This paper reports the basic results obtained from an investigation of the microstructure of lithium niobate (LiNbO3) crystals fabricated by using the focused ion beam (FIB) sputtering method. The morphology characterization of the microstructure is explained in terms of scanning electron microscopy observations based on the geometrical integrity, surface smoothness, cross-sectional area, aspect ratio, etc. The optimized micro-fabrication conditions and the sample morphology characterization are presented, and the sputtering rate of Ga ions for a 30-keV energy milling of the LiNbO3 crystal is calculated, which is lower than the theoretical one gained from “The Stopping and Range of Ions in Matter” (SRIM) sputtering simulation due to the redeposition effect being enhanced with increasing milling depth

Planar waveguides have been fabricated in z-cut lanthanum gallium silicate (LGS) crystal by 6.0 MeV oxygen-ionsimplantation with a dose of 6×10^15 ions/cm². The waveguides were characterized by the prism-coupling method. The dark modes are measured before and after annealing at 600, 700, 750 and 800℃ for different times in air. The refractive index profiles are reconstructed by using a reflectivity calculation method. TRIM'98 code is used to simulate the process of the oxygen-ion implantation into LGS. It is found that the refractive-index change may be partly due to the damage induced by nuclear collision, and the waveguide has thermal stability below 600℃. In this paper, the kinetics of a cloned special glucosidase, named ginsenosidase type III hydrolyzing 3-O-glucoside of multi-protopanaxadiol (PPD)-type ginsenosides, were investigated. The gene (bgpA) encoding this enzyme was cloned from a Terrabacter ginsenosidimutans strain and then expressed in E. coli cells. Ginsenosidase type III was able to hydrolyze 3-O-glucoside of multi-PPD-type ginsenosides. For instance, it was able to hydrolyze the 3- O-β-D-(1→2)-glucopyranosyl of Rb1 to gypenoside XVII, and then to further hydrolyze the 3-O-β-D-glucopyranosyl of gypenoside XVII to gypenoside LXXV. Similarly, the enzyme could hydrolyze the glucopyranosyls linked to the 3-O- position of Rb2, Rc, Rd, Rb3, and Rg3. With a larger enzyme reaction Km value, there was a slower enzyme reaction speed; and the larger the enzyme reaction Vmax value, the faster the enzyme reaction speed was. The Km values from small to large were 3.85 mM for Rc, 4.08 mM for Rb1, 8.85 mM for Rb3, 9.09 mM for Rb2, 9.70 mM for Rg3(S), 11.4 mM for Rd and 12.9 mM for F2; and Vmax value from large to small was 23.2 mM/h for Rc, 16.6 mM/h for Rb1, 14.6 mM/h for Rb3, 14.3 mM/h for Rb2, 1.81mM/h for Rg3(S), 1.40 mM/h for Rd, and 0.41 mM/h for F2. According to the Vmax and Km values of the ginsenosidase type III, the hydrolysis speed of these substrates by the enzyme was Rc>Rb1>Rb3>Rb2>Rg3(S)>Rd>F2 in order.

PSMB1 (proteasome subunit beta type 1) is a core component of the 20S proteasome, and based on its structure, it might have a crucial function in the transcription of certain genes. Rheb (Ras homolog enriched in brain) is a Ras-like small GTPase that acts as an upstream positive regulator of the mechanistic target of rapamycin complex 1 (mTORC1) pathway. We cloned and characterized PSMB1 (KY310590.1) to determine its function in cell cycle progression and proliferation of goat fetal fibroblasts (GFbs). Further, by yeast two-hybrid screen and coimmunoprecipitation, we conformed that PSMB1 interacts directly with Rheb. An siRNA was designed and expressed targeting PSMB1 mRNA in GFbs and inducing cell cycle arrest. Rheb overexpression in GFbs significantly increased the number of S phase cells and growth efficiency compared with control cells. These data indicate that PSMB1 and Rheb have important functions in the cell cycle and proliferation of GFbs, indicating that their interaction governs many processes in GFbs.

Mischmetal of Bayan Obo ore was utilized to prepare the high performance (Pr7.34Nd21.86)1-xMMxFe68.7Al0.1-Cu0.12Co0.88B ribbons using melt-spinning method. Phase composition and magnetic properties were investigated at room temperature. The ribbons mainly consist of R2Fe14B phase in isotropic nanostructure. Both coercivity and maximum energy product decrease with the increase of MM content. The magnetic parameters of the ribbons with MM = 20 % in mass are Br = 7.38 kGs, Hcj = 13.66 kOe, (BH)max = 11.81 MGOe. Henkel plots were applied to demonstrate the exchange coupling interaction between grains.

Wang, Dong Xing,Shah, Asif,Zhou, Lei,Zhang, Xue Feng,Liu, Chun Jing,Huang, Hao,Dong, Xing Long Korean Society of Microscopy 2015 Applied microscopy Vol.45 No.4
Carbon-coated Ni, Cu and Sn nanocapsules were investigated by means of X-ray diffraction (XRD), transmission electron microscopy (TEM) and a four-point probe device. All of these nanocapsules were prepared by an arc-discharge method, in which the bulk metals were evaporated under methane ($CH_4$) atmosphere. Three pure metals (Ni, Cu, Sn) were typically diverse in formation of the carbon encapsulated nanoparticles and their different mechanisms were investigated. It was indicated that a thick carbon layers formed on the surface of Ni(C) nanocapsules, whereas a thin shell of carbon with 1~2 layers covered on Cu(C) nanocapsules, and the Sn(C) nanocapsules was, in fact, a longger multi-walled carbon nanotubes partially-filled with metal Sn. As one typical magnetic/dielectric nanocomposite particles, Ni(C) nanocapsules and its counterpart of oxide-coated Ni(O) nanocapsules were compared in the electrically conductive behaviors for further applications as the electromagnetic materials.

Curcumin, a polyphenol compound derived from the rhizome of the plant Curcuma longa L. has been verified as an anticancer compound against several types of cancer. However, understanding of the molecular mechanisms by which it induces apoptosis is limited. In this study, the anticancer efficacy of curcumin was investigated in human gastric adenocarcinoma SGC-7901 cells. The results demonstrated that curcumin induced morphological changes and decreased cell viability. Apoptosis triggered by curcumin was visualized using Annexin V-FITC/7-AAD staining. Curcumin-induced apoptosis of SGC-7901 cells was associated with the dissipation of mitochondrial membrane potential (MMP) and the release of cytochrome c into the cytosol. Furthermore, the down-regulation of Bcl-2 and up-regulation of Bax that led to the cleavage of caspase-3 and increased cleaved PARP was observed in SGC-7901 cells treated with curcumin. Therefore, curcumin-induced apoptosis of SGC-7901 cells might be mediated through the mitochondria pathway, which gives the rationale for in vivo studies on the utilization of curcumin as a potential cancer therapeutic compound.

Multi-target drug design, in which drugs are designed as single molecules to simultaneously modulate multiple physiological targets, is an important strategy in the field of drug discovery. QT-011, a tamibarotene-furoxan derivative, was here prepared and proposed to exert synergistic effects on antileukemia by releasing nitric oxide and tamibarotene. Compared with tamibarotene itself, QT-011 displayed stronger antiproliferative effects on U937 and HL-60 cells and was more effective evaluated in a nude mice U937 xenograft model in vivo. In addition, QT-011 could release nitric oxide which might contribute to the antiproliferative activity. Autodocking assays showed that QT-011 fits well with the hydrophobic pocket of retinoic acid receptors. Taken together, these results suggest that QT-011 might be a highly effective derivative of tamibarotene and a potential candidate compound as antileukemia agent.
