      • A Comparison of Friction Force Calibration in Lateral Force Microscope

        The main principle of two widely used methods which were proposed by Ruan and Bhushan, and by Ogletree and Carpick are introduced. Experiments were conducted using the two methods to measure friction force between AFM probe and silicon sample quantitatively. To characterize the frictional properties, the conversion factors of the two methods by which lateral electronic signal is converted into actual friction force were calculated. The experimental results show that that the conversion factors were extraordinarily different from each other. Further research should be done to identify the reasons for these differences.

        Novel miR-1958 Promotes Mycobacterium tuberculosis Survival in RAW264.7 Cells by Inhibiting Autophagy Via Atg5

        Autophagy is crucial for immune defense against Mycobacterium tuberculosis (Mtb) infection. Mtb can evade host immune attack and survival within macrophages by manipulating the autophagic process. MicroRNAs (miRNAs) are small, non-coding RNAs that are involved in regulating vital genes during Mtb infection. The precise role of miRNAs in autophagy with the exits of Mtb remains largely unknown. In this study, we found miR-1958, a new miRNA that could regulate autophagy by interacting with 3'UTR of autophagy-related gene 5 (Atg5). In addition, Mtb infection triggered miR-1958 expression in RAW264.7 cells. What's more, miR- 1958 overexpression blocked autophagic flux by impairing the fusion of autophagosomes and lysosomes. Overexpression of miR-1958 reduced Atg5 expression and LC3 puncta while inhibition of miR-1958 brought an increase of Atg5 and LC3 puncta; the opposite results were observed in detection of p62. The survival of Mtb in RAW264.7 cells transfected with mimic of miR-1958 was enhanced. Taken together, our research demonstrated that a novel miR-1958 could inhibit autophagy by interacting with Atg5 and favored intracellular Mtb survival in RAW264.7 cells.

        Axial compression ratio limit values for steel reinforced concrete (SRC) special shaped columns

        This paper presents the results of experimental investigation, numerical calculation and theoretical analysis on axial compression ratio limit values for steel reinforced concrete (SRC) special shaped columns. 17 specimens were firstly intensively carried out to investigate the hysteretic behavior of SRC special shaped columns subjected to a constant axial load and cyclic reversed loads. Two theories were used to calculate the limits of axial compression ratio for all the specimens, including the balanced failure theory and superposition theory. It was found that the results of balanced failure theory by numerical integration method cannot conform the reality of test results, while the calculation results by employing the superposition theory can agree well with the test results. On the basis of superposition theory, the design limit values of axial compression ratio under different seismic grades were proposed for SRC special shaped columns.

        Seismic behavior of T-shaped steel reinforced high strength concrete short-limb shear walls under low cyclic reversed loading

        This paper presents an experimental study of six steel reinforced high strength concrete T-shaped short-limb shear walls configured with T-shaped steel truss under low cyclic reversed loading. Considering different categories of ratios of wall limb height to thickness, shear/span ratios, axial compression ratios and stirrup reinforcement ratios were selected to investigate the seismic behavior (strength, stiffness, energy dissipation capacity, ductility and deformation characteristics) of all the specimens. Two different failure modes were observed during the tests, including the flexural-shear failure for specimens with large shear/span ratio and the shear-diagonal compressive failure for specimens with small shear/span ratio. On the basis of requirement of Chinese seismic code, the deformation performance for all the specimens could not meet the level of ‘three’ fortification goals. Recommendations for improving the structural deformation capacity of T-shaped steel reinforced high strength concrete short-limb shear wall were proposed. Based on the experimental observations, the mechanical analysis models for concrete cracking strength and shear strength were derived using the equivalence principle and superposition theory, respectively. As a result, the proposed method in this paper was verified by the test results, and the experimental results agreed well with the proposed model.

        Autophagy of bovine mammary epithelial cell induced by intracellular Staphylococcus aureus

        Bovine mastitis is a common disease in the dairy industry that causes great economic losses. As the primary pathogen of contagious mastitis, Staphylococcus aureus (S. aureus) can invade bovine mammary epithelial cells, thus evading immune defenses and resulting in persistent infection. Recently, autophagy has been considered an important mechanism for host cells to clear intracellular pathogens. In the current study, autophagy caused by S. aureus was detected, and the correlation between autophagy and intracellular S. aureus survival was assessed. First, a model of intracellular S. aureus infection was established. Then, the autophagy of MAC-T cells was evaluated by confocal microscopy and western blot. Moreover, the activation of the PI3K-Akt-mTOR and ERK1/2 signaling pathways was determined by western blot. Finally, the relationship between intracellular bacteria and autophagy was analyzed by using autophagy regulators (3-methyladenine [3-MA], rapamycin [Rapa] and chloroquine [CQ]). The results showed that S. aureus caused obvious induction of autophagosome formation, transformation of LC3I/II, and degradation of p62/SQSTM1 in MAC-T cells; furthermore, the PI3K-Akt-mTOR and ERK1/2 signaling pathways were activated. The number of intracellular S. aureus increased significantly with autophagy activation by rapamycin, whereas the number decreased when the autophagy flux was inhibited by chloroquine. Therefore, this study indicated that intracellular S. aureus can induce autophagy and utilize it to survive in bovine mammary epithelial cells.
