        The structural and non–linear dynamic analysis for radioactive waste container

        In recent years, the development of radioactive waste containers for nuclear facility decommissioning and dismantling is a critical issue because the Taiwan domestic boiling water reactor nuclear power plant is going to be decommissioned. The main purpose of this research is to design a metal container that meets the structural requirements of related regulations. At first, the shielding analysis was performed by varying dimensions of radioactive waste to determine the storage efficiency of the container. Then, a series of structural analyses for operational and accidental conditions of the container with full load were conducted, such as lifting, stacking, and drop impact conditions. On the other hand, the field drop impact tests were carried out to ensure structural integrity. The present research demonstrates the structural safety of the developed container for decommissioned nuclear facilities in Taiwan.

      • Aberrant Methylation of RASSF1A gene Contribute to the Risk of Renal Cell Carcinoma: a Meta-Analysis

        The aim of this study was to assess the diagnostic value of RASSF1A methylation in renal cell carcinoma. Systematically search were performed using the Pubmed, ProQest and Web of Science for all articles on the association between RASSF1A methylation and renal cell carcinoma before 15 April 2015. After the filtration, 13 studies involving 677 cases and 497 controls met our criteria. Our meta-analysis suggested that hypermethylation of RASSF1A gene was associated with the increased risk of RCC(OR:4.14, 95%CI:1.06-16.1). Stratified analyses showed a similar risk in qualitative detection method(OR:28.4, 95%CI:10.2-79.6), body fluid sample(OR:12.8, 95%CI:5.35-30.8), and American(OR:10.5, 95%CI:1.97-55.9). Our result identified that RASSF1A methylation had a strong potential in prediction the risk of Renal cell carcinoma.

        Differential Diagnosis of Brain Diseases Using In Vivo Proton Magnetic Resonance Spectroscopy at 3 Tesla: A Preliminary Study

        The purpose of this study was to evaluate the usefulness of in vivo 3T $^1H$ MRS with short TE for prescreening various brain diseases. Together with ten normal volunteers, 12 brain tumor patients(2 lymphomas, 5 malignant gliomas) and 5(benign meningiomas) and 10 brain ischemic disease patients(6 acute and 4 subacute infarctions) participated. Lymphomas showed increased intensities of Cho and Lac. Likewise, gliomas showed increased Cho and Lac, but with decreased NAA and ${\beta}\;{\gamma}$-Glx; in higher grade of gliomas, Lac, Cho, mI and Lip predominantly increased with decrease of NAA. Benign meningiomas showed increased Cho, Lac and ${\beta}\;{\gamma}$-Glx; with decreased of NAA. The alanine peak at 1.47 ppm is a neuronal marker for meningiomas. Infarctions showed increased Lac and Lip and decreased NAA, ${\alpha}$-Glx and ${\beta}\;{\gamma}$-Glx where Lac increased with decreased of ${\alpha}$-Glx in acute, and Cho, Lac and Lip increased with decrease of NAA in subacute. Elevated Lac and decreased NAA levels were more aggravated in subacute. Clinical application of the $^1H$ MRS with short TE at 3T is able to povide valuable spectral information for prescreening various brain diseases by monitoring the changes of disease-specific cerebral metabolite concentrations in vivo, and consequently, it can be applicable to assessment of differential diagnosis and malignancy as well.

        Biocatalysis and Bioprocess Engineering : Overproduction of the Escherichia coli Chaperones GroEL-GroES in Rhodococcus ruber Improves the Activity and Stability of Cell Catalysts Harboring a Nitrile Hydratase

        Three combinations of molecular chaperones from Escherichia coli (i.e., DnaK-DnaJ-GrpEGroEL- GroES, GroEL-GroES, and DnaK-DnaJ-GrpE) were overproduced in E. coli BL21, and their in vitro stabilizing effects on a nitrile hydratase (NHase) were assessed. The optimal gene combination, E. coli groEL-groES (ecgroEL-ES), was introduced into Rhodococcus ruber TH3. A novel engineered strain, R. ruber TH3G was constructed with the native NHase gene on its chromosome and the heterologous ecgroEL-ES genes in a shuttle plasmid. In R. ruber TH3G, NHase activity was enhanced 37.3% compared with the control, TH3. The in vivo stabilizing effect of ecGroEL-ES on the NHase was assessed using both acrylamide immersion and heat shock experiments. The inactivation behavior of the in vivo NHase after immersion in a solution of dynamically increased concentrations of acrylamide was particularly evident. When the acrylamide concentration was increased to 500 g/l (50%), the remaining NHase activity in TH3G was 38%, but in TH3, activity was reduced to 10%. Reactivation of the in vivo NHases after varying degrees of inactivation was further assessed. The activity of the reactivated NHase was more than 2-fold greater in TH3G than in TH3. The hydration synthesis of acrylamide catalyzed by the in vivo NHase was performed with continuous acrylonitrile feeding. The final concentration of acrylamide was 640 g/l when catalyzed by TH3G, compared with 490 g/l acrylamide by TH3. This study is the first to show that the chaperones ecGroEL-ES work well in Rhodococcus and simultaneously possess protein-folding assistance functions and the ability to stabilize and reactivate the native NHases.

        Institutional resilience of Taiwan’s semi-presidential system: the integration of the president and premier under party politics

        Over the past three decades, the semi-presidentialism has been adopted in most new democracies. It is also the constitutional order in most democracies, which can be divided into three categories: established, post-Leninist, and postcolonial democracies. Semi-presidentialism is a political system with dual executive branches. Moreover, because of these dual executive branches, the constitutional order of semi-presidentialism might be similar to a presidential system if the president is the de facto head of government. It might also be similar to a parliamentary system if the prime minister is the de facto head of government with the support of the parliamentary majority. Taiwan has been considered as a semi-presidential country since 1997. According to Taiwan’s constitutional amendments, its president is directly elected, and the premier (prime minister) and cabinet are responsible to the legislature. Dual executive system in Taiwan has been effective and flexible. I attempt to examine the institutional resilience of Taiwan’s constitutional function, which means that, in properly responding to social movements, its government has continued to function well. In addition, the president has been able to continue serving in office in spite of political crises. A vague constitutional design and a presidentialized party system are two reasons for this.

      • Clinical outcomes on tension-releasing suture appendage on single-incision sling devices for postoperative voiding dysfunction involving undue tape tension

        Objective: To determine the clinical outcomes of adding tension releasing suture(TRS) appendage for manipulation of over-tensioned single-incision slings(SIS) as a means to relieve post-operative voiding dysfunction. Material and Method: This was a retrospective case series study conducted in the in a tertiary referral center from January 2010 to July 2017 after gaining Institutional Review Board approval. The records of patients with urodynamic stress incontinence(USI) without needing concurrent procedures that underwent anti-incontinence surgery using MiniArc, Solyx, and Ajust with voiding dysfunction were collated and analyzed. The primary outcome measure was the recovery of normal post-void residual urine(PVR) after TRS manipulation. The secondary outcome measures were the pain intensity noted during manipulation (quantified through Visual Analog Scale), and the continence rate (assessed through 1. Objective cure- 1-hour pad test weight <2g and absence of USI; and 2. Subjective cure- index score of <1 on question #3 on UDI-6: "Urine leakage related to physical activity, coughing, or sneezing?"). Results: There were 73 patients with high post-void residual (PVR) urine. The forty-two(9.5%) patients with over-tensioned sling were managed with TRS manipulation while the 31 patients(7%) with high PVR and no sling over-tension were managed with intermittent catheterization. All patients in both groups regained normal PVR. The TRS manipulated group demonstrated an objective cure of 92.9%(39/42) and subjective cure of 91%(38/42). Pain experienced during TRS manipulation significantly increased with the Ajust system (p=0.018). There were 3 patients who had persistence of USI, 2 from MiniArc and 1 from Solyx. Conclusion: The TRS manipulation is a well-tolerated procedure that can effectively relieve voiding dysfunction for over-tensioned SIS without affecting continence cure rates.

        Molecular cloning and expression analysis of the first two key genes through 2-C-methyl-D-erythritol 4-phosphate (MEP) pathway from Pyropia haitanensis(Bangiales, Rhodophyta)

        Pyropia haitanensis (T. J. Chang et B. F. Zheng) N. Kikuchi et M. Miyata is one of the most commercially usefulmacroalgae cultivated in southeastern China. In red algae, the biosynthesis of terpenoids through 2-C-methyl-Derythritol4-phosphate (MEP) pathway can produce a direct influence on the synthesis of many biologically importantmetabolites. In this study, two genes of cDNAs, 1-deoxy-D-xylulose-5-phosphate synthase (DXS) and 1-deoxy-Dxylulose-5-phosphatereductase (DXR), which encoding the first two rate-limiting enzymes among MEP pathway werecloned from P. haitanensis. The cDNAs of P. haitanensis DXS (PhDXS) and DXR (PhDXR) both contained complete openreading frames encoding polypeptides of 764 and 426 amino acids residues, separately. The expression analysis showedthat PhDXS was significant differently expressed between leafy thallus and conchocelis as PhDXR been non-significant. Additionally, expression of PhDXR and its downstream gene geranylgeranyl diphosphate synthase were both inhibitedby fosmidomycin significantly. Meanwhile, we constructed types of phylogenetic trees through different algae and higherplants DXS and DXR encoding amino acid sequences, as a result we found tree clustering consequences basically in linewith the "Cavalier-Smith endosymbiotic theory." Whereupon, we speculated that in red algae, there existed only completeMEP pathway to meet needs of terpenoids synthesis for themselves; Terpenoids synthesis of red algae derivativesthrough mevalonate pathway came from two or more times endosymbiosis of heterotrophic eukaryotic parasitifer. Thisstudy demonstrated that PhDXS and PhDXR could play significant roles in terpenoids biosynthesis at molecular levels. Meanwhile, as nuclear genes among MEP pathway, PhDXS and PhDXR could provide a new way of thinking to researchthe problem of chromalveolata biological evolution.

      • Facile synthesis of Nafion-supported Pt nanoparticles with ultra-low loading as a high-performance electrocatalyst for hydrogen evolution reaction

        <P><B>Abstract</B></P> <P>x%Pt-Naf-CV (Pt-Nafion-Cyclic Voltammetry) catalysts with homogeneously distributed platinum nanoparticles and ultra-low Pt loading are successfully synthesized by using a facile potential cycling approach. The as-synthesized 0.8%Pt-Naf-CV catalyst exhibits an enhanced electrocatalytic activity for hydrogen evolution reaction (HER) in 0.5 M H<SUB>2</SUB>SO<SUB>4</SUB> solution, which obtains a low overpotential of 34 mV at 10 mA cm<SUP>−2</SUP>. The linear sweep voltammetry (LSV) curve of 0.8%Pt-Naf-CV catalyst is almost consistent with that of commercial Pt/C. However, the 0.8%Pt-Naf-CV catalyst displays a more excellent stability and durability in comparison with commercial Pt/C. Besides, the Pt loading of Pt/C (Pt–10 wt%) is about 10 times that of 0.8%Pt-Naf-CV catalyst. The improved electrocatalytic performances are derived from the synergistic effects of Pt and Nafion. The Nafion plays a significant role as a dispersant, carrier and structure directing agent on the morphology and size of the Pt catalyst. This result contributes a promising method to enhance the catalytic activity and reduce the amount of Pt.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        Seamless Transition Strategy for Wide Speed-Range Sensorless IPMSM Drives with a Virtual Q-axis Inductance

        Hybrid rotor position estimation methods that integrate a fundamental model and high frequency (HF) signal injection are widely used for the wide speed-range sensorless control of interior permanent-magnet synchronous machines (IPMSMs). However, the direct transition of two different schemes may lead to system fluctuations or system instability since two estimated rotor positions based on two different schemes are always unequal due to the effects of parameter variations, system delays and inverter nonlinearities. In order to avoid these problems, a seamless transition strategy to define and construct a virtual q-axis inductance is proposed in this paper. With the proposed seamless transition strategy, an estimated rotor position based on a fundamental model is forced to track that based on HF signal injection before the transition by adjusting the constructed virtual q-axis inductance. Meanwhile, considering that the virtual q-axis inductance changes with rotor position estimation errors, a new observer with a two-phase phase-locked loop (TP-PLL) is developed to accurately obtain the virtual q-axis inductance online. Furthermore, IPMSM sensorless control with maximum torque per ampere (MTPA) operations can be tracked automatically by selecting the proper virtual q-axis inductance. Finally, experimental results obtained from an IPMSM demonstrate the feasibility of the proposed seamless transition strategy.
