        Portable Low-Cost MRI System Based on Permanent Magnets/Magnet Arrays

        Portable low-cost magnetic resonance imaging (MRI) systems have the potential to enable “point-of-care” and timely MRI diagnosis, and to make this imaging modality available to routine scans and to people in underdeveloped countries and areas. With simplicity, no maintenance, no power consumption, and low cost, permanent magnets/magnet arrays/magnet assemblies are attractive to be used as a source of static magnetic field to realize the portability and to lower the cost for an MRI scanner. However, when taking the canonical Fourier imaging approach and using linear gradient fields, homogeneous fields are required in a scanner, resulting in the facts that either a bulky magnet/magnet array is needed, or the imaging volume is too small to image an organ if the magnet/magnet array is scaled down to a portable size. Recently, with the progress on image reconstruction based on nonlinear gradient field, static field patterns without spatial linearity can be used as spatial encoding magnetic fields (SEMs) to encode MRI signals for imaging. As a result, the requirements for the homogeneity of the static field can be relaxed, which allows permanent magnets/magnet arrays with reduced sizes, reduced weight to image a bigger volume covering organs such as a head. It offers opportunities of constructing a truly portable low-cost MRI scanner. For this exciting potential application, permanent magnets/magnet arrays have attracted increased attention recently. A magnet/magnet array is strongly associated with the imaging volume of an MRI scanner, image reconstruction methods, and RF excitation and RF coils, etc. through field patterns and field homogeneity. This paper offers a review of permanent magnets and magnet arrays of different kinds, especially those that can be used for spatial encoding towards the development of a portable and low-cost MRI system. It is aimed to familiarize the readers with relevant knowledge, literature, and the latest updates of the development on permanent magnets and magnet arrays for MRI. Perspectives on and challenges of using a permanent magnet/magnet array to supply a patterned static magnetic field, which does not have spatial linearity nor high field homogeneity, for image reconstruction in a portable setup are discussed.

        Identification of seven novel virulence genes from Xanthomonas citri subsp. citri by Tn5-based random mutagenesis

        To identify novel virulence genes, a mutant library of Xanthomonas citri subsp. citri 29-1 was produced using EZ-Tn5 transposon and the mutants were inoculated into susceptible grapefruit. Forty mutants with altered virulence phenotypes were identified. Nine of the mutants showed a complete loss of citrus canker induction, and the other 31 mutants resulted in attenuated canker symptoms. Southern blot analysis revealed that each of the mutants carried a single copy of Tn5. The flanking sequence was identified by plasmid rescue and 18 different ORFs were identified in the genome sequence. Of these 18 ORFs, seven had not been previously associated with the virulence of X. citri subsp. citri and were therefore confirmed by complementation analysis. Real-time PCR analysis showed that the seven genes were upregulated when the bacteria were grown in citrus plants, suggesting that the expression of these genes was essential for canker development.

      • Survivin Gene Functions and Relationships between Expression and Prognosis in Patients with Nasopharyngeal Carcinoma

        This study aimed to investigate the relationship between prognosis and protein and mRNA expression of an apoptotic inhibitor gene, survivin, in patients with nasopharyngeal carcinoma. Furthermore, functions of the survivin gene in the CNE2 nasopharyngeal carcinoma cell line were assessed. Immunohistochemistry and in situ hybridization were used in detecting the survivin protein and mRNA in 44 nasopharyngeal carcinoma specimens, and 30 chronic nasopharyngitis samples as controls. Survivin gene expression in CNE2 cell line was suppressed with an shRNA (short hairpin RNA). The positive ratios of expression for survivin protein and mRNA in nasopharyngeal carcinoma were 79.5% and 75.0% respectively, obviously higher than in the control group (p<0.01), and there is very good consistency between the two methods. The mean survival time of patients with higher survivin protein or mRNA expression was shorter than in patients with lower levelsv(p<0.01). Proliferation of the CNE2 cell line was distinctly inhibited by the shRNA. The results indicate that overexpression of the survivin gene plays an important role in onset and development of nasopharyngeal carcinoma, and it may be helpful for prognostic appraisal.

        Protective effects of l-theanine on rats with dextran sulfate sodium-induced infl ammatory bowel disease

        The aim of this study is to evaluate the antiinflammatory and protective eff ects of L -theanine in infl ammatorybowel disease (IBD) and to identify the underlyingmolecular mechanisms. Rats were pre-treated with L -theanineat 0, 50, 200, or 800 mg/kg/day. IBD was induced inrats using dextran sulfate sodium (DSS). Histopathologicalanalysis suggests that L -theanine can suppress DSS-inducedIBD with signifi cant inhibition of infl ammation in large andsmall intestinal tissues. Moreover, the 200 mg/kg/day L -theanine-treated DSS group had higher body and small intestineweights, a lower disease activity index and expression ofinfl ammatory factors than the DSS group without pre-treatment. In RNA sequencing and tandem mass tag labelinganalyses, large number of mRNAs and proteins expressionlevel diff ered when compared with the DSS-induced ratswith and without 200 mg/kg/day L -theanine pre-treatment. Moreover, Kyoto Encyclopedia of Genes and Genomes pathwayanalysis indicates the anti-infl ammatory activities ofL -theanine in DSS-induced IBD, with a high representationof genes in “Cholesterol metabolism” and “Retinol metabolism”pathways. Analysis of protein–protein interaction networksfurther indicates the involvement of these two pathways. These studies suggest that medium-dose L -theaninepre-treatment could ameliorate DSS-induced IBD throughmolecular mechanisms involving cholesterol and retinolmetabolism.
