        Recombinant Human Granulocyte Colony-Stimulating Factor Promotes Preinvasive and Invasive Estrogen Receptor-Positive Tumor Development in MMTV-erbB2 Mice

        Purpose: We investigated whether recombinant human granulocyte colony-stimulating factor (rhG-CSF) could promote the development of preinvasive and invasive breast cancer in mouse mammary tumor virus (MMTV-erbB2) mice with estrogen receptor- positive tumors. Methods: MMTV-erbB2 mice were randomly divided into three experimental groups with 20 mice in each group. MMTV-erbB2 mice were treated with daily subcutaneous injections of vehicle or rhG-CSF (low-rhG-CSF group, rhG-CSF 0.125 μg; vehicle-rhG-CSF group, normal saline 0.25 μg; and high-rhG-CSF group, rhG-CSF 0.25 μg) at 3 months of age. Cellular and molecular mechanisms of G-CSF action in mammary glands were investigated via immunohistochemistry and reverse transcription polymerase chain reaction. Results: Low, but not high, rhG-CSF doses significantly accelerated mammary tumorigenesis in MMTV-erbB2 mice. Short-term treatment with rhG-CSF could significantly promote the development of preinvasive mammary lesions. The cancer prevention effect was associated with reduced expression of proliferating cell nuclear antigen, cluster of differentiation 34, and signal transducers and activators of transcription 3 in mammary glands by >80%. Conclusion: We found that G-CSF was regulated by rhG-CSF both in vitro and in vivo. Identification of G-CSF genes helped us further understand the mechanism by which G-CSF promotes cancer. Low doses of rhG-CSF could significantly increase tumor latency and increase tumor multiplicity and burden. Moreover, rhG-CSF effectively promotes development of both malignant and premalignant mammary lesions in MMTV-erbB2 mice.

        Glioma-Associated Oncogene Homolog1 (Gli1)-Aquaporin1 pathway promotes glioma cell metastasis

        Glioma-Associated Oncogene Homolog1 (Gli1) is known to be activated in malignant glioma; however, its downstream pathway has not been fully explained. The aim of this study was to explore the role of Gli1-Aquaporin1 (AQP1) signal pathway in glioma cell survival. Our data suggests that both Gli1 and AQP1 are upregulated in glioma tissues, as in comparison to in normal tissues. These up-regulation phenomena were also observed in glioma U251 and U87 cells. It was demonstrated that Gli1 positively regulated the AQP1 expression. By luciferase reporter gene and ChIP assay, we observed that this modulation process was realized by combination of Gli1 with AQP1 promotor. In addition, knock down of Gli1 by siRNA interference reduced the viability of glioma cells as well as suppressed cell metastasis. Also, the inhibitory effects of cell survival by silenced Gli1 were abrogated by AQP1 overexpression. In summary, glioma cell survival is a regulatory process and can be mediated by Gli1-AQP1 pathway. [BMB Reports 2016; 49(7): 394-399]

        Traditional Chinese medicine for COVID-19 pandemic and emerging challenges: An online cross-sectional survey in China

        Background We aimed to investigate use of infection control behaviours, preventative and therapeutic interventions, and outcomes among respondents to an online survey during the COVID-19 pandemic in China. Methods The survey was designed by an international team, translated and adapted to simplified Chinese, including 132 kinds of traditional Chinese medicine (TCM) preparation recommended by guidelines. It was distributed and collected from February to May 2021, with data analysed by WPS spreadsheet and wjx.cn. Descriptive statistics were used to describe demographics and clinical characteristics, diagnosis, treatments, preventative behaviours and interventions, and their associated outcomes. Results The survey was accessed 503 times with 341 (67.8%) completions covering 23 provinces and four municipalities in China. Most (282/341, 82.7%) respondents reported no symptoms during the pandemic and the majority (290/341, 85.0%) reported having a SARS-CoV-2 PCR test at some point. Forty-five (13.2%) reported having a respiratory infection, among which 19 (42.2%) took one or more categories of modern medicine, e.g. painkillers, antibiotics; 16 (35.6%) used TCM interventions(s); while seven respondents combined TCM with modern medicine. All respondents reported using at least one behavioural or medical approach to prevention, with 22.3% taking TCM and 5.3% taking modern medicines. No respondents reported having a critical condition related to COVID-19. Conclusion We found evidence of widespread use of infection control behaviours, modern medicines and TCM for treatment and prevention of COVID-19 and other respiratory symptoms. Larger scale studies are warranted, including a more representative sample exploring TCM preparations recommended in clinical guidelines.       • Lack of Association of Glutathione S-transferase M3 Gene Polymorphism with the Susceptibility of Lung Cancer

        Objective: The conclusions of published reports on the relationship between the glutathione S-transferase M3 (GSTM3) A/B gene polymorphism and the risk of lung cancer are still debated. This meta-analysis was performed to evaluate the association between GSTM3 and the risk of lung cancer. Methods: Association investigations were identified from PubMed, Embase, and Cochrane Library, and eligible studies were included and synthesized using a meta-analysis method. Results: Eight reports were included into this meta-analysis for the association of GSTM3 A/B gene polymorphism and lung cancer susceptibility, covering 1,854 patients with lung cancer and 1,926 controls. No association between the GSTM3 A/B gene polymorphism and lung cancer was found in this meta-analysis (B allele: OR = 1.25, 95% CI: 0.89-1.76, P = 0.20; BB genotype: OR = 1.53, 95% CI: 0.71-3.32, P = 0.28; AA genotype: OR = 0.85, 95% CI: 0.59-1.23, P = 0.39). Conclusions: The GSTM3 A/B gene polymorphism is not associated with lung cancer susceptibility. However, more studies on the relationship between GSTM3 A/B gene polymorphism and the risk of lung cancer should be performed in the future.

        Development of AFLP and SCAR Markers Linked to a Recessive Genic Male Sterile Gene (Bnms3) in Rapeseed for Marker-assisted Selection

        9012AB is a recessive genic male sterility (RGMS) line in rapeseed, of which the male sterility is controlled by two pairs of recessive duplicate male sterile genes (Bnms3 and Bnms4) interacting with a recessive epistatic suppressor gene (esp). The recessive homozygosity at the esp locus (espesp) can suppress the expression of the recessive male sterility trait in homozygous plants (Bnms3Bnms3Bnms4Bnms4) and result in fertility restoration. A F2 population of 188 plants, derived from self-pollinated progenies of a 9012 AB fertile plants (BnMs3Bnms3Bnms4Bnms4EspEsp), was conducted to identify molecular markers linked to the recessive male sterility gene (Bnms3). By amplified fragment length polymorphism (AFLP) assay combining with bulked segregant analysis (BSA), 13 markers linked to Bnms3 were identified. Linkage analysis indicated that 13 AFLP markers were tightly linked to the Bnms3 gene with a genetic distance varying from 1.3 cM to 7.1 cM. Among them, one marker was co-dominant marker, 6 markers were in coupling phase with Bnms3, and the others were in repulsion phase with Bnms3 gene, One AFLP marker with a genetic distance of 1.4cM was further converted into a SCAR marker successfully, which have been applied in marker-assisted selection of RGMS lines and their temporary maintainers effectively.

        Gene Mapping and Expression Analysis of a Novel Mutant reproduce organs absent (roa) in Rice

        Mutant plays an important role in function analysis in plant. A rice flower mutant reproduce organs absent (roa), showing a stable inheritance during several years of study, was identified in rice (Oryza sativa L. ssp. Indica) cultivar Jinhui10 treated with EMS and used in this study. This mutant showed following: elongated palea and pedicle; absence of inner three whorls of floral organs; multi-whorls glume like organs inside the lemma/palea; spikelet meristem like organ upon the pedicle. These phenotypes suggested that ROA is a key gene in rice spikelet development. Genetic analysis confirmed that the mutant traits were controlled by a single recessive nuclear gene. By gene mapping, ROA was restricted between two SSR markers RM221 and RM1342 on the chromosome 2. It concluded that ROA was a novel gene involving in flower development in rice. Besides, the mutation of ROA influenced the transcription level of floral homeotic genes; the expression of floral homeotic genes decreased in roa panicle compared with wild-type, and it suggested that ROA affected flower development by influencing the expression of floral homeotic genes.

        nonstop glumes (nsg), a novel mutant affects spikelet development in rice

        The spikelet is a unique structure of grass plants,and its development involved with complicated molecular regulation network. nsg (nonstop glumes) mutant affecting spikelet development was identified from EMS-treated Jinhui10(Oryza sativa L. ssp. indica.). Mutant plants had normal glumes (inner rudimentary glume, empty glumes and lemma/palea) and pedicel at the early flowering stage, but had longer ones at later stage. An extra glume-like organ was found in 84 %of mutant individuals. The number of stamens decreased in most mutant individuals whereas three stigmas or two carpels were found in some mutant individuals. The mutant phenotype suggests that NSG is involved in the whole rice spikelet development. NSG was mapped to a 15 kb region on the chromosome 4. According to sequence analysis, a gene encoding a protein with C2H2 domain exhibited a 13 bp insertion, causing a frame shift in genomics DNA and cDNA in nsg. This gene was identified as the candidated gene of NSG. The mutation of NSG influenced the transcription level of some floral hometic genes. The expression of OsMADS4, OsMADS16, DL and OsMADS3 decreased distinctly, and OsMADS1 increased in nsg panicle, suggests that NSG affected spikelet development through influencing the expression of floral hometic genes.

        A 2-40 Gb/s PAM4/NRZ Dual-mode Wireline Transmitter with 4:1 MUX in 65-nm CMOS

        This paper presents a 2-40 Gb/s dual-mode wireline transmitter supporting the four-level pulse amplitude modulation (PAM4) and non-return-tozero (NRZ) modulation with a multiplexer (MUX)-based two-tap feed-forward equalizer (FFE). An edgeacceleration technique is proposed for the 4:1 MUX to increase the bandwidth. By utilizing a dedicated cascode current source, the output swing can achieve 900 mV with a level deviation of only 0.12% for PAM4. Fabricated in a 65-nm CMOS process, the transmitter consumes 117 mW and 89 mW at 40 Gb/s in PAM4 and NRZ at 1.2 V supply.

        Identification of MicroRNAs as potential biomarkers for detecting ischemic stroke

        Background: Increasing epidemic of ischemic stroke (IS) makes it urgent to understand the pathogenesis and regulatory mechanism, previous studies have described microRNAs (miRNAs) is part of the brain's response to ischemia. Objective: The aim of this study was to screen potential biomarkers for the prediction and novel treatment of IS. Methods: Differentially expressed miRNAs were screened from three newly diagnosed IS patients and three controls by RNA sequencing technology. Furthermore, target prediction databases were then used to analysis the target genes of different expressed miRNAs, and the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway database were used to identify the functions and the main biochemical and signal pathways of differentially expressed target genes. Results: Our results revealed that 27 miRNAs were differentially expressed in IS, among which, hsa-miR-659-5p was the most highly increased and was first found to be associated with IS. In addition, KEGG pathway analyses showed that differentially expressed miRNAs were mainly significantly enriched in lysosome pathway, cytokine-cytokine receptor interaction pathway, spliceosome pathway, base excision repair pathway. Conclusions: miRNAs were involved in IS pathogenesis, and hsa-miR-659-5p, hsa-miR-151a-3p and hsa-miR-29c-5p as the three highest |log2FoldChange| regulation in this study, which may be the biomarkers of IS and need further study.
