, S65) were selected from 1,196 strains of bacteria which were isolated from rhizospere in vegetable root rot suppressive soil. Selected 2 strains were identified to be a species to Pseudomonas fluorescens S4(PS4) and Pseudomonas fluorescens S65 (PS65). The highest of inhibitory activity was produced in 523 synthetic broth medium at pH 7.0 and 25℃ during 3 day culture. The isolate strains were resistant to the agricultural chemicals such as benomyl, proamocab hydrochloride and fosethyl-Al-folpet, and the antibiotics such as vancomycin, penicillin and lincomycin, only PS4 was resistant to erythromycin.

      • 응집제 생산 균주의 분리 및 탐색

        The microorganism producing a kind of flocculant was screened from soils. The isolated strain was identified as Agrobacterium sp.from the results of morphological and biochemical characteristics. The optimum temperature and optimum initial pH for flocculant production of the isolated strain were 30℃ and pH 7.5, respectively.

        Pseudomonas sp.에 의한 채소병원균의 생물학적 억제

        채소 등 작물의 부패균인 Erwinia carotovora subsp. carotovora의 생육을 억제하는 길항세균을 경작지 표토층으로부터 분리하고 배양조건에 따른 억제력변화를 조사하고, 길항관련 유전자의 소재를 확인하였다. 분리균들중 가장 우수한 억제력을 갖는 S4, S14, S65균주를 최종 선발하고 Pseudomonas 근연종으로 동정하였다. 523 배지를 사용하여 배양조건에 따른 억제력변화를 조사한 결과 배지초기 pH를 7∼8로 조정하여 30℃에서 3일간 배양하였을 때 가장 높은 억제력을 보였고, 배지성분중 탄소원으로 mannitol과 sorbitol, 무기염으로 CaCl_2를 첨가 배앙했을 때 효과적이었으나 질소원에 대한 감수성은 낮았다. 분리균들의 토양정착시 시비에 의한 효과가 있었으며,benomyl, fosethyl-Al-folpet, penicillin, lincomycin 등의 화학농약 및 항생물질에 대한 내성이 있었다. 이들의 DNA 분포양상은 S14 균주만 plasmid DNA를 확인할 수 있었고 길항관련 유전자는 chromosomal DNA에 있음을 확인하였다. For the selection of powerful antagonistic bacterium for biological control of soil borne Erwinia carotovora subsp. carotovora causing rot of vegetable, excellent strains (S4, S14, S65) were selected from 1,196 strains of bacteria which were isolated from rhizosphere in vegetable root rot-suppresive soil. Strains were identified to be Pseudemonas species with Api 20NE kit. Antagonistic substance was produced in 523 synthetic broth medium at pH 7∼8 and 30℃ during 3 days culture. The substance was stable in the pH range of 6 to 9. When the basal medium was supplemented with mannitol and sorbitol as carbon source and calcium chloride as metal salt, the production of the inhibitory substance was increased. The inhibitory acitivity was increased by the addition of fertilizer in soil. The isolated strains were resistant to the agricultural chemical such as benomyl and fosethyl-Al-folpet, and the antibiotics such as penicillin and lincomycin. We had found that Pseudomonas sp. S14 strain had a single plasmid. After treated with acridin orange for curing, we confirmed the existence of antagonistic gene in the chromosomal DNA.
