Acquisition of a reference Tetranychusstrains that are completely susceptible to acaricides and retain identical genetic backgrounds to acaricide-resistant strains is an essential step in elucidating mechanisms of resistance. To establish both completely susceptible and acaricide-resistant strains for this purpose, I collected Tetranychus mite populations from various regions in South Korea including both heavily cultivated and remote regions. Suitability as a susceptible or resistant reference strain was tested by determining species identity as Tetranychus urticae along with baseline susceptibility to common acaricides. The majority of mite populations collected from cultivated areas belonged to a monophyletic group of the previously reported green-type T. urticae as determined by mtCOI- and ITS2-based phylogenetic analysis. These strains showed relatively reduced levels of susceptibility to the acaricides tested, suggestive of the development of resistance. Among them, the AbaR strain was classified as a minor group in the T. urticae complex. The UD strain, originally collected from a remote island region, was found to be susceptible to the acaricides tested and generated an independent phylogenetic branch. The UD strain was also considered a minor group in the T. urticae complex. Phenotypic analysis based on morphological characters confirmed that both the AbaR and UD strains were statistically undistinguishable from the major green-type T. urticae. Taken together, I propose that the UD strain be used as a susceptible reference strain as it provides baseline susceptibility to acaricides and a wild-type genetic background for the resistance studies.

Isaacsicalanus paucisetus Fleminger, 1983, a monotypic species of the family Spinocalanidae Vervoort, 1951, was first reported from a hydrothermal vent field in the East Pacific Rise off the mouth of the Gulf of California. The mitochondrial cytochrome oxidase I(mtCOI) DNA barcodes are considered a useful tool to assist traditional taxonomy and species discrimination in calanoid copepods. However, the mtCOI DNA barcodes of I. paucisetus have not been reported due to the species rarity and the difficulty of sampling. In this study, we firstly determined the mtCOI DNA barcodes of the I. paucisetus newly collected from a hydrothermal vent in the North Fiji Basin of the southwestern Pacific. All mtCOI DNA barcodes of I. paucisetus were identical and intraspecies variations of spinocalanid species were 0.0-3.0%. Interspecies and intergeneric variations were 13.4-25.2% and 16.7-24.1%, respectively. The DNA barcodes of I. paucisetus obtained in the present study would be helpful for understanding taxonomic relationships of widespread spinocalanid species.

벼 줄기속 가해 벼밤나방 유사 유충과 벼밤나방 성페로몬 트랩 포획 성충의 mtCOI 부분영역과 ITS2 영역 비교

서해안 지역 논에서 벼 줄기속을 가해하고 있는 벼밤나방 유사 유충과 논 주변에 설치한 벼밤나방 성페로몬 트랩에 포획된 벼밤나방 유사 성충으로부터 게놈 DNA 를 추출하고 미토콘드리아 시토크롬옥시다제I(mtCOI) 부분영역을 비교하였다. mtCOI 염기서열로부터 추론된 아미노산 서열의 경우 한 위치에서 국내 채집 나방 모두가 GenBank 데이터베이스에 등록되어 있는 중국산 벼밤나방 일부와 다른 아 미노산(Gly vs. Ser) 변이가 관찰되었다. 그리고 동일 지역 채집 나방에서도 일부 추론된 아미노산에서 변이가 있었다. 이러한 결과가 벼밤나방 종내변이 때문인지 다른 종이 섞여서 나타난 종간변이 때문인지는 앞으로 명확한 규명이 필요할 것으 로 보인다. 한편 종동정을 위한 참고자료로서 핵내 리보솜 RNA 유전자의 ITS2(rRNA internal transcribed spacer 2) 전체영역을 해독하고 비교하였다. 최근 우리나라에 발생하여 피해를 주고 있는 벼 줄기속 가해 나방의 종류를 밝히고 분포 지역과 생리·생태, 약제저항성 등에 관한 연구가 필요할 것으로 보인다.

Heterorhabdus papilliger (Claus, 1863) is newly reported from the Tsushima Warm Current realm of the southern Korean waters. Its morphological diagnostic characteristics generally agreed well with the original description and the previous records of H. papilliger. The female of H. papilliger can be recognized by the genital somite, which in lateral view has a more or less rounded genital prominence and an uninflated posterior ventral margin; the second exopodal segment of male right leg 5 with the medial projection with a large, rounded, plumose proximal lobe, and a poorly developed distal lobe. The genetic difference for the partial mtCOI gene between Korean specimens and H. papilliger from Spain and Japan of the same clade is 0.4%, while the difference between Korean specimens is 0.5%. However, the interspecific difference for the mtCOI gene between H. papilliger from the Korean waters and the other Heterorhabdus species is in the range of 14.7-20.8%, suggesting that the former is a valid species.

Two species, P. mictodonta (Marenzeller, 1879) and P. wilsoni (Glasby and Hsieh, 2006), are recognized and redescribed. In this study, mtCOI gene showed a good resolution as molecular marker for species identification of the P. nuntia species group of Korea.

미토콘드리아 COI DNA 영역의 뉴클레오티드 차이를 이용한 팥나방(Matsumuraeses phaseoli)과 어리팥나방(M. falcana)의 PCR 판별법

콩(Glycine max)과 팥(Vigna angularis) 등 콩과(Fabaceae) 작물 해충인 팥나방 (Matsumuraeses phaseoli)과 어리팥나방(M. falcana) (Lepidoptera: Tortricidae)은 형태적으로 매우 유사하여 종 구별이 힘든 것으로 알려져 있다. 본 연구에서는 PCR-SSP (PCR with Sequence Specific Primers) 방법으로 두 유사종을 빠르고 정확하게 구별할 수 있는 판별법을 찾고자 두 종의 미토콘드리아 시토크롬 옥시다아제 I (mtCOI) DNA 부분영역(439bp)의 염기서열을 해독하였다. 그리고 다른 나방 종의 mtCOI 염기서열과 함께 나열하여 비교한 후 팥나방과 어리팥나방에서 종 특이적으로 차이가 나는 단일 뉴클레오티드를 찾아 염기서열 특이 프라이머 조합을 만들고 PCR을 실시하였다. PCR 산물들을 전기영동 한 결과 어리팥나방은 245bp, 팥나방은 409bp와 245bp 사이즈의 밴드가 특이적으로 나타났으며 멸강나방과 왕담배나방에서는 밴드가 나오지 않았다. 이러한 결과로부터 본 연구에서 만든 염기서열 특이 프라이머 조합이 성페로몬트랩과 포장에서 잡힌 팥나방과 어리팥나방의 성충과 유충, 번데기의 종 구별을 위해 유용할 것으로 기대된다.

The tea mosquito bug, Helopeltis theivora Waterhouse (Miridae: Heteroptera) is a polyphagous pest that affects a number of economically important crops in India. Partial sequences of mitochondrial cytochrome c oxidase 1 (mtCOI) were used to study host associated genetic variation on H. theivora collected from tea (Camellia sinensis L.), cashew (Anacardiumoccidentale L.), acalypha (Acalypha indica L.) and cocoa (Theobroma cacao L.). The results suggest that host plants have a significant effect on the genetic variation of bugs collected from two wide geographical locations (South India and North-East India). Haplotyping data analysis of mtCOI reveals six host plant specific haplotypes of H. theivorawith minimumgenetic divergence (0.8%) between two haplotypeswithin tea as compared to other plants (2.6–11.1%). Further phylogenetic (NJ, ML and BA) analysis evidence three host specific distinct clades. The most surprising result that emerged from our studies is the harmony between clustering of H. theivora and host plant phylogeny. The results indicate that host plants do have selection pressure and substantial effect on genetic variation in H. theivora and such adaptive evolution may have repercussions for IPM (Integrated Pest Management) strategies of H. theivora in India.
Phylogenetic Analysis of Sweetpotato Whitefly Bemisia tabaci in Bangladesh

The sweetpotato whitefly Bemisia tabaci (Gennadius) is one of the most important sap sucking pests causing economic losses in a variety of vegetables in Bangladesh and as well as around the world. In the present study, the mtCOI sequence of B. tabaci was analysed using samples collected from different host plants (Potato, Brinjal, Tomato, Sweet potato, Bean) from district Gazipur, Patuakhali, Rajshahi and Nilphamari of Bangladesh. Phylogenetic analysis of our samples and relative sequences of B. tabaci in NCBI database was shown three independent clusters. Samples in Bangladesh were most similar with those of Thailand, Vietnam, Pakistan, China and India but did not show any B and Q aggressive biotypes.
