• Calpain-dependent Beclin1 cleavage stimulates senescence-associated cell death in HT22 hippocampal cells under the oxidative stress conditions

        Nguyen, Huynh Quoc,Zada, Sahib,Lai, Trang Huyen,Pham, Trang Minh,Hwang, Jin Seok,Ahmed, Mahmoud,Kim, Deok Ryong Elsevier 2019 Neuroscience Letters Vol.701 No.-

Abstract

Oxidative damage in neurons including glutamate excitotoxicity has been linked to increasing numbers of neuropathological conditions. Under these conditions, cells trigger several different cellular responses such as autophagy, apoptosis, necrosis and senescence. However, the connection between these responses is not well understood. In this study, we found that the 60-kDa BECN1 was specifically degraded to a 40-kDa fragment in hippocampal HT22 cells treated with 5 mM glutamate. Increased BECN1 cleavage was specifically associated with a decrease in cell viability under oxidative stress. Interestingly, this BECN1 cleavage was specifically inhibited by a calpain inhibitor ALLN but was not affected by other protease inhibitors. Also, the BECN1 cleavage was not detected in calpain-4-deficient cell lines. Furthermore, calpain cleaved BECN1 at a specific site between the coiled-coil domain and Bcl2 homology 3 domain, which is associated with the anti-apoptotic protein Bcl-2. Moreover, some cellular senescence markers, including β-galactosidase, p21, p27Kip1, p53 and p16INK4A, increased proportionally to those of BECN1 cleaved fragments. These results suggest that calpain-mediated BECN1 cleavage under oxidative conditions is specifically associated with cell death induced by cellular senescence.

Highlights

BECN1 was specifically degraded to a 40-kDa fragment accompanying with decrease of cell viability under oxidative stress.

BECN1 cleavage was inhibited by calpain inhibitor but not by other protease inhibitors.

Calpain cleaves BECN1 at a specific site between CCD and BH3 domains.

BECN1 degradation stimulates senescence-associated cell death at the oxidative stress.

In this paper, a nonlinear optimal control of a 3-D overhead crane is investigated, where time varying rope length is considered in the control design. The Euler-Lagrange equations is employed to derive the dynamic model of the overhead crane. The proposed optimal control law guarantees the minimum of the sway angles together with the control efforts. The numerical simulations and experiments are carried out to verify the effectiveness of the proposed control method, where the con-sistency between numerical simulation and experiment results are shown.

Prediction-based energy policy for mobile virtual desktop infrastructure in a cloud environment

        Nguyen, T.D.,Hung, P.P.,Dai, T.H.,Quoc, N.H.,Huynh, C.T.,Huh, E.N. Elsevier science 2015 Information sciences Vol.319 No.-

Using cloud services from mobile devices has become a growing trend because of its mobility and convenience. However, mobile devices join and leave cloud services more frequently than traditional computers, which causes energy inefficiency in a cloud data center. Waste, in the form of energy and cooling requirements, particularly occurs when a mobile device disconnects from a service, but the cloud servers, known as virtual machines (VMs), continue running. The VMs should transition to lower-power states instead remaining active. However, transition to a lower-power state causes a service delay when users reconnect to the service because VMs in a lower-power state are not ready to serve. Therefore, an efficient energy policy must not only maximize energy savings but also minimize service delays. In this paper, we propose two approaches to energy efficiency: an Instant Energy Policy (IEP) that can quickly find an appropriate low-power state based on a predicted disconnection time and a Prediction-based Energy Policy (PrEP) that determines when to transition VMs to a low-power state and when to return them to the active state based on each users activity history. IEP predicts the unknown disconnection time using the multisize sliding windows workload estimation technique, which supports a non-stationary environment. This method can quickly obtain an energy policy, but it is limited when disconnection time fluctuates widely. PrEP presents an improved approach to achieve an optimal global result with respect to both energy consumption and service delay. Through simulations with a real-world dataset collected by the MIT Human Dynamics Lab, we show that PrEP provides approximately 20% power saving over the benchmark policies while guaranteeing minimal service delay.

The Effect of Institutions on Productivity Spillovers from FDI to Domestic Firms: Evidence in Vietnam

Huynh Quoc Vu,Pham Thi Bich Ngoc,Nguyen Le Hoang Thuy To Quyen 사람과세계경영학회 2022 Global Business and Finance Review Vol.27 No.3

Purpose: The purpose of this study is to investigate the impact of formal institutions and their components on productivity spillovers from FDI enterprises to domestic firms’ TFP in developing countries like Vietnam. Design/methodology/approach: The study, conducted in two steps to explore the relationship, is to estimate the firm's TFP in accordance with the semi-parametric method of Levisohn and Petrin (2003). Regression is in accordance with the equation with panel data and adjusted by Driscoll and Kraay standard errors. An unbalanced panel data, related to more than 61,600 Vietnamese manufacturing firms from 2012 to 2017, is combined with the Provincial Institutional Quality Survey (PAPI index) and IO table. Findings: The local institutions have a positive impact on promoting learning ability and increasing productivity of domestic firms, especially small and medium enterprises (SMEs) that take better advantage of this effect. Vertical linkages with FDI enterprises assist local firms to increase productivity while horizontal linkages bring in negative effects. Domestic enterprises with high productivity (in the top 25%) receive positive spillover effects from horizontal linkages and vertical linkages and gain positive impacts of the institution on productivity whereas the group of low-productivity enterprises records negative impacts. This research highlights those enterprises operating in the region where institutions have transparency, accountability, participation in comments and effective corruption control can absorb spillovers and improve their productivity as well as the transparency and corruption control are recognized as having a positive impact through horizontal linkages. Research limitations/implications: With the limitation of research data being conducted only on manufacturing enterprises, there is a lack of data on the impact of service enterprises. The study only stops at understanding the impact of formal institutional effects on productivity spillovers whereas informal institutional effects will be studied in the future. Furthermore, the productivity spillovers of FDI enterprises are explored in general besides other FDI forms that will have different productivity spillovers like offshore. From the results of this study, the governments of developing countries should improve their institutions to encourage local enterprises to take the advantage of spillover effects from FDI enterprises as well as pay more attention to regional factors by supplementing development priority policies based on the capacity of each region. Institutional quality at provincial level has a positive impact on productivity spillovers; consequently, it is essential to have policies for further institutional improvement. Originality/value: This is the first research paper on the impact of the institutional factor at provincial level on firm’s productivity in developing countries like Vietnam. Theoretically, the impact of formal institutions on spillover effects from FDI enterprises is also clarified. In addition, our findings have implications for local economic development policies: vertical linkages promote domestic firms to increase their productivity while horizontal linkages of FDI enterprises in the same industry generate adverse impacts. This paper suggests some feasible solutions for SMEs in developing countries towards their productivity improvement.
