        Bombyx mori b-tubulin Promoter for High-level Expression of Heterologous Genes

        We previously isolated 9 clones that show stronger signal compared to Bombyx mori cytoplasmic actin gene (BmA3) by using a dot blot hybridization. In this study, we focused on one clone among these clones which has high amino acid similarity with b-tubulin gene of B. mori. This clone was ubiquitously expressed in all tissues and developmental stage of B. mori. As result of promoter assay using dual luciferase assay system, we found the highest transcription activity region (-750/-1) in the 5’-flanking region of b-tubulin gene, which has about 47 fold more intensive promoter activity than BmA3 promoter. Moreover, the b-tubulin promoter was normally regulated in Bm5, Sf9, and S2 cells. Therefore, we suggest that b-tubulin promoter may be used more powerful and effectively for transgene expression in various insects containing B. mori as a universal promoter.

        에너지 하베스팅 센서네트워크에서 데이터의 긴급성을 고려한 QoS기반 MAC프로코콜 설계

        에너지 하베스팅 센서네트워크에서는 노드의 전력상황을 고려하여 중계노드를 선택하는 매체접근제어 프로토콜이 요구된다. 기존의 EH-WSN (Energy harvesting wireless sensor network)의 연구들은 전력측면을 강조하다보니 센싱된 데이터의 긴급성과 같은 QoS (Quality of Service)를 크게 고려하지 못했다. 데이터의 긴급성에 따라 요구되는 전력 상황과 전송지연이 달라지도록 하여 데이터의 QoS에 맞는 매체접근이 이루어질 수 있도록 해야 한다. 중계노드의 경우 긴급하지 않은 상황에서 전력에 상관없이 모든 데이터를 중계하게 되면 막상 긴급 데이터가 발생할 경우 전력문제로 인해 지연이 크게 발생하는 상황이 생길 수 있다. 본 논문에서는 데이터의 긴급성에 따라 중계노드에 요구되는 전력상황을 다르게 설정함으로써 긴급데이터 발생 시에 발생할 수 있는 전력부족상황을 최소화하는 매체접근제어프로토콜을 설계하고 시뮬레이션을 통해 그 성능을 분석하였다. 성능분석결과 긴급데이터의 시간지연 및 신뢰성 성능이 향상됨을 확인할 수 있었다. In the EH-WSN (Energy harvesting wireless sensor network), a MAC (medium access control) protocol is required to select a relay node considering the power status of a node. Existing EH-WSN studies emphasize the power aspect, so it does not consider the QoS like the urgency of the sensed data. The required power and transmission delay must be changed according to the urgency so that the medium access control according to the data QoS can be performed. In case of relay node, relaying data without consideration of data urgency and node power may cause delay due to power shortage in case of urgent data. In this paper, we designed a MAC protocol that minimizes the power shortage that can occur during emergency data generation. For this, relay node requirements are set differently according to the urgency of data. The performance was analyzed through simulation. Simulation results show the reduced latency and improved reliability of urgent data transmission.

        Bioconversion and growth performance of Hermetia illucens in single fruit by-products

        As agricultural production increases due to population growth, agricultural by-products that are generated at all production stages increase commensurately. Black soldier fly (BSF) (Hermetia illucens) treatment has potential as an environmentally friendly process to combat the environmental pollution caused by agricultural by-products. This study examined the utilization of BSF larvae in the decomposition of the by-products of apples and mandarins, fruits commonly produced in South Korea. The BSF test larvae were fed apple pomace or mandarin waste, and the control larvae were fed calf feed. Larval weight and size were measured at 4-day intervals until larvae reached the pre-pupal stage. Larval development time, survival rate, and BSF fecundity rate were calculated for all three substrates. Waste reduction and bioconversion ratios were also calculated. The developmental time of larvae fed with apple pomace and mandarin waste was greater than that of the control larvae. The average weight of larvae fed with the fruit by-products was less than that of the control. There was no significant difference in the survival rate of BSF larvae or the fecundity rate of BSF between the substrates used in this study. BSF larvae decomposed 48.0% and 61.5% of apple pomace and mandarin waste, respectively. The bioconversion efficiency rates of BSF larvae fed with apple pomace and mandarin waste were 9.1% and 12.1%, respectively. These results indicated that decomposition of single fruit by-products by BSF larvae is an environmentally friendly and effective bioconversion process.

        Mo¨ssbauer parameters of natural iron have been measured from 25K to 298K. The isomer shift was not linear with absolute temperature, except at high temperature, because of quantized lattice levels. We have obtained the Debye and einstein temperature by fitting the kinetic temperature proposed by Mazo and Kirwood. We have found out the following results; 1) Isomer shift values tend to increase, while temperature decrease. 2) Hyperfine splitting values tend to increase, while temperature decrease. 3) Debye temperature is 264 K. 4) Einstein temperature is 202 K.

        Influence of starvation on the larval development of the Black Soldier Fly, Hermetia illucens (Diptera: Stratiomyidae)

        The rearing of black soldier flies in Korea is affected by seasonal factors such as temperature and sun light; for this reason, it requires a great deal of effort to rear and maintain black soldier flies during the winter. In this study, we investigated how starvation affects larval development. After hatching the larvae, they were fed for 5 or 10 d and then starved for a certain period to see how they recovered. The length and width of larvae were estimated to be 18.18 to 21.96 mm, 5.19 to 6.04 mm, respectively. Larvae survivorship to the pupal stage was significantly different between groups and periods of starvation. The groups of fed for 5 d then starved showed a high survival rate until 20 d starvation and then the survival of larvae rapidly decreased. The survival rate of the larvae was abruptly decreased for 20 d starvation in the groups of fed for 10 d and starved, and then gradually decreased until 60 d starvation thereafter. Our research attempted to influence larvae development through starvation and provides basic information on how to culture the black soldier fly effectively and economically throughout the year.

        Enzymatic activity of Endoplasmic Reticulum Oxidoreductin 1 from Bombyx mori

        Most proteins produced in the endoplasmic reticulum (ER) of eukaryotic cells fold via disulfide formation (oxidative folding). Oxidative folding is catalyzed by protein disulfide isomerase (PDI) and PDI-related ER protein thiol disulfide oxidoreductases (ER oxidoreductases). In yeast and mammals, ER oxidoreductin-1s (ERO1s) supply oxidizing equivalent to the active centers of PDI. We previously identified and characterized the ERO1 of Bombyx mori (bERO1) as a thioredoxin-like protein that shares primary sequence homology with other ERO1s. Here we compare the reactivation of inactivated rRNase and sRNase by bERO1, and show that bERO1 and bPDI cooperatively refold denatured RNase A. This is the first result suggesting that bERO1 plays an essential role in ER quality control through the combined activities of bERO1 and bPDI as a catalyst of protein folding in the ER and sustaining cellular redox homeostasis.
