        Trehalase 에 관한 연구 ( 1 ) Sarcoma 180 복수암에 걸린 Mouse Trehalase 에 관하여

        The activity of trehalase (α, α-trehalose 1-D-glucohydrolase, ED 3. 2. 1. 28) in Sarcoma 180 ascites tumor cells and ascitic fluid which harvested at 7 days after transplantation has been studied. The activity of trehalase was measured by the method of Lapp and Mason. The results obtained were as follows; 1. The activity of trehalase could not be found in the Sarcoma 180 ascites tumor cells. 2. The activity of trehalase in ascitic fluid was 938.54 units/㎖. 3. In the ascitic fluid, the trehalase activity is relatively low compared with those in insect, microorganism, Ascaris suum and mouse kidney. 4. Trehalase from ascitic fluid has been purified about 58 fold by ammonium sulfate precipitation, DEAE-cellulose column chromatography and Sephadex G-200 gel filtration. 5. The optimal pH for enzymatic activity was 5.4, and optimal temperature was 60℃. 6. The enzyme was specific for trehalose as substrate. 7. The product of the reaction was identified as glucose by paper chromatographic method.

      • 사람태반 미토콘드리아의 Superoxide Dismutase의 정제 및 성상

        Cytotoxic superoxide radical(O_2^-) is a common intermediate of oxygen reduction reaction in the respiring cells and therefore these cells should be protected against the cytotoxic effect of superoxide radicals. At the present time, three types of superoxide dismutase, copper- and zinc-containing, iron-containing and manganese- containing superoxide dismutase, have been isolated from both eukaryotic and prokaryotic cells. The purification and properties of cytosolic superoxide dismutase of human term placenta have been reported previously from this laboratory. In this study, therefore, the experiment was carried out to characterize mitochondrial .superoxide dismutase of the placenta. Mitochondria of human placenta were isolated from human term placenta cell homogenate by differential centrifugation and the mitochondrial superoxide dismutase was purified by DEAE-cellulose column chromatography. The results were summarized as follows: 1. The distribution of superxode dismutase in terms of activity, in the cytosolic and mitochondrial fractions was found being 85%(7.4 units/g) and (1.3 units/g) respectively. 2. The distribution of mitochondrial superoxide dismutase in intermembrane space and matrix of mitochondria was found being 47% and 53% respectivly. 3. The mitochondrial superoxide dismutase was purified approximately 52 fold. The molecular weight of the purified enzyme was estimated to be 85,000 by Sephadex G-100 gel filtration method. 4. Purified mitochondrial superoxide dismutase contained 1.82 atoms of manganese per molecule and was very similar to the superoxide dismutase previously isolated from other eukaryotes. 5. The activity of manganese-containing superoxide dismutase was inhibited about 10% by 1mM cyanide but it is inhibited as much as 88% by ethanol-chloroform(5:1, v/v) mixture. 6. The ultraviolet absorption spectrum of purified mitochondrial superoxide dismutase was also similar to those of the previously prepared eukaryotic enzyme. 7. The activity of xanthine oxidase of human term placenta was found to be 23.6 units/g and the relative activity in the cytosolic and mitochondrial xanthine oxidase was 83% and 17%, respectively.

        Mouse에 이식한 Sarcoma 180 복수암세포 및 복수에서 trehalase ($\alpha$, $\alpha-$ trehalose 1-D-glucohydrolase, EC 3. 2. 1. 28)의 분포를 관찰하였으며 이 효소를 부분 정제하였다. Trehalase의 활성도는 Lapp and Mason 법으로 측정하였으며 효소의 정제는 ammonium sulfate에 의한 침전 DEAE-cellulose column chromatography 및 Sephadex G-200 chromatography 등을 행하여 다음과 같은 결혼을 얻었다. 1. Sarcoma 180 복수암세포는 trehalase가 함유되어 있지 않다. 2. 7일만에 채취한 복수에서 이 효소의 활성도는 938.54 unit/ml이었다. 3. 복수의 이 효소의 활성도는 곤충, 기생충 및 미생물에 비하여 매우 낮았다. 4. 복수의 이 효소를 약 58배 정제하였으며 비활성도는 414.5이었다. 5. 이 효소의 반응 최적온도는 $60^{\circ}C$이며 반응최적 pH는 5.4 이다. 6. 이 효소는 glucose를 함유한 disaccharide의 기질중 trehalose에만 반응성이 있다. 7. 반응 생성물이 glucose임을 paper chromatography로 확인할 수 있었다. 8. 이상과 같은 결과로 보아 Sarcoma 180 복수암세포를 함유한 마우스의 복수에 trehalase가 존재함을 알 수 있었다.       • 석탄점화온도의 직접적인 측정에 의한 아역청탄 촤산화 반응율에 대한 실험적 연구

        The experiment was designed to study the coal char oxidation kinetics. The kinetics has been estimated using the Semenov's thermal spontaneous ignition theory adapted to coal char particle ignition temperature. The ignition temperature of coal char particle is obtained by a method of direct measurement of the particle temperature. The ignition temperature is detected by means of a thermocouple which is used as both a heating and a measuring element. The ignition temperatures for bituminous coal YAKUTUGOL have been measured at 4 particle sizes in the range of 0.4 - 1 ㎜. The ignition temperature of the particle increases with the raising of the diameter. The results were used to calculate the activation energy and the pre-exponential factor. As a result, the kinetics are in agreement with coal char kinetics from other investigations.

        A Phased-Based Focusing Mechanism and English Locative Inversion Constructions

        This paper presents an alternative phase-based theory of English Locative Inversion Construction (LIC). In discussing some previous analyses, it has been noticed that some additional syntactic mechanisms such as EPP checking of a locative PP, a null expletive Pro, or a Focus phrase at the vP edge are necessary to account for the complicated discourse-related properties of LIC. On the other hand, we pay careful attention to the information structure of a locative PP, a logical subject, and an unaccusative verb appearing in LIC. In analyzing them, it is assumed here that the spell-out domain of a phase is exactly identical with the focus domain of the phase. This focusing mechanism of phase can straightforwardly explain why the locative PP and unaccusative verb of LIC have a Topic or topicalized interpretation while the logical subject has a Focus interpretation. Under our proposed structure, only logical subject remains in the focus domain of the CP phase (i.e., TP). Without postulating any subsidiary syntactic operations, therefore, we can provide a better theory in analyzing English LICs.

        Wh-scrambling as D-linking Movement

        This paper addresses that Wh-scrambling in Korean is closely related to the discourse-based operation required by information structure of a sentence; especially, it can be analyzed as movement of D-linked Wh-phrases in the sense of Pesetsky (1987). This paper stands against the traditional view that Wh-scrambling is analyzed as either an optional Wh-movement or Focus movement. Rather, providing the counterevidence that Korean Wh-scrambling does not have any typical properties Wh-movement and Focus movement usually show in other languages, this paper has throughly argued that in certain syntactic and semantic respects, scrambling appears to mark a direct object Wh-phrase D-linked by placing it in the left periphery of a sentence. As a result, this paper suggests that scrambled Wh-phrases are derived from D-linking movement, which is a narrow syntax operation to the edge (more specifically. Spec, ΔP) of CP for checking a discourse feature (i.e. [+Δ]). From a semantic perspective, D-linked Wh-phrases should be interpreted as non-quantificational, presupposed and specific. These semantic properties are sharply distinct from the ones of typical Wh- or Focused phrases.

        A Non-raising Approach to Matrix Object in Korean ECM Constructions

        This paper presents a non-raising and discourse-based approach to Korean ECM constructions. Against the popular view that matrix object of ECM is a result of SOR, it is suggested in this paper suggests that matrix object is a sentential contrastive Topic adjunct, which is base-generated in the matrix clause. It is thoroughly argued that the previous analyses based on obligatory or optional SOR are not entirely free from certain theoretical stipulations caused by the movement-related operations such as movement chain, case filter, and Projection Principle. These difficulties are mainly due to the fact that Korean ECM constructions have a fully projected embedded clause (i.e. CP), instead of defective TP. For example, the availability of overt or covert embedded subject in the ECM construction is one of the empirical drawbacks under the SOR analysis. On the other hand, a non-raising analysis provided here is based on the assumption that matrix object of Korean ECM is a sentential contrastive Topic adjunct, which has a prosodic prominence. The greatest advantage of our analysis is that it is totally free from all the syntactic problems shown in the previous analyses. In other words, matrix object of Korean ECM is not a result of either optional or obligatory SOR. As an alternative approach to the SOR analysis, a non-raising analysis of Korean ECM constructions is on better theoretical and empirical ground.

      • 우리나라 降雨量의 確率分布型에 관한 硏究

        The purpose of this study deals the derivation of the maximum rainfall depth related to the duration of rainfall and frequncy for 14 meteorological stations in Korea. The rainfall data used in this study are duration of 10min, 60min, 8hrs and 24hrs. The 24 years historical records are consist of the year from 1961 to 1984. The five distributions of the rainfall frequency are selected such as 2 parameter log-normal, Type I extremal, log-Pearson Type III, Gamma and Pearson Type III. The results are concluded as follows; 1.The relationship between rainfall depth with duration and frequency are fairly good approximation in function of *****(p. 218) 2.The results will be the effective standard to determine the design storm for sewer or culvert design. 3.According to this study, the probability distribution of rainfall in Korea shows that there are no difficulties to choose any five type of distributions.

      • 혼합연료 가스의 열면점화 특성에 관한 실험적 연구

        In this study, the experimental of the ignition of methan-air mixture by a Hot-surface has been carried out. It was introduced to an experimental method of flame visualization and ignition delay by hot-surface. The flame was visualized using schlieren method, and reliable ignition delay time was obtained by synchronizing PDS and thermocouple. Based on the data obtained through schlieren method, it was found that the flame front of the mixed gas’s initial ignition was first formed at the contact point with the hot-surface and then developed to the nozzle. The P.D.S. could measure to the intensity of the flame luminous and the ignition temperature was obtained by a thermocouple. Reliable ignition delay was determined by synchronizing these two methods.
