        Expression and Possible Role of Phospholipase C $\beta1$ and $\gamma1$ in Mouse Oocyte Maturation and Preimplantation Embryo Development

        Phospholipase C (PLC)는 다양한 세포주에서 세포내 신호전달에 중요한 역할을 한다고 알려져 있으나, 생쥐 난자성숙 과정과 착성전 배아발생 과정에서 PLC의 역할과 발현은 아직 연구된 바 없다. 본 연구에서는 난자성숙과 착상전 배아발생 과정에서 생쥐의 PLC \beta 1과 \gamma 1의 유전자 발현을 조사하기 위하여 한 개의 난자 혹은 배아에서 추출된 total RNA를 사용하여 경쟁적 RT-PCR 방법으로 mRNA를 정량하였다. PLC \gamma 1의 유전자 발현을 조사하기 위하여 한 개의 난자 혹은 배아에서 추출된 total RNA를 사용하여 경쟁적 RT-PCR 방법으로 mRNA를 정량하였다. PLC \gamma 1의 유전자는 전혀 발현하지 않았다. 수정후 PLC \betta 1과 \gamma 1의 유전자 발현은 상실기 배아에서 증가하기 시작하여 포배기 배아에서는 현저히 증가하였다. 난자성숙과 착상전 배아발생 과정에서 protein kinase C(PKC) 신호전달체게에 의한 PLC의 역할을 조사하기 위하여 PKC의 억제제인 sphingosin, PKC의 촉진제인 $diC_{8}$, 그리고 PLC의 억제제인 U73122의 효과를 조사하였다. Spihingosine은 처리후 1시간 이내에 대조군에 비해 20% 정도의 난자성숙을 촉지하였으나 U73122는 유효한 효과를 보이지 않았다. U73122는 상실기 배아의 compaction을 억제하였으나 $diC_{8}$에 의하여 부분적으로 극복되었다. 이상의 결과는 PLC \beta 1과 \gamma 1 유전자가 생쥐의 착상전 발생단계에서 특이젖으로 발현하고 있으며, 난자성숙과 착상전 초기배아에서 PKC-PLC 신호전달체계가 관여하고 있으리라 사료된다. It has been wel known that phospholipase C(PLC) plays an important role in the intracellular signaling in a variety of cell types. However, involvement of PLC in mouse oocyte maturation and preimplantation embryo development remains unknown. The present study examined the expression patterns of the mouse PLC \beta 1 and \gamma 1 during oocyte maturatio and preimplantation embryo development study examined the expression patterns of the mouse PLC \beta 1 and \gamma 1 during oocyte maturation and preimplantation embryo development by the competitive reverse transcription-polymerase chain reaction (RT-PCR method). PLC \gamma 1 mRNA (0.1 fg) was readily detected in germinal vesicle (GV)-stage oocyte and its level was reduced as meiotic resumption proceeded. PLC-\beta 1 mRNA (<0.1 fg) as detected at low level at GV-stage oocytes and scarcely detected at germinal vescle breakdown (GVBD)-stage oocytes. After fertilization, both PLC \beta 1 and \gamma 1 mRNA levels began to increase at morula-stage embryos (0.2 fg) and were more prominent in blastocyst-stage embryos(1 fg). to elucidate the possible involvement of PLC via protein kinase C(PKC) pathway during oocyte maturation and preimplantation embryo development , the effects of sphingosine (PKC inhibitor), sn-$diC_{8}$(PKC activator) anc U73122 (PLC ingibitor) were examined. Treatment of GV-stage oocytes with sphingosine (20 \mu M) facilitated the meiotic resuption by 10-20 over the control within 1 h as judged by GVBD, whereas U73122 failed to show any significant effect. U73122 (10 \mu M) effectively blocked the compaction of morula, while sn-$diC_{8}$(50 \mu M). In summary, the present study shows that the mouse PLC \beta 1 and \gamma 1 are expressed in a developmental stage-specific manner and PLC-PKC pathway may be involved in early preimplantation embryo development.

        The immune thrombocytopenia(ITP) is defined as a platelet count of less than 100,000/ L. It is generally known as characterized by the bleeding manifestations of skin and/or mucosa like ecchymosis due to low platelet count, but reports of the related intraosseous lesions are not common. The idiopathic bone cavity(IBC) is an empty space of the bone, which occurs mainly in the long bones. It is found predominantly in the mandible in case of the maxillofacial area. In general, it appears as an isolated unilocular lesion without the correlation of the teeth. Although the cause of the IBC is supposed to be associated with hemostatic problems, the etiology is unclear and it was not disclosed the relevance of specific systemic disease. In this present case, IBCs that occurred in mandible of patient who has IPT was treated by curettage with platelet transfusion.

      • Alteraation of Gonadotropin-releasing Hormone and Luteinizing Hormone ${\beta}$-Subunit mRNA Levels in Neonatally Estrogenized Female Rats

        Treatment of newborn female rats with gonadal steroids induces permanent sterility in adulthood. We investigated the alteration in expression patterns of gonadotropin-releasing hormone (GnRH) and luteinizing hormone (LH) in neonatally estrogenized sterile rats (ESR). Newborn female rats received daily injections of 17${\beta}$-estradiol (E, 10 ${\mu}$g) from the day of birth (day 1) to postnatal day 5. Controls were subjected to vehicles over the same period. All animals were sacrificed on week 7 after birth. Hypothalamic GnRH mANA levels were markedly higher in all ESR than in controls, while hypothalamic GnRH contents in ESR increased in proportion to the frequency of daily administration of E. However, both pituitary LH6 mRNA and serum LH levels were inversely decreased by the same treatment. The data indicate that neonatal exposure of E equally elevates the expression of GnRH gene, but reduces the secretion of GnRH, accordingly leading to attenuation of LH6 gene expression and circulating LH levels. The temporal effect of E and/or progesterone (P) on GnRH and LH6 mRNA levels was also examined in ESR. Newborn female rats were daily injected with E (10 ${\mu}$g) or vehicle for five successive days from day 1 and ovariectomized at week 5. They were implanted with E (235 ${\mu}$g/ml) two days prior to week 7, injected with P (1 mg) 42 h later, and sacrificed 7 h after P administration. In ovariectomized controls, hypothalamic GnRH mRNA levels were dropped to half by treatment of E and restored by subsequent treatment of P. The negative feedback action of E on GnRH mRNA levels observed in ovariectomized rats was completely blocked by neonatal exposure of E. The change in pituitary LH mRNA levels was similar to that in hypothalamic GnRH mRNA levels. Taken together, the results suggest that neonatal treatment of E alters the synthesis and release of GnRH in adulthood and furthermore blocks the negative feedback regulation of E which occurs normally after ovariectomy.

        Non-medical use of drugs to enhance performance at Olympic Games by athletes has been prohibited by the International Olympic Committee(IOC) since 1968 on medical and ethical grounds. IOC wants to protect athlete from harmful side effect of drugs due to misuse of it. It also, ethically, wants to have Games run on fair base, not fortified by performance enhancing drugs. The banned substance include stimulants, narcotic analgesics, anabolic, steroids, β-blockers and diuretics. In order to prevent the positive reaction in the doping test induced by herb medicine, this study was done on about Bojungchisheup-tang. The laboratory set up doping analyses methods to cover as many drugs as possible without sacrificing sensitivity and specificity within one procedure. Its screening method considered of four different procedures. The results were negative. According to the above results, Bojungchisheup-tang taken by athletes would show the negative reaction in the doping test. So its prescription for athletes can be given without worries of the doping test.
