Static and fatigue behavior of through-bolt shear connectors with prefabricated HFRC slabs

Twelve push-out test specimens were conducted with various parameters to study the static and fatigue performance of a new through-bolt shear connector transferring the shear forces of interface between prefabricated hybrid fiber reinforced concrete (HFRC) slabs and steel girders. It was found that the fibers could improve the fatigue life, capacity and initial stiffness of through-bolt shear connector. While the bolt-hole clearance reduced, the initial stiffness, capacity and slippage of through-bolt shear connector increased. After the steel-concrete interface properties were improved, the initial stiffness increased, and the capacity and slippage reduced. Base on the test results, the equation of the load-slip curve and capacity of through-bolt shear connector with prefabricated HFRC slab were obtained by the regression of test results, and the allowable range of shear force under fatigue load was recommended, which could provide the reference in the design of through-bolt shear connector with prefabricated HFRC slabs.

Autophagy is crucial for immune defense against Mycobacterium tuberculosis (Mtb) infection. Mtb can evade host immune attack and survival within macrophages by manipulating the autophagic process. MicroRNAs (miRNAs) are small, non-coding RNAs that are involved in regulating vital genes during Mtb infection. The precise role of miRNAs in autophagy with the exits of Mtb remains largely unknown. In this study, we found miR-1958, a new miRNA that could regulate autophagy by interacting with 3'UTR of autophagy-related gene 5 (Atg5). In addition, Mtb infection triggered miR-1958 expression in RAW264.7 cells. What's more, miR- 1958 overexpression blocked autophagic flux by impairing the fusion of autophagosomes and lysosomes. Overexpression of miR-1958 reduced Atg5 expression and LC3 puncta while inhibition of miR-1958 brought an increase of Atg5 and LC3 puncta; the opposite results were observed in detection of p62. The survival of Mtb in RAW264.7 cells transfected with mimic of miR-1958 was enhanced. Taken together, our research demonstrated that a novel miR-1958 could inhibit autophagy by interacting with Atg5 and favored intracellular Mtb survival in RAW264.7 cells.
Expression Vectors for Human-mouse Chimeric Antibodies

The production of recombinant antibodies has been generally recognized as time-consuming and labor-intensive. The aim of our study is to construct mammalian expression vectors containing the cDNA encoding the human constant regions and murine variable regions to massively and cost-effectively produce full-length chimeric antibodies. Unique restriction sites flanking the Ig variable region were designed to allow for the replacement of variable regions generated by PCR. Western blot analysis of the chimeric antibodies revealed that the expressed products were of the predicted size, structure and specificity. The usefulness of the vectors was confirmed by construction of human-mouse chimeric antibody-HCAb which secretes murine antibody against the human colorectal cancer. Selected in medium containing gradually increasing methotrexate (MTX), clones with increased expression of the product gene can be efficiently generated. The secretion of recombinant chimeric antibody-HCAb yielded $30\;pg\;cell^{-1}\;day^{-1}$ at $10^{-6}\;M$ MTX. With this high-level expression from pools, the convenient and rapid production of over 100 milligram amounts per liter of recombinant antibodies may be achieved, which indicates the significant roles of pYR-GCEVH and pYR-GCEVL in the production of chimeric antibodies.
Co-simulation of self-propelled artillery based on virtual prototype technology

In present study, a virtual prototype of self-propelled artillery was constructed based on virtual prototype technology, and the whole impact process of artillery firing simulation was completed using ADAMS and AMEsim software. Results showed that the constructed artillery firing simulation system was reasonable. It could add a recoil dynamic response suitable for live firing to the tested artillery and could detect the vibration response characteristics of the artillery through the test device. Through numerical simulation, a quantitative analysis of artillery firing stability, stress state, and fatigue life under different working conditions was performed. All simulation results were in good agreement with the real situation. This research provides a certain reference for the evaluation of artillery firing stability and lays a good theoretical foundation for the engineering development and practical application of the test device.

In this study, an efficient transformation system for the medicinal plant Anisodus acutangulus was successfully developed and optimized using Agrobacterium rhizogenes. Three bacterial strains, A4, R1601, and modified C58C1 and three explant types, leaf blade, petiole, and stem, were examined. The highest transformation efficiency of 94.44% was achieved using strain C58C1 with stem explants. Over 20 independent hairy root lines were successfully established with strain C58C1 us-ing stem explants, all of which contained the rolB and rolC genes as confirmed by polymerase chain reaction (PCR). Out of four media compositions, the liquid 1/2 MS medium was found the most suitable for hairy root growth. The maximum bio-mass of one hairy root line increased up to 80 times in liquid 1/2 MS medium after a 30 day culture period. Different hairy root lines displayed a varied capacity for tropane alkaloid production and the best hairy root line (T4) from the C58C1-stem combination produced up to 10.21 mg/g (dw) of hyoscyamine, which was about 1.5-fold higher than in the wild type plants. To our knowledge, this is the first report to demonstrate the production of tropane alkaloids in hairy roots of A. acutangulus.
Nanophase ZnV2O4 as stable and high capacity Li insertion electrode for Li-ion battery

Spinel ZnV2O4 nanoparticles are synthesized by a hydrothermal method and its properties are characterized using XRD, SEM, TEM, and electrochemical test. The structural and morphological characterizations show that ZnV2O4 sample has high purity and well crystallization with crystal size less than 20 nm. The as prepared electrode shows stable capacity over 660 mAh g-1 in the voltage range of 0.01-3.0 V at 50 mA g-1. The reaction mechanism with lithium ion is also investigated through ex-XRD and -TEM. It shows that the pristine ZnV2O4 is transformed to isostructural spinel LixV2O4 (x close to 7.6) and metal Zn phase during the first lithiation process. Then the spinel LixV2O4 seems to perform a topotactic intercalation reaction mechanism and that the in-situ formed LixV2O4 can still keep its spinel matrix while allowing more than 5.7 lithium reversibly into/out over 50 cycles.
Influence of Screw Rotors Tip Angle on Mixing Performance for One Novel Twin-screw Kneader

Twin-screw kneader is an efficient polymer processing equipment. In this paper, the mixing performance of one novel intermeshing counter-rotating twin-screw kneader with different tip angles of the male rotor is simulated using the mesh superimposition technique (MST). Statistical analysis is carried out for the flow field using particle tracking technique, and distributive mixing performance is evaluated using the residence time distribution and segregation scale, while the dispersive mixing performance is estimated using the parameters such as shear rate, stretching rate and mixing index. The results show that the best distributive mixing performance is achieved when the tip angle is 0˚, while the optimal dispersive mixing performance is obtained when the tip angle is 20˚. The results in this paper provide a data basis for the selection of parameters and optimization of the performance for the screw rotors.

Stacked-nanoflake Li4Ti5O12 spinel was synthesized via the pyrolysis of a LieTi copolymeric precursor formed by in situ polymerization of LiOH and [Ti(OC4H9)4] and acrylic acid. XRD and SEM characterization shows that the powders calcined at 700 C for 3 h was well-crystallized particles with submicron diameter. Chargeedischarge measurement showed the Li4Ti5O12 electrode had displayed excellent rate capability and delivered reversible capacity of 171, 158, 148, 138 and 99 mAh g1 at rates of 0.1C, 0.5C, 1C, 2C and 4C, respectively. The test electrode also showed excellent cyclability as the capacity retains 96.1% after 60 cycles between 0.5 and 2.5 V.
