Ginsenoside Rg1 ameliorates Alzheimer's disease pathology via restoring mitophagy

Background: Alzheimer's disease (AD) is a common form of dementia, and impaired mitophagy is a hallmark of AD. Mitophagy is mitochondrial-specific autophagy. Ginsenosides from Ginseng involve in autophagy in cancer. Ginsenoside Rg1 (Rg1 hereafter), a single compound of Ginseng, has neuroprotective effects on AD. However, few studies have reported whether Rg1 can ameliorate AD pathology by regulating mitophagy. Methods: Human SH-SY5Y cell and a 5XFAD mouse model were used to investigate the effects of Rg1. Rg1 (1µM) was added to β-amyloid oligomer (AβO)-induced or APPswe-overexpressed cell models for 24 hours. 5XFAD mouse models were intraperitoneally injected with Rg1 (10 mg/kg/d) for 30 days. Expression levels of mitophagy-related markers were analyzed by western blot and immunofluorescent staining. Cognitive function was assessed by Morris water maze. Mitophagic events were observed using transmission electron microscopy, western blot, and immunofluorescent staining from mouse hippocampus. The activation of the PINK1/Parkin pathway was examined using an immunoprecipitation assay. Results: Rg1 could restore mitophagy and ameliorate memory deficits in the AD cellular and/or mouse model through the PINK1-Parkin pathway. Moreover, Rg1 might induce microglial phagocytosis to reduce β-amyloid (Aβ) deposits in the hippocampus of AD mice. Conclusion: Our studies demonstrate the neuroprotective mechanism of ginsenoside Rg1 in AD models. Rg1 induces PINK-Parkin mediated mitophagy and ameliorates memory deficits in 5XFAD mouse models.
Active control of amplitude and phase of high-power RF systems in EAST ICRF heating experiments

The EAST ICRF system operating space has been extended in power and phase control with a low-level RF system for the new double-strap antenna. Then the multi-step power and periodic phase scanning experiment were conducted in L-mode plasma, respectively. In the power scanning experiment, the stored energy, radiation power, plasma impedance and the antenna's temperature all have positive responses during the short ramp-ups of PL;ICRF. The core ion temperature increased from 1 keV to 1.5 keV and the core heating area expanded from jZj 5 cmto jZj 10 cm during the injection of ICRF waves. In the phasing scanning experiment, in addition to the same conclusions as the previous relatively phasing scanning experiment, the superposition effect of the fluctuation of stored energy, radiation power and neutron yield caused by phasing change with dual antenna, resulting in the amplitude and phase shift, was also observed. The active control of RF output facilitates the precise control of plasma profiles and greatly benefits future experimental exploration.

This paper presented an improved analytical method for calculating the open-circuit magnetic field in the surface-mounted permanent magnet machines accounting for slots and eccentric magnet pole. Magnetic field produced by radial and parallel permanent magnet is equivalent to that produced by surface current according to equivalent surface-current method of permanent magnet. The model is divided into two types of subdomains. The field solution of each subdomain is obtained by applying the interface and boundary conditions. The magnet field produced by equivalent surface current is superposed according to superposition principle of vector potential. The investigation shows harmonic contents of radial flux density can be reduced a lot by changing eccentric distance of eccentric magnet poles compared with conventional surface-mounted permanent-magnet machines with concentric magnet poles. The FE(finite element) results confirm the validity of the analytical results with the proposed model.

This paper presented an improved analytical method for calculating armature-reaction field in the surface-mounted permanent magnet machines accounting for opening slots. The analytical model is divided into two types of subdomains. The current of the armature is centralized in the center of the slots. The field solution of each subdomain is obtained by applying the interface and boundary conditions of the model. Two 30-pole/36-slot prototype machines with different slot-opening width are used for validation. The FE (finite element) results confirm the validity of the analytical results with the proposed model. The investigation shows that the wider the slot-opening width is, the smaller the peak value of radial and circumferential components of flux density, and the analytical armaturereaction field produced by centralized current in the slots is similar with the armature-reaction field produced by distributed current in the slots in the FE.
