        Standardization of Acaricidal Bioassays for Developing Natural Acaricides against Three Mite Species (Arachnida: Acari)

        To establish a system of effective bioassays for natural acaricides against three mite species, influences of bioassay system, solvent drying time, and leaf-dipping time were investigated. Among the tested organic solvents, including diethyl ether, hexane, and acetone, acetone was found to be the most suitable for developing natural acaricides, whereas diethyl ether and hexane were not suitable. Solvent drying and leaf-dipping times of acetone were suitable at 10 min and 60 s, respectively. Application method of spraying (direct contact) bioassay was more effective than other methods. These results could be used in the standardization of bioassay systems for natural acaricides.

        Optimization of the microemulsion formulation of curcuma oil and evaluation of its acaricidal efficacy against Tetranychus cinnabarinus (Boisduval) (Acari: Tetranychidae)

        The microemulsion formulation (hereafter formulation) of curcuma oil and its acaricidal efficacy against Tetranychus cinnabarinus Boisduval (Acari: Tetranychidae) were optimized in the laboratory to evaluate their spray effectiveness of oviposition inhibition and repellence. Ethovision XT6 was used to analyse the effects of the sublethal concentrations (LC 20 ) of curcuma oil and the formulation on the behaviors of T. cinnabarinus. The results showed that Tween-80 was the best surfactant, Isopropanol was the best co-surfactant and K m = 2:1 was the best condition for the formulation. The prepared microemulsions are stable under conditions of centrifugation and incubation for extended periods. The results showed that the effect of the spray bioassays of the formulation against T. cinnabarinus continuously increased during the experiment, but for curcuma oil almost no longer increase observed when the exposure time went beyond 24 h. Moreover, compared with curcuma oil (LC 50 = 0.716%), the spray bioassay of the formulation (LC 50 = 0.035%) was stronger against T. cinnabarinus. The repellency of the formulation to T. cinnabarinus was stronger with increasing exposure time, but that of curcuma oil declined after 12 h of exposure. The mobile distance of T. cinnabarinus treated with the formulation continuously declined during the experiment but that due to the curcuma oil almost no longer declined when the treatment time reached 12 h. The maximum mobile frequency of T. cinnabarinus treated by curcuma oil and the formulation was recorded at 6 h and 12 h, respectively. Thus, the formulation is a promising candidate as a botanical acaricide of green vegetables.

        천연물 제초제 개발을 위한 전식물체 수준의 경엽처리 검정법 확립

        본 연구는 식물정유의 활용과 개발을 포함한 천연물 제초제의 개발을 효과적으로 추진하기 위한 방안으로서 식물정유 성분의 특성이 고려된 개선된 전식물체(whole-plant) 검정법을 확립하기 위해 수행되었다. 잡초 4초종을(피, 바랭이, 자귀풀, 어저귀) 이용하는 전식물체 경엽살포 검정법은 검정 화합물 량이 충분할 경우, 전식물체 생장점 점적처리법은 검정화합물이 미량일 때 추천할 수 있는 방법으로 판단되었다. 각 식물정유 처리시 초기 처리농도는 2,500~5,000 ㎍ mL-1 수준이 필요하였으며 계면활성제는 Tween 20 보다 NOP를 첨가하였을 경우에 보다 높은 살초활성을 나타내었다. 식물정유와 같이 휘발성이 있는 천연물의 활성을 평가하기 위해서는 약제를 처리한 후 과습 상태에 일정시간 두면 제초활성 발현이 증진되었다. 또한 전식물체 생장점 점적처리법은 경엽살포 검정법에 비해 약 1/100의 약제처리량(부피), 약 1/200의 성분량(무게)에서도 살초효과의 검정이 가능하였다. This study was carried out to establish an improved bioassay system, whole-plant bioassay which is more effective in developing natural herbicides for foliar treatment such as herbicidal essential oils. Two bioassay systems using four weed species (Echinochloa crus-galli, Digitaria sanguinalis, Aeschynomene indica, and Abutilon theophrasti), spraying method and spotting method, were established. Spraying method is applicable if the amount of test compounds is enough, while spotting method is useful for the small amount of test compounds. The initial application rate was desirable at 2,500~5,000㎍ mL-1. Herbicidal activities were higher in the NOP treatment when compared to the Tween 20 treatment. To efficiently evaluate volatile compounds such as essential oils, if the compound-treated pots were incubated in dew chamber for about 10hrs, better results were obtained in the degree and stability of herbicidal responses. When the efficiency of bioassay systems established in this study was compared, the spraying method was minimized four times to the conventional method that has beed used for screening of synthetic compounds in KRICT. On the other hand, in the spotting method, screening for development of a natural herbicides was possible even in level of 1/100 test volume and 1/200 amounts of test compound compared to the spraying method.

        The effects of petroleum oil and lime sulfur on the mortality of Unaspis yanonensis and Aculops pelekassi in the laboratory

        Arrowhead scale (AHS), Unaspis yanonensis (Kuwana) (Hemiptera: Diaspididae), and pink citrus rust mite (PCRM), Aculops pelekassi (Keifer) (Acari: Eriophyoidae), are important arthropod pests of organic citrus orchards in Jeju. This study was conducted to evaluate the stage-specific mortality effects of petroleum spray oil (PSO) (AI 95%) and lime sulfur (LSF) (AI 22%) against AHS and PCRM in the laboratory. The developmental stages of AHS were divided into the 1st nymph, the three sub-stages of the 2nd nymph (N2a=early soft body, N2b=hardened body, and N2c=late apolysis), and the two sub-stages of adult females (A3a=early age, and A3b=middle age). The developmental stages of PCRM were divided into eggs, the 2nd nymphs, and adults. PSO 100× and LSF 80× resulted in high mortality against the 1st nymphs of AHS. The effects of PSO and LSF were significantly different among the application rates on 2nd nymphs of AHS and were higher on N2a. The PSO and LSF treatments significantly affected egg hatch rate of PCRM: LSF 50× (2.4%), LSF 100× and PSO of 50× and 200× (17.8% to 20.4%), and LSF 200× (39.0%). PSO and LSF also significantly affected the mortality of the 2nd nymphs and adults of PCRM. PSO and LSF resulted in low direct mortality on A3a and A3b of AHS, but induced an abnormal morphology. PSO treatment resulted in a loose attachment of AHS scale cover on the leaf surface and LSF treatment resulted in an abnormal or lacking scale cover.
