사람에 있어서 免疫담당세포의 하나인 T 細胞는 몇몇 subpooulation으로 나누어지고 있으며, 그중 ?? 및 ??를 同定하는 수단으로 사용되는 牛赤血球抗體중에서 우선 純粹 IgG抗體를 分離, 精裂하였으며, 이 精裂된 IgG抗體는 標準製品과의 比較實驗에서 ?? 細胞의 一致되는 成積을 얻을 수 있었다. Antisera to ox red blood cell were prepared by intraperitioneal hypermultiple injections without adjuvant in outbred white rabits. Purified IgG fraction from these rabbits anti-ox red blood cell antiserum for T subset assay was obtained by precipitation with 50% saturated ammonium sulphate followed by DEAE-cellulose chromatography and Sephadex G-150 gel filtration. These purified IgG fraction was compared with Cappel company standard IgG fraction for ?? subpopulation assay. We used home-made IgG fraction and obtained favorable results in ?? subpopulation assay as Cappel company standard IgG fraction.

For production of tetanus toxoid(TT) specific human monoclonal antibodies, anti-TT antibody secreting peripheral B lymphocytes were separated by rosetting with TT-coated SRBC. And, B lymphoblastoid cell line(BLCL) was established by Epstein-Barr Virus(EBV) transformation methods. Stable BLCLs were established after three times of cloning by limiting dilution. To establish the maximal antibody producing condition, several environmental factors were tested. The optimal condition for maximum antibody prodiction was 4 day culture of 1 x 105cells/ml concentration in RPMI 1640 complete culture media supplemented with 20% fetal calf serum, and the maximum concentration of secreted antibody was about 900ng/ml. The antibody production of BLCL was decreased during long-term culture after establishment of cell line, but antibody production was maintained by repeated cloning by limiting dilution.
韓國正常人의 末稍血液淋巴球의 Natural Kiier(NK)活性値에 關한 硏究

Spontaneous human lymphocyte-mediated cytotoxicity(SLMC) against target of cultured myeloid cell(K562) was examined in healthy subjects of Korean. Lymphocyte-mediated natural cytotoxicity against target cell was assayed by using a ?? release microcytotoxicity system. These assay system involve reactivity in 37℃, CO₂ contained incubator between effector cell(PBL) and target cell, K562 at 40 to 1 of L/T ratio. The SLMC of lymphocytes from healthy donors were compared between the male and female. The result showed differences of certain degree in activity in both group. In our study, the results of NK activity in male and female are definitely different. This results was consistent with other investigators.
Case Report : Primary Squamous Cell Carcinoma of the Liver Initially Presenting with Pseudoachalasia

Pseudoachalasia secondary to primary squamous cell carcinoma (SCC) of the liver is extremely rare and has not been reported until now. Here, we report a unique case of primary SCC of the liver initially presenting with progressive dysphagia along with short periods of signifi cant weight loss. A 58-yearold man initially presented with progressive dysphagia along with significant weight loss over brief periods of time. The radiographic and manometric fi ndings were consistent with achalasia. Subsequent esophagogastroduodenoscopy revealed a moderately dilated esophagus without evidence of neoplasm or organic obstruction. However, firm resistance was encountered while traversing the esophagogastric junction (EGJ), although no mucosal lesion was identifi ed. Due to the clinical suspicion of the presence of a malignant tumor, endoscopic ultrasonography (EUS) and computed tomography scans of the chest and abdomen were obtained. A huge hepatic mass with irregular margins extending to the EGJ was found. EUS-guided fine-needle aspiration was performed, and the mass was diagnosed as a primary SCC of the liver by immunohistochemical staining. (Gut Liver 2012;6:275-279)
Human-Mouse Heterohybridoma를 이용한 사람 항암세포 항체(Monoclonal Antibody)의 생산

Human lymphocytes from peripheral blood, lymph node, spleen, and ascite of cancer patients were fused with mouse P3x 63Ag 8.653 myeloma cells. With 8 patients, 13 fusions were performed. Human-mouse heterohybridomas usually appeared between day 7 and 14, and 613 clones(average; 13 clones per 10' lymphocytes) were successfully propagated. Human immunoglobulins(Ig) were detected from 7 hybridoma culture supernatants by ELISA, and levels of Ig synthesis ranged from 0.1 to 1.5pg/ml. Pretreatment of lymphocytes with mitogen(pokeweed mitogen, or lipopolysacchar ide) increase neither fusion frequency nor yield of Ig-secreting hybridomas. Most of hybridomas were unstable for Ig secretion and only I of the 7 clones produced Ig over 3 month period. We observed the loss of human chromosome in the hybrid clones which stopped secreting lg. Monoclonal antibodies, HDB7 was demonstrated to react with autologus tumor cells, but not stroma, lymphocytes, or allogeneic tumor cells. Our results suggest that human monoclonal antibodies specific against autologous tumor cells can be generated by human-mouse hybridization.
